Cloning and Diversity Analysis of Coding Sequence of Myostatin Gene in Huili Black Goat

Abstract: To explore genetic differentiation conditions of Huili Black goat, primers were designed according to myostatin (MSTN) gene of ordinary goats, the coding sequence of MSTN gene in Huili Black goat was amplified and cloned by PCR and aligned with the nucleotide sequence of other species. The results showed that, the complete coding sequences of MSTN gene in Huili Black goat was 1128 bp, and it consisted of the exon 1 (372 bp), exon 2(375 bp), exon 3 (381 bp); the homology with Jianchang Black goat and Qianbei Ma goat were the highest, which reached 97.4%, as a family, they had the closest genetic relationship; the homology with cattle, Rhesus monkey and dog species were the lowest and they were distantly related to the Huili Black goat; the coding sequence of MSTN gene encoded 375 amino acids, the contents of all kinds of amino acids were very different, the leucine (Leu) was the most abundant protein, reaching 8.80%. This research provided a molecular genetics basis for the genetic resources protection and development of Huili Black goat.
