Abstract: In order to gain polyclonal antibody against α-casein, the commercial α-casein were linked with carrier protein BSA and OVA, respectively, as immunogen (α-casein-BSA) and coating antigen (α-casein-OVA). The reaction products were identified by nondenaturing PAGE and SDS-PAGE. New Zealand White rabbits were immunized with the immunogen (α-casein- BSA) to produce polyclonal antibody. The results showed that the titer of the antibody was 1∶256000 and had high cross reactivity with κ-casein. It could be used to detect the total amount of α-casein and κ-casein in milk.
