紫外—可见分光光度法测定枫蓼水煎液中总黄酮含量

Determination of Total Flavonoids in Fengliao Decoction by UV-visible Spectrophotometry

摘要: 本试验旨在建立测定枫蓼水煎液中总黄酮含量的紫外—可见分光光度法。以芦丁为对照品,用亚硝酸钠—硝酸铝—氢氧化钠为显色体系,采用紫外—可见分光光度法于510 nm波长处测定其吸光度。结果表明总黄酮浓度在0.008~0.048 mg/mL范围内,芦丁浓度与吸光度的线性关系良好。总黄酮加样平均回收率为100.97%,RSD=1.66%。本试验结果表明所用的方法简便、准确,可用于牛耳枫、辣蓼药材的半成品及成品中总黄酮含量的检测。

Abstract: A UV-visible spectrophotometry method was established for the determination of total flavonoids in Fengliao decoction. Rutin was the control, NaNO2-Al(NO3)3-NaOH was the color developing reagent and the detection wave length was 510 nm. The standard curve was linear in the range of 0.008 to 0.048 mg/mL for total flavonoids, the average recovery of total flavonoids was 100.97% with RSD of 1.66%. The method established was fast and reliable, and could be used to determine the content of total flavonoids in half finished product and finished product of Polygonum hydropiper L. and Daphniphyllum calycinum Benth..
