Development of Gold Immunochromatography DiagnosticStrips for Detecting Akabane Virus

Abstract: We developed akabane virus gold immunochroamatography diagnostic strips with the “sandwich” assay format, which is suitable for field fast test. Akabane virus specific McAbs 3A and 2C were purified, purified McAbs were labeled with the colloidal gold synthesized by trisodium citrate disoxidation method. Goat anti-mouse IgG and purified McAb were separately sprayed on the nitrocellulose membrane as the control line (C line) and the test line (T line) to detect akabane virus in the samples. Test results indicated that akabane virus gold immunochroamatography diagnostic strips established in this study were sensitive (with sensitivity of 10 TCID50), specific, rapid, stable and simple. It is very suitable for basic veterinary medical laboratories to do fast large-scale field screening test, and has very important significance to akabane virus fast diagnose.
