牛精子蛋白胶内酶切条件优化及问题分析

摘要: 针对银染蛋白的胶内酶切质谱鉴定成功率较低的技术现状,对影响牛精子蛋白酶切鉴定的酶量、酶切时间、凝胶存储时间等影响因素进行了摸索,并优化了挖点、脱色、脱水、酶切消化和抽提肽段等操作细节,同时对质谱峰图中出现的污染问题进行了分析解决,提高了蛋白鉴定的成功率,为牛精子蛋白质的质谱鉴定奠定了技术基础。

Abstract: The achievemet ratio of identifying protein of sliver stained through MALDI-TOF-MS is low. The following variables were studied to effect of in-gel digestion of silver-stained 2-D gels with bovine spermatozoa protein :enzyme concentration, degestion time and dry gel. And optimization of in-gel digestion of silver-stained 2-D gels is including spot excision, destaing, digestion and extraction. Contaminating protein was analyzed and solution in this paper in order to enhance achievement ratio of protein identity
