The Influence of Different Decalcifying Fluid on the Section Staining of Articular Joint Tissue of Rabbit Knee

Abstract: To investigate the effect of different decalcifying fluid on the color of articular joint tissue section of rabbit knee,to aid at the histological evaluation of hyaline cartilage and distribution of fibrocartilage polysaccharide. Prepare New Zealand rabbits,after the fully fixation of the whole rabbit knee joints in 10% neutral formalin,the tissues were washed with running water,then decalcified with 4 different decalcifying solutions (10% HNO3 decalcifying solutions,3% HNO3 decalcifying solutions,modified AF decalcifying solutions and modified 30% hydrochloric acid). The tissue sections were stained with hematoxylin-eosin (HE),alcian blue and safranin-O,and observed under a microscope to evaluate the morphology of chondrocytes and the cartilage matrix. Results:①The effects of HE staining: first is 3% HNO3 decalcifying solutions group,next is modified 30% hydrochloric acid group,once again is 10% HNO3 decalcifying solutions and modified AF decalcifying solutions group. ②The effect of alcian blue and safranin-O staining in 3% HNO3 decalcifying solutions group and modified AF decalcifying solutions group were better than other groups.
