Effect of Copper Complexes Derived from Schiff ’s Base of Lysine on the Rumen Fermentation of Sheep

Four male Xinjiang Merino sheep,with 40 kg of average live weight,fitted with permanent cannulas in the rumen,were supplemented with three dosages (0,4,8 g/sheep·d)of Copper complexes derived from Schiff’s base of Lysine in three periods respectively to study the effect of Copper complexes derived from Schiff’s base of Lysine on rumen fermentation. The results showed that the intake of DM of corn straw was increased 12.5%at the level of 8 g/sheep·d(P<0.05). The dosage of 4 g/sheep·d had no effect on rumen pH value(P>0.05),but that of 8 g/sheep·d tended to slow up the decrease of rumen pH value 3 h after feeding. The dosage of 4 g/sheep·d did not influence the concentration of ammonia nitrogen(P>0.05),but that of 8 g/sheep·d decreased the concentration of ruminal ammonia nitrogen. The ruminal concentrations of acetic and total volatile fatty acid (TVFA) were decreased,however,the propionic acid and butyric acid were increased,and rumen fermentation type was changed from acetic to propionic acid after addition of the Copper complexes derived from Schiff’s base of Lysine. This experiment indicated that the Copper complexes derived from Schiff base of Lysine could regulate the rumen fermentation. 

A total of 96 1day healthy AA broilers of commercial strain were chosen in this experiment, the birds were divided into 3 groups: control, test groupⅠand Ⅱ, four replicates of 8 birds were in a group. The total experimental time was 49 days. During 1~21 days, birds in test groupⅠand Ⅱ were fed basal diet supplemented with 0.1% alfalfa glycosides, respectively. During 22~35 days and the 36~49 days, birds in groupⅠwere fed basal diet supplemented with 0.1% alfalfa glycosides, and birds in groupⅡ were fed basal diet supplemented with 0.2 % alfalfa glycosides. The results showed that: ①Dietary supplementation of 0.1% alfalfa glycosides effects significantly the feed intake(FI) of the broilers in 22~35 days,but the performance difference of other groups and experimental times is not significant (P>0.05). ②Dietary supplementation of alfalfa glycosides to the different date age has the influence on serum and liver GSHPx, SOD and MDA level in different extend.

A study was conducted to evaluate the Saccharicterpenin effects on production performance, intestinal canal micro-flora population, biochemical parmeter of blood on the piglets early weaning(PEW). Treatments were as follows: treatment Ⅰ is the control group, the group treatment Ⅱ,Ⅲ,Ⅳ,Ⅴ respectly contained 250 mg/kg Saccharicterpenin, 500 mg/kg Saccharicterpenin, 250 mg/kg Saccharicterpenin plus 4 mg/kg Flavomycin, 4 mg/kg Flavomycin. The results showed that: Supplemented with 500 mg/kg Saccharicterpenin of Flavomycin plus 250 mg/kg Saccharicterpenin, the number of TP、GLO、G/A were improved, the group treatment Ⅲ is the best; IgA is significant improved(P<0.05), the group treatment Ⅱ,Ⅲ,Ⅳ,Ⅴ was higher 43.21%, 104.89%,73.21%,27.74% than the control, IgG,the group treatment Ⅱ,Ⅲ,Ⅳ,Ⅴ was higher 10.50%,12.07%,11.05%,10.26% than the control, group; treatment Ⅱ,Ⅲ,Ⅳ,Ⅴ on the Serum of BUN was higher 2.67%,2.96%, 2.81%, 2.52% than the control, but no significant difference to GLU(P>0.05), The number of AST, ALT, ALP of the group treatment Ⅲ was higher than others, but the group treatment Ⅱ,Ⅲ,Ⅳ,Ⅴ of LDH was lower 2.71%,4.02%, 2.96%, 1.65% than the control.

The activity of SOD and GSHPx and the level of MDA of lymphocyte membrane in the modle chicken with Vitamin E (VE)deficiency that produced by using low content of VE feedstuff and supplying unsaturated fatty acid in the diet were detected by Test Kit;the membrane fluidity of lymphocyte in the same modle chicken was detected through fluorescence polarization. The results showed that activity of SOD and GSH-Px and fluidity of lymphocyte membrane were significantly lower in experimental chicken than control ones(P<0.05,P<0.01);the MDA content were significantly higher than control ones (P<0.05).The results indicated that VE plays an important role in maintaining the function of lymphocyte membrane in chicken. 

The fat is indispensable component ingredients and nutriment of poultry, which not only keeps poultry’s growing, breeding, and producing normally, but also has a great effect on poultry’s immune organ, tissue and regulate its immune response. So, a survey was made on the effect of fat on poultry’s immune function such as immune organ growth, body’s immunity, cell-mediated immunity, non-special immunity, and production of Cytokines. This could provide the theory basis for preparing rational diets of poultry and improving their ability against diseases in poultry’s production practice.

Genetic diversity of Pinghu royal jelly bee and Suwang No.1 bee were evaluated with 6 microstaellite loci, the genetic variability within breeds and genetic differentiation between breeds were estimated, the results showed that in 6 microstaellite loci, the 40 alleles were found, the number of alleles per locus ranged from 4 to 10, the average expected heterozygosity and PIC of all loci were 0.5078 and 0.4708 respectively. Mean numbers of effective alleles of Pinghu royal jelly bee and Suwang No.1 bee were 0.4981 and 0.5007, inbreeding index(FST), Reynolds' genetic distance and gene flow between two populations were 0.037,0.03770 and 6.507 respectively, these results indicated that the heterozygosity and the genetic diversity of two bee populations were very high.

The prolactin (PRL) gene was studied as a candidate gene for the prolificacy of Jining Grey goats. Single nucleotide polymorphisms of exon 1 to exon 5 of PRL gene were detected in high fecundity breeds (Jining Grey, Boer, Wendeng dairy goat) and low fecundity breed (Inner Mongolia Cashmere goat) with five pairs of primers by PCR-SSCP. For the first time in goats, the entire exon sequence of PRL gene was obtained by means of clone sequencing. No single nucleotide polymorphism was detected in four goat breeds. This result indicated that the detected loci of PRL gene had no significant effect on prolificacy of Jining Grey goats.

The 493bp fragment of Callipyge gene of 113 individuals was amplified by PCR, of which 35 were from Boer goat, 38 were from Nanjiang brown goat and 40 were from Chengde polled goat. There was a variation of C184T discovered by sequencing. Molecular genetic diversity of Callipyge gene with restriction endomuclease Fok I was studied by PCR-RFLP. The polymorphism was existed in Boer goat and Nanjiang brown goat. There were 3 genotypes, which was CC, CT and TT, respectively. The frequency of genotypes was 0.6571, 0.3143 and 0.0286 respectively in Boer goat. The frequency of genotypes was 0.8947, 0.1053 and 0.0000 respectively in Nanjiang brown goat. However, there is all CC genotype in Nanjiang brown goat. It suggested that there was a relationship of the variation of C184T to the trait of Callipyge in goat. 

With the method developed by our research, using agar without neutral red, plaques were picked under an inverted microscope, we plaque-purified 3 strains H5N1 subtype AIV chicken embryo isolates and obtained thirteen plaque-purified strains with marked difference in size and shape. Our research indicate addition of suitable amount of trypsin could facilitate plaque formation with marked increase in plaque diameter and plaque-forming units, its optimal concentration being 50 μg/ml, when the concentration of calf serum is 2%. TCID50, EID50 and LD50 testing were done on embryo isolates and the plaque-purified strains, when It was found in the study that for the size and shape of purified strains which came from the same embryo isolate exist notable difference, the larger the plaque the higher the virulence, and that the virulence of the largeplaque strains approached that of the embryo isolates, which indicate that virus particles producing large plaques were dominant in embryo isolates. Analysis of the nucleotide sequences and deduced amino acid sequences of HA genes of the 13 plaque-purified strains revealed that the cleavage site amino acid sequences of the HA protein were all conforming to structures assigned to the molecular characteristics of highly pathogenic notifiable avian influenza virus.

Balearica pavonina has a small quantity and it's significant to develop artificial reproduction and breeding. This research extracted genome DNA from feathers and amplified the EE0.6 gene of Balearica pavonina specifically with two combinations of primers. The results showed that male birds displayed one distinct amplification band,whereas two bands were amplified for female ones. With the results, the sex of Balearica pavonina could be identified successfully. It is meaningful for the reproduction of the birds.

E0 gene was taked back from the pGEM-T Easy-E0 recombinant plasmid cleaved by BamH I and Pst I, was cloned into the EGFP. High pure recombinant plasmid of pEGFP- E0 was transfected into PK-15 cell, BHK-21 cell, VERO cell by LipofectamineTM2000, examine the cell for fluorescence using a UV microscope. All could find the fluorescence in PK-15 cells, BHK-21 cells, VERO cells. In 24 h, 48 h and 72 h, count of the the cell for fluorescence were increasing, count of the cell for fluorescence were most in PK-15 cells, BHK-21 cells in 72 hs, VERO cells. count of the cell for fluorescence were most in PK-15 cells in 72 h. Detection indicated expressed E0 protein mainly locate in the plasma of the PK-15 cells. After cleaved by BamH I and Pst I, PCR and indirect immuno fluorescence assay, detection indicated large number of expressed E0 protein are in the PK-15 cells.After PK-15 cells and PK-E0 cells infected by HCLV, the titer of HCLV was the highest in 72 h, then began to decrease. Detection indicated large number of expressed E0 protein caused increase of the titer of HCLV.

To observe and measure the Cardiac anatomic characteristic of Wuzhishan miniature pig inbredline, twenty individuals with different month-age were anatomized, and obtained the related anatomic parameters. This study showed that the cardiac gross morphology, the chambers division of heart and cardiac internal structure in the 2 and 4 months Wuzhishan miniature pig were the same, and in accord with that of human, but the cardiac size and some anatomic parameters were markedly different from that of adult, the diameter of blood vessel in the cardiac basal part were different from that of other local pig breeds and human. The anatomic study on the heart provided the basic data for the utilization of the Wuzhishan miniature pig inbredline for comparative medicine and clinical xenotransplantation.

In order to study the reaction and mechanism of nanometer titanium dioxide with macromolecule. The effects of different concentrations of nanometer titanium dioxide treated with different reaction time, temperature, visible light and UVA on the cellulase activity were reported in this paper. The results indicated that TiO2 treated with visible light and UVA could make cellulase inactive. And the inactive effect is in direct proportion to the concentration, temperature and the reaction time.

No significant difference fund Between all groups' DMI and FCR; Even SNF shown an increased tendency, supplementation of rapeseed (350g/h/d), rape oil (150g g/h/d) and fatty acid (200g g/h/d) did not affect the composition of milk SNF(P>0.05). Supplementation of fatty acid (200g g/h/d) increased milk fat significantly, but .that of rapeseed (350g/h/d), rape oil (150g g/h/d) had no any positive effect on the composition of milk SNF (P>0.05). group1 profited extract 1.08 Yuan kg/h/d, group2 lost 0.06 Yuan, group3 lost 0.14 Yuan kg/h/d;Above results shown supplemented rapeseed (350g/h/d) to the dairy cow whose milking day are 90-150, increase economical efficiency, supplementation of fatty acid enhanced the milk quality.

16 Staphylococcus aureus strains isolated from bovine mastitis of two intensive dairy farms were typed with RAPDPCR. Amplification with 3 primers, only primer OLP11 produce band patterns that have characteristics. Selection of anneal temperature and concentration of template shows that 33℃ and original concentration could produce clear, distinguishable and repeatable band patterns. The result shows that 2 groups were divided for the 16 strains and 14 belong to same group. Related coefficient of the band patterns for most strains is only 0.04, which demonstrated that these strains have similar genetic background.

Judging from the clinical data, we find that Left displacement of the abomasums has 295 examples, accounts for 81.04%, Right displacement of the abomasum has 69 examples, accounts for 18.96%, there are 231cases among the cows of the 1st parity, accounts for 63.46%, 84 cases among the cows of the 2nd parity, accounts for 23.08%,37 cases among the cows of the 3rd parity, account for 10.16%,9 cases among the cows of the 4th parity, account for 2.47%,2 cases of the cows above the 5th parity, account for 0.55%. 357 head of cattle were cured, the cure rate is 98.08%. Besides, other information has also been carried out counting, such as, the morbidity among different season, the serum electrolyte changes, the regular rumination and defecation of the illed cattle after the operation.

The antiserum was prepared with the mixture of OFL-BSA,DIF-BSA and NORBSA.The Ci-ELISA method for the detection of ofloxacin ,difloxacin and norfloxacin was established with it.To ofloxacin ,difloxacin and norfloxacin.The regression equations about their standard curves were y=-0.1984x+0.9687(r=0.9885),y=-0.1738x+0.9150 (r=0.9985) and y=-0.2004x+0.9572 (r=0.9954),respectively.The 50% inhibition concentration(IC50) were 231.57,245.32 and 186.14ng/ml,respectively. The limits of detect (LOD) were 1.62,1.86 and 1.07ng/ml ,respectively. The coefficients of variation for the Ci-ELISA method were less than 5.44% in intra-assay and 12.45% in inter-assay. The recoveries in milk ranged from 71.70% to 94.53%.

150 wool samples including 3 types of fineness(≤18 μm,18~20 μm and ≥20 μm) were taken, and the variations of fiber diameter along and within staple were studied. The results demonstrate that the variation of fiber diameter along staple is similar in different types of fineness and agrees with the seasonal change of feedstuff, and the wool with fineness being less than 18 μm has the least variations of fiber diameter(P<0.01); Contrarily, the variation of diameter between fibers within staple is increased in company with the reducing of fineness(P<0.01).
