二喹噁啉羟酸全抗原偶联比HPLC测定方法的建立

摘要: 为建立一种准确、可靠的测定人工抗原偶联比方法,本试验应用高效液相色谱法方法测定全抗原二喹噁啉羟酸—牛血清白蛋白(QCA-BSA)合成中游离二喹噁啉羟酸的含量。采用Waters 2695分离系统、2489紫外检测系统;流动相:甲醇∶水(60∶40);柱温:30 ℃;流速:1 mL/min; 检测波长320 nm。试验结果表明,游离二喹噁啉羟酸的线性范围为0.2~128 μg/mL,线性方程为y=17526x+4648,R2=1,RSD为0.28%,全抗原QCA-BSAⅠ、Ⅱ、Ⅲ的偶联比分别为1.76∶1、28.60∶1、65.63∶1。建立的方法准确、可靠,适用于喹噁啉类人工抗原偶联比的测定。

Abstract: To establish an accurate and reliable determination method for the coupling ratio of the artificial antigen, the HPLC was applied to the determination of free 2-quinoxalinecarboxylic acid of the artificial antigen in this research. The Waters 2695 highly effective liquid phase chromatographs was used by ultraviolet detector 2489 at 320 nm, the mobile phase was methanol∶water(60∶40), column worked at 30 ℃ and the flow rate was 1 mL/min. The results showed that the linearity range of 2-quinoxalinecarboxylic acid was 0.2 to 128 μg/mL,y=17526x+4648,R2 was equal to 1.RSD was 0.28%. This method was accurate and reliable which could be used to determinate the coupling ratio of the artificial antigen.
