鸭肠炎病毒基因组末端的克隆及序列分析

Cloning and Sequence Analysis of Genomic Termini of Duck Enteritis Virus

摘要: 根据已知的鸭肠炎病毒基因组序列设计2对引物RTLT1、RTLT2,取基因组自连产物进行PCR反应,分别得到全长1607、1978 bp的序列。将PCR产物克隆至pMD18-T载体,对重组质粒进行PCR和酶切鉴定,将阳性质粒测序,结果显示2对引物扩增的重叠序列部分完全一致。该段序列G+C含量高达75%,含有大量直接和反向重复序列,并发现4个末端特征序列,分别为具有回文结构的α序列、包含两测末端接合位点的β序列、高度保守序列的γ 序列和AnTn序列。将该序列与马疱疹病毒Ⅰ型(EHV-1)、牛疱疹病毒Ⅰ型(BHV-1)、水痘带状疱疹病毒(VZV)、猪伪狂犬病毒(PRV)末端序列进行比较,证实所得到的序列为鸭肠炎病毒基因组末端序列,这些特征序列的发现对进一步了解DEV复制机制有很大帮助。

Abstract: Two pairs of primers RTLT1 and RTLT2 were designed according to the published nucleotide sequence of the duck enteritis virus (DEV).The genomic DNA of DEV was extracted and self-lignased. 1607 bp and 1978 bp genomic fragments were amplified using PCR which were both cloned into pMD18-T vector. After identification of recombinants by PCR and Pst I digestion, the positive plasmids were sequenced. The result showed the sequences of the overlap region of two PCR products were identical. The G+C content of the overlap sequence reached 75%. Some direct and inverted repeated sequences and four characteristic sequences, α palindromes, β sequence surrounding the teminal joint, highly conserved γ sequences and AnTn sequence were found. Compared with the report of the DNA termini of equine herpesvirus 1, bovine herpesvirus 1, varicella zoster virus and pseudorabies virus, the sequence was confirmed to be the termini of duck enteritis virus genome. These sequence elements were proposed to be important for further understanding of DEV replication.
