猪伪狂犬病病毒LX株的分离与鉴定

Abstract

Abstract: To isolate pseudorabies virus (PRV), the morbid materials were collected from some swine farm of Liaoning province and inoculated on BHK-21 cell after disposal.After blind transfer of culture,two portions showed typical cytopathic effect(CPE). TCID₅₀ of virus was 10^-7.8/mL.By the indirect immunofluorescence experiment,the yellow-green fluorescence occurred in the cell cytoblastema.The virus appeared typical porcine pseudorabies virus form, diameter was about 140 to 200 nm,circle with peolos and peolomer. It was inactivated by chloroform, ether,acid and heat. The cell cytoblastema which showed CPE were tested by RT-PCR, and the result was postive. The experimental animals appeared obvious clinical symptom and dead after inoculation.The results showed that PRV was successfully isolated from Liaoning province.
