牦牛Toll样受体5基因的克隆及序列分析

Abstract

Abstract: This study aimed to investigate the characteristics of the TLR5 gene from yak. According to the published cattle TLR5 gene sequence in the GenBank, we designed special primers for the TLR5 gene. The results of sequence analysis showed that the length of the cloning TLR5 cDNA was 2582 bp，contained a complete 2577 bp open reading frame(ORF). It encoded a peptide of 858 amino acids whose N-terminal signal peptide contained 21 amino acids, and the protein had a calculated molecular weight of 97.981 ku and a PI of 4.85. The results of protein prediction showed that TLR5 protein perform as the hydrophilic. Sequence homology analysis showed that yak had higher homology with wild yak, cattle, sheep and goat. The results showed that TLR5 gene sequence was conserved in mammals. The successful cloning of TLR5 gene of yak provided important basis for the study of molecular mechanism of innate immune response.
