猪鼻支原体抗血清的制备与鉴定

Preparation and Identification of Mycoplasma hyrhinis Antiserum

摘要: 猪鼻支原体(Mycoplasma hyrhinis,Mhr)是猪鼻腔的常在菌。将猪鼻支原体与弗氏佐剂共乳化,免疫新西兰大白兔,获得猪鼻支原体兔抗血清。Western blotting检测其特异性;建立间接ELISA方法检测其抗体效价;将获得的猪鼻支原体兔抗血清分别加入猪鼻支原体和猪肺炎支原体培养物中,进行生长抑制试验。结果显示,制备的猪鼻支原体兔抗血清与猪鼻支原体具有反应原性;抗体效价达1:160 000以上;猪鼻支原体兔抗血清可以显著抑制猪鼻支原体的生长,而对猪肺炎支原体的生长影响较小。本试验结果为鉴定和检测猪鼻支原体提供了方法,也为猪肺炎支原体的分离提供了参考。

Abstract: Mycoplasma hyrhinis (Mhr) was a common inhabitant in the nasal cavity of pigs.To prepare antiserum of Mhr, we immuned New Zealand White rabbit with Mhr which was emulsified with freund's adjuvant.Western blotting analysis showed that antiserum had the immuneoreactivity to Mhr.The antibody titer was more than 1:160 000 detected by established indirect ELISA.The growth inhibition test confirmed that antiserum could significantly inhibit the growth of Mhr, but the effect on the growth of Mycoplasma pneumoniae (Mhp) was weak.The above results would provide means for identification and detection of Mhr, and also provide references for the separation of Mhp.
