取代基对黄酮类化合物抑制脂多糖诱导巨噬细胞释放一氧化氮活性的影响

Influence of Substituent on the Anti-inflammatory Effect of Flavonoids against Lipopolysaccharide-induced Nitric Oxide

摘要: 为给黄酮类非甾体抗炎药的研发提供试验依据,本试验以脂多糖诱导巨噬细胞(RAW264.7)释放一氧化氮(NO)为试验体系,研究了5种取代基对黄酮类化合物体外抗炎活性的影响。具体评价了黄酮、5,7-二羟基黄酮、5,7,4'-三羟基黄酮、芹菜素-7-葡萄糖苷及蒙花苷的体外抗炎活性。结果表明5,7,4'-三羟基黄酮能明显抑制脂多糖诱导RAW264.7细胞NO的释放,5,7,4'-三羟基黄酮的4'羟基是其活性的必需基团。

Abstract: Aim to provide experimental data for the research and development of flavonoids as non-steroidal anti-inflammatory drugs (NSAIDS),the substituent effects on the anti-inflammatory effect of five flavonoids against lipopolysaccharide (LPS)-induced nitric oxide (NO) have been studied. Flavone (FLA),5,7-dihydroxyflavone (DFL),5,7,4'-trihydroxyflavone (TFL),apigenin-7-glucoside (APG) and linarin (LIN) were carried out to investigate for anti-inflammatory effects against LPS-induced NO. TFL significantly inhibited generation of NO on LPS stimulated RAW264.7 cells. The 4'-OH was essential for the anti-inflammatory effect of TFL against LPS-induced NO.
