Determination of Residual Avermectins in the Cattle Muscle Using IAC-HPLC with Fluorescence Detection

Abstract: A HPLC method was developed with immunoaffinity chromatography columns(IAC)as purify process to detect the residual of avermectin,ivermectin,doramectin and eprinomectin in bovine muscle tissue.Samples were extracted with methanol,purified by IAC and followed by derivatization with acetic anhydride and 1-methylimidazole in acetonitrile.The optimum time required for the completion of derivation reaction of the 4 drugs was 100 min at 96 ℃.Recoveries ranged from 77.3 % to 119.5% with CV from 1.5% to 18.9% when these drugs were spiked in bovine muscle at levels of 5,10,50 and 100 ng/g.The limit of detection of the method was 0.8 ng/g in bovine muscle tissue.This method could be used to detect the residual of avermectins in bovine muscle tissue.
