高效液相色谱法测定不同粒径淫羊藿中淫羊藿苷的溶出量

Determination of Dissolution Amount of Icariin in Different Particle Sizes of Epimedium by HPLC

摘要: 本研究旨在建立高效液相色谱法测定不同粒径淫羊藿中淫羊藿苷的溶出量。采用Shimadzu VP-ODS(150 mm×4.6 mm,5 μm)为色谱柱,以乙腈:水(25:75,V/V)为流动相,流速为1 mL/min,在270 nm波长下进样10 μL进行检测,结果显示淫羊藿苷在0.08~0.40 mg/mL范围内与峰面积呈良好的线性关系(r=0.9999),相对标准偏差(RSD)为1.62%,平均回收率为97.11%~101.08%,该含量测定方法简单、快速,所测定的淫羊藿苷溶出量随着淫羊藿粒径的变小而增加,但粉碎到100目以上时,淫羊藿苷的溶出量不再随粒径的减小而增加。

Abstract: A HPLC method for the determination of icariin in different particle sizes of Epimedium was developed. Chromatography was performed on a Shimadzu VP-ODS column(150 mm×4.6 mm,5 μm). An acetonitrile-water (V:V=25:75) as the mobile phase was used on the wavelength of 270 nm. The results showed that the calibration curve was linear in the range from 0. 08 to 0.40 mg/mL with r=0.9999. The relative standard deviation was 1.62% and the recovery was from 97.11% to 101.08%. The method was accurate and rapid for the determination of icariin in different particle sizes of Epimedium. And the dissolution amount of icariin was increased with the diminution of particle size. But when the size surpassed 100 meshes, the dissolution amount remained constant.
