乳汁中RNA提取方法的建立

摘要: 用牛初乳作为试验材料,进行乳汁体细胞的提取及RNA的提取与扩增。结果显示:牛初乳中含有大量的体细胞(SCC约为20万/ml,活力约为60%),从体细胞中提取RNA,所提取的RNA完整性和纯度可以用来合成cDNA,管家基因GAPDH扩增条带正确,表明可以用合成的cDNA作为模板进行其它基因的扩增,为奶牛在基因工程方面的研究提供分子生物学技术平台。

Abstract: The cow colostrum was applied as the experimental material, and the milk somatic cells were extracted and RNA was extracted and amplicated. The result shows that the colostrum contains a large number of somatic cells(SCC is about 200,000 /ml and the activity of somatic cells is about 60%). The integrality and purity quotient of the extracted RNA can be used to produce DNA. By RT-PCR amplication, the bands of house-keeping gene GAPDH was reasonable, which indicates that produced DNA can be amplificated as the template for other genes. It provides the primary basis of molecular biological analysis for the cow in the aspect of the genetic engineering research.
