Isolation and the Full-length Genome Sequencing of Avian Reituculoendotheliosis Virus GD09 Isolate

Abstract: An avian reticuloendotheliosis virus (REV),designated as GD09,was isolated and identified by DF-1 cell culture,PCR and immunofluorescence assay specific to REV from broiler breeder suspected with REV infection in Guangdong province. Based on the public full-length proviral genome sequence of avian reticuloendotheliosis virus strain,six pairs of primers were synthesized,and the full-length proviral genome of GD09 isolate was sequenced by the method of fragment amplification. The full-length proviral genome sequence of GD09 isolate was 8295 bp;its size had a little difference from that of published full-length genome sequences,but its structure corresponded with typical retroviral genome structure for completely copy ability,the known oncogenes were not included in its genome. According to the gp90 gene sequence comparison of GD09 strain with those of the other REV strains at home and abroad,the sequences of gp90 gene of GD09 strain and phylogenetic analysis showed that the gp90 gene of GD09 strain had the closest genetic relationship to HA9901 strain.
