蓖麻毒素诱导的小鼠淋巴细胞的凋亡

The Apoptosis of Mouse Iymphocytes Induced by Ricin Toxin

摘要: 无菌制备小鼠脾淋巴细胞,以不同浓度蓖麻毒素(0、0.3、3、30、300 μg/mL)对细胞进行处理,于不同时间段(3、6、12、24 h)采用MTS法对细胞活力进行检测。结果表明,蓖麻毒素对淋巴细胞有明显的毒性作用,3 μg/mL的毒素处理细胞24 h后,细胞活力降为60%左右。为研究蓖麻毒素是否是通过诱发凋亡作用而引起细胞死亡,本试验对典型的凋亡特征进行了检测,首先用流式细胞仪对细胞的线粒体膜电位进行了检测,证明经毒素处理细胞后,其电位发生显著变化(P<0.05);Hoechst33258染色和DNA 梯度裂解检测结果同时也证明,接毒细胞发生了明显的调亡作用。此结果可为深入研究蓖麻毒素的毒理作用机制提供理论依据。

Abstract: Prepared mouse splenic lymphocytes under aseptic conditions, then treated cells with different concentratio of ricin toxin (0,0.3,3,30,300 μg/mL) and detected its viability at different durations(3,6,12,24 h) by MTS method. The result showed that RT had significant toxic effect on lymphocytes and its viaility only 〖JP2〗60% of control cells when treated by RT for 24 h.〖JP〗〖LM〗〖WT〗〖WT5”BZ〗 In order to invest that if the cell death induced by RT according to the apoptotic pathway, we detected series of typical apoptotic character in the experiment, membrane potential detection, Hoechst33258 staining and DNA ladder. All the results illuminated that RT could induce apoptosis of lymphocytes indubitable. And also, this conclusion can provide parts of theories for the toxic action’s further study of ricin.
