细菌人工染色体文库基因组DNA制备技术

Research on the Preparation of Bacterial Artificial Chromosome Library Genomic DNA

摘要: 试验旨在对基因组DNA的制备进行研究,为细菌人工染色体(bacterial artificial chromosome,BAC)文库的构建奠定基础。以豁眼鹅全血为试验材料,提取高质量的基因组DNA,分别采用Hind Ⅲ、EcoR Ⅰ和BamH Ⅰ 3种限制性内切酶对所提基因组DNA进行部分酶切,并利用控制酶切时间、设置不同的Hind Ⅲ酶切浓度梯度对基因组DNA进行部分酶切。结果表明,Hind Ⅲ为最佳限制性内切酶,并得到了最佳酶切用量(40 U/μL)。该方法所制备的基因组DNA质量较好,可用于BAC文库的构建。

Abstract: This paper introduced a preparing method of genome DNA for a Huoyan goose and it laid a foundation for the constructing of bacterial artificial chromosome library .The genome DNA was extracted from a goose’s blood and digested by three restriction enzymes of Hind Ⅲ,EcoR Ⅰ and BamH Ⅰ. Meanwhile,different concentration gradients were set in order to get the optimal concentration of restriction enzyme.The results showed that Hind Ⅲ was the best enzyme and the optimal concentration of restriction enzyme was 40 U/μL. The genome DNA abstained by this method had a good quality and could be used to construct the bacterial artificial chromosome library.
