Establishment of Loop-mediated Isothermal Amplification for Detection of Swine Influenza Virus

Abstract: The objective of this study was to establish a loop-mediated isothermal amplification(LAMP) method for swine influenza virus (SIV). Six primers were designed according to the SIV NP gene sequence. Then specificity, sensitivity and reproducibility of the test were carried out. In the results, the developed system was specific for the detection of H1N1, H1N2, H3N2, avian-like H1N1 subtype SIV and H1N1 subtype influenza A, there was no cross-reactivity with porcine reproductive and respiratory syndrome virus, porcine parvovirus, classical swine fever virus, pseudorabies virus, porcine circovirus type 2 and Japanese encephalitis virus. The sensitivity of this assay was 100 copies/μL plasmid DNA. These results showed that LAMP method had higher sensitivity and specificity, and could be used for the detection of SIV.
