玉竹提取物A对小鼠免疫性肝损伤治疗作用的研究

The Therapeutic Effects of EA-PAOA on Liver Injury Induced by Concanavalin A in Kunming Mice

摘要: 本试验旨在研究玉竹提取物A(extraction A of polygonatum odoratum, EA-PAOA)对刀豆蛋白A(ConA)诱导的小鼠免疫性肝损伤的治疗作用,探讨其发挥此作用的可能机制。将昆明小鼠随机分成正常对照组、模型对照组、试验1组(EA-PAOA 0.5 g/kg)、试验2组(EA-PAOA 1 g/kg)、试验3组(EA-PAOA 2 g/kg)。除正常对照组外,其余各组均尾静脉注射20 mg/kg的ConA 0.5 mL,每天06∶00注射1次,连续5 d。每天注射ConA 2 h后,EA-PAOA 3个试验组分别腹腔注射不同浓度的EA-PAOA 0.5 mL,每天3次,共4 d,第5天只在注射ConA 后2 h注射1次EA-PAOA。末次注射EA-PAOA后6 h,摘眼球取血低温保存,待测谷丙转氨酶活性(ALT)、3个细胞因子;取肝待行肝脏组织病理学检查注射;取脾待行淋巴细胞增殖试验(MTT法)和流式细胞术以检测T淋巴细胞亚群。结果显示,EA-PAOA 3个试验组血清中,ALT、IFN-γ、TNF-α水平明显降低,IL-10水平明显升高,能显著抑制T淋巴细胞的转化增殖,与模型对照组相比差异极显著(P<0.01);但流式细胞术检测T淋巴细胞亚群发现,5组CD4+T和CD8+ T淋巴细胞的数量和比例并无明显差别,都与正常对照组无显著差异;EA-PAOA 3个试验组肝脏组织病理改变较模型对照组明显改善。表明EA-PAOA可能是通过抑制T淋巴细胞的转化增殖,抑制其活性,调节Th1和Th2的数量及比例,降低血清中IFN-γ、TNF-α水平,提高IL-10水平,以此发挥其治疗免疫性肝损伤的功能。

Abstract: The objective of this paper was to investigate the therapeutic effects of EA-PAOA on the immune hepatic injury in Kunming mice and study its therapeutic mechanisms. Kunming mice were divided into five groups, which were normal control, model control, test 1(EA-PAOA 0.5 g/kg), test 2(EA-PAOA 1 g/kg), test 3 (EA-PAOA 2 g/kg). Model control and three test groups, respectively, received intravenous injections of Concanavalin A(20 mg/kg) 0.5 mL, once one day, five days in all. Two hours after the first injection of Concanavalin A, three test groups were administrated intraperitoneal injections of EA-PAOA 0.5 mL, three times one day, five days in all.Six hours after the last injection of EA-PAOA, blood were collected, the liver was reserved for routine light microscopy and the spleen was aimed to do T-cell proliferaction activity tests. We tested the effects of EA-PAOA on T lymphocyte subsets of spleen with flow cytometry. Compared to model control group, in the three EA-PAOA tests, the level of serum ALT,IFN-γ,TNF-α were lower,serum IL-10 were higher(P<0.01) .EA-PAOA could obviously inhibit T-cell proliferactive activity. But the number of CD4+ T and CD8+ T lymphocyte in spleen with flow cytometry hadn’t much difference. The results indicated that EA-PAOA had distinct anti-liver injury function. Therefore, we suppose that the therapeutic mechanisms of EA-PAOA could be to inhibit T-cell proliferaction activity, increase serum concentration of IL-10, and decrease serum concentration of IFN-γ,TNF-α. 
