Nutrient Digestibility and Growth of Sulking Calves Fed Various Quality Forages

10 healthy Holstein calves with similar maternal body measurement, birth weight, body measurement, were selected randomly to two groups and fed corn straw(FCS) and hay (H) respectively. The nutrient digestibility and growth performance were measured. The results showed that H had lower concentrate intake (P>0.05) and higher Forage (P<0.01) and dry matter intake (P>0.05) than FCS. Compared with FCS, NDF (P<0.01), ADF (P<0.01), crude protein (CP) (P>0.05), Ash (P>0.05) of digestibility, daily gain (P>0.05), chest girth (P>0.05), chest width (P>0.05) of H were higher. It was concluded that supplementing hay would increase the dry matter intake and nutrient digestibility of sulking calves and improve the growth performance of sulking calves and make a good condition for finishing.

As the experimental materials, chickens were treated with acute and chronic cold stress (12±1℃) in order to explore the effect of cold stress on antioxidative function. Through the detection of T-AOC, CAT activity, GSH-Px activity, SOD activity, MDA contents in hypothalamus and serum, it is demonstrated that cold stress could change antioxidative function, induce oxidative stress, increase the content of free radicals, and finally result in the body damage.

Intracytoplasmic sperm injection (ICSI) procedure has become the treatment of choice in cases of male factor infertility and in cases of failed conventional IVF cycles. In bovine, the technique is mainly used to produce sexed embryos and transgenic animals, and to research the mechanism of fertilization. Research status, main results and progress of the recent years were reviewed in this paper. At the same time, the main problem of bovine ICSI were expatiated. And it has also made a prospect of the application of ICSI in human and animals.

The DNA extraction from the hairs of the Panthera Tigris Altaica, cooking extraction method in water was studied, and the comparison with the traditional phenol and chloroform method was done. The cooking extraction method was the method from which genomic DNA was obtained when the cells were broken and proteins were denatured after the cells were cooked and cooled quickly. The extracting results of the two methods were detected through agrose gel electrophoresis, PCR amplification and PAGE gel electrophoresis, the OD value, density and purity. The analysis result indicated that the cooking extraction method in water is a quick, simple, economic and efficient method to extract DNA from the hairs of the Panthera Tigris Altaica.

Based on gene pooling and sequencing, a new SNPs was found on the bovine MyoD1 gene, its transition C-T leads to a reaplacement of glicine with serine. Polymorphism and relationship with the carcass traits was analysised by PCR-RFLP in different populations. The transition of this locus fit in with Hardy-Weinberg equilibrium in Chinese-simmental cattle and Luxi cattle, but not in Qinchuan cattle. Statistical analysis revealed higher value in back fat and thickness of thigh (P<0.05), not in average daily growth, yield of carcass, intramuscular fat, marbling and yenderness.

To establish a preliminary procedure for making interspecific chimeras between cattle and buffalo by Aggregating Blastomeres Method. Chimeric embryos were reconstructed by aggregating blastomeres from early bovine and buffalo embryos isolated by either enzymatic or mechanical method in a droplet of culture medium containing 100 μg/ml phytohemagglutinin (PHA). Aggregating buffalo morulae with bovine embryos at either 8-cell or blastula stage did not differ significantly in their aggregation rate (85.7% vs 77.8%, P>0.05) and proportion developing to blastocysts (72.2% vs 42%; P>0.05). There were also no significant difference in aggregation rate (92.3% vs 86.7%, P>0.05) and blastocyst development (58.3% vs 46.2%; P>0.05) between the chimeric embryos derived from 8-cell embryonic blastomeres isolated by 0.25% Pronase and microsurgery. In conclusions,①The aggregation rate and embryonic development of chimeric embryos are not influenced by the stages of embryos to be aggregated; ②Methods for isolation of blastomeres (enzymatic treament and microsurgery) do not affect their aggregation rate and embryonic development.

The 892 bp fragment (97 bp of part of exon 8 (the full length of exon 8 was 100 bp), 683 bp of intron 8, 70 bp of exon 9 and 42 bp of part of intron 9)of prolactin receptor (PRLR) gene was amplified successfully in Jining Grey goats by PCR and cloned into pGEM-T Easy vector. The positive clones were further identified by PCR analysis. The nucleotide sequence was detected and the peptide sequence of this fragment was deduced. This sequence shared 99.4 %, 97.01%, 89.22%, 89.22%, 88.02% nucleotide homology with the published mRNA of PRLR gene of sheep, cow, human, rat and mouse separately, and the amino acid homology was 100%, 94.55%, 81.88%, 81.82%, 83.64% separately.

Total RNA from our laboratory stored spleens of four Sanhuang-chickens, one Wuji-chicken and one Zhenzhu-chicken were extracted respectively using Trizol reagent (Gibco/BRL). First-strand cDNA (fcDNA) were generated from total RNA using primer P15T: (5’-CTG ATC TAG AGG TAC CGG ATC CTT TTT TTT TTT TTT T-3’). According to the sequence of chicken BF2gene in GenBank, Two pairs of primers containing EcoRⅠ and Hind Ⅲ restriction sites was designed to clone BF2 genes extracellular domains.After being purified and recovered, the PCR product was inserted into the pGEM-T Easy vector. The recombinant pGEM-T Easy/BF2(1-5) plamid was digested by double enzymes (EcoRⅠand Hind Ⅲ ). The DNA fragments of interest were subcloned into into pMAL-p2X expressed vector between the EcoR I and Hind Ⅲ restriction sites (named p2X-BF2(1-5), respectively) and transfected into E. coli TB1. The clones were sequenced on both strands by the Shanghai Sangon Biotechnology Company.The recombinant plasmids p2X-BF2(1-5) were identified by EcoRⅠ and Hind Ⅲ. The engineering TB1 strains containing p2X-BF2(1-5) were induced by IPTG and SDS-PAGE analysis was carried out to identify the expressed protein.The result revealed the BF2 gene extracellular domains were composed of 807 bp-819 bp nucleotides encoding 269-273 amino acid residues. The expressed MBP-eBF2 fusion proteins were purified using the amylose affinity resin column and DEAE-Sepharose ion exchange chromatography, and cleaved by the factor Xa protease. The purified fusion proteins were identified by western blot and create a basis for reconstruct their complex identifying the virus-derived antigenic peptides.

Three gene fragments were designed and synthesized according to the preference of E.coli.The gene of Cecropin P1 was amplified by PCR. The target gene fragment modified was cloned into vector T-easy, and then inserted into expression plasmid pcDNA3.1(+),the expression vector construction was completed. It was confirmed that the rombinant plasmid was successfully constructed by restriction endonuclease analysis and DNA sequencing, This may lay a foundation in research of antimicobial activities and antimicrobial mechanism in the gene of Cecropin P1.

The result of using the national standard to measure the alkaline phosphate (ALP)in milk is as follows: the activity of enzyme in the whole milk and skimmed milk is 403.345 μg/ml, 148.8147 μg/ml respectively;the coefficient of variation is 1.30%,2.99%, respectively.All these point this method is good and stable.The difference compounded to the national standard: the best dillution ratio of milk samples is 1∶25, the right consistency of phenol is 50 μg/ml. The conclusion by simulating pasteurization: ALP is inactived at 63℃ for 30 min.

IBV HN99strain was multiplied and purified by chick embryo kidney cell (CEKC), and the SPAFAS Jinan laboratory certified it was pure. Virus neutralization test (VN) revealed that the serotype of HN99strain was similar to IBV T strain. The inactivated vaccine of HN99 strain was prepared and it showed absolutely safety, moreover, chickens were immunized with the vaccine at different dose by muscle injection, three weeks later, 100 %protection were induced after challenging with HN99 strain, the results indicated that the IBV HN99 strain had fine Immunological Properties.

Based on the published sequences of picornavirus, 4 pairs of RT-PCR primer were designed. Total RNA were extracted from chick embryo allantoic fluid by RNAiso Reagent,one cDNA fragment were successfully amplified from total RNA extracted from DHV A66chick embryo allantoic fluid by using TakaRa
RNA PCR Kit(AMV)Ver3.0. After ligating cDNA to the plasmid vector pMD18T and introducing into E. coli DH5а, the recombinant plasmids were screened by ampicillin and checked by the restriction enzyme EcoRⅠ and SalⅠ digestion. Then the recombinant plasmids were sequenced. Compare and analyse of homologies of nucleotide showed that identities of DHV A66 and 3 published DHV were up to 96.2%~96.8%. This indicated partial cDNA library of DHV A66 strain were constructed successfully.

The antibody of S gallinarum in 3monthsold mothball breeding chicken which from a farm in Rongchang county, Chongqing city, were investigated by plate agglutination test (PAT). The result shows that the positive rate of
S gallinarum was 10.3% (115/1113), which includes 3.6% of X species cock, 1.3% of X species hen, 17.6% of Y species cock and 11.8% of Y species hen. Experiment demonstrates that the S gallinarum existed badly in this farm, furthermore, there was appreciable difference between mothball breeding chicken with different specie and gender, which were infected. 

The temperature in cowshed was drop down by adopting physieal measure, to insure cow's heath in summer and promote milk performance. 10 cows of stable milk performance were rardomly divided into two groups. For lower temperature, electric fans and water-pouring instruments were installed in cowshed. The results indicuted: compared wich CK, the temperature decreased by 10%,12% and 9.2% respectively. Through test, the humidity decreased by 0.33%, that of CK decreased by 0.16%, but hadn’t asigmficant difference(P>0.05).The results indicated: physical drop in temperature benefited improving dairy cow performance.

Pseudorabies continued to spread throughout the swine farms in China and caused large economic loss for Chinese swine industry, It cause reproductive failure in breeding herds and neurological disorder and high mortality in piglet. pseudorabies virus (PRV) is also an important primary pathogen in porcine respiratory disease syndrome (PRDC). 1940 serum samples from 46 large scaled swine farms located in 8 provinces were determined by differential PRV blocking gE-ELISA, it showed that 657 (33.87%) samples were positive for antibody to gE of PRV, and 25 swine farms were positive for antibody to gE of PRV. Among these swine farms, the highest positive rate of field PRV infection is 75.76% (125/165). While some well-managed swine farms which performed scientific PRV immune procedure were negative for virulent field PRV infection. Gene deleted PRV vaccine(AUSKIPRAGN) and commercial gE-ELISA reagent provide an ideal tool for the
control and eradication of pseudorabies. 

49 strains E Coli were separated from the disease meterals of several table chicken feeding factories in the contiguous area of Henan,Shandong and Hebei provinces,and 42 strains were confirmed as belonging to 11 serum types.The chief serum types are O2,O78,O1,O74,and O11,with the percentage of 26.2%,19.0%,11.9%,9.5% and 9.5% resp,which indicated the complexity of serum types in this area.The testing of sensitivity to drugs,showed that the 49 separated strains of E Coli are most Sensitivity to amikain florfenicol and cefotaxime,with the highsensitive strains reaching 94% ~100%.So they can be the chief choosed drugs to prevent and cure this epidemic disease.

For studying of the difference of cashmere quality in Alashan of Inner Mongolia, Chaqia town of Qinghai and Ping Liang Gansu, China, the cashmere was tested, which were collected from three areas respectively, that is Fineness, elongation, strength, length by Hand arranging, Short cashmere content. Results:the cashmere fineness , elongation rate and the length by Hand arranging are 16.14±1.73 μm , 46.18%±2.93% and 49.56±10.83 mm respectively, they are extremely notable (P<0.01) greater than which of the cashmere of Chaqia town of Qinghai and Ping Liang of Gansu.
