Establishment of Indirect Hemagglutination Assay for Antibodies Detection of APP

Abstract: In order to understand the infection status of Actinobacillus pleuropneumoniae in Jiangxi province, we developed indirect hemagglutination assay (IHA) for detection of antibody by using APP isolates of Jiangxi province. IHA consisting of glutaraldehyde-tannic-fixed rabbit red blood cells (RRBC) coated with the supernatant of repetitive freeze-thawed and sonicated APP was established. The positive sera of hog cholera virus, porcine reproductive and respiratory syndrome virus, pseudorabies virus, Escherichia coli, Salmonella, Haemophilus parasuis showed negative results. Different batches of IHA antigen showed good repeatability. The results showed that the specificity and repeatability of IHA were good, which created the foundations for the serological survey.
