With the total RNA from C3-B7 hybridoma cells which secrete monoclonal antibody against NDV HN protein, the VH(heavy-chain variable region) and VL(lightchain variable region)genes encoding the monoclonal antibody were amplified by RT-PCR. The purified pruduct was cloned to pMD18-T vector and the recombined plasmids were transformed into DH5α. Colonies were selected and the specific recombinant plasmid was identified by colony PCR. The result shows that the VH gene encoding 119 aa and the VL gene encoding 110 aa were 357 bp and 332 bp, respectively.This establish the foundation for the construction and expression of single chain Fv(svFv).

377 isolates recovered from diseased chickens in northern region of Henan province from the year 2005 to 2007, which were conformed as escherichia coli strains by the biochemical test, were tested for antimicrobial susceptibility. All isolates displayed high resistance against FQs. The rates of resistance against enrofloxacin, ciprofloxacin, norfloxacin were 94.9%, 93.9%, 94.9%, respectively. 235 isolates which displayed high resistance against enrofloxacin (MIC>32 μg/ml) were further screened for the presence of the qnr genes using a PCRbased strategy. The results showed that there was only one strain in which qnrlike genes was positive among these detected strains. This qnrlike gene was named qnrA after sequencing analysis.

Splenic lymphocyte isolated and cultured from cattle spleen was exposed to different of levels of sodium fluoride in 24 h. Detecting GSH-Px activity, SOD activity, NOS avtivity and contents of MDA and NO of splenic lymphocyte. The results showed that GSH-Px activity and SOD activity were higher and other indexes were lower in the lymphocyte exposed to fluorine than control ones and there was doseeffect relationship. Conclusion: it is demonstrated that fluorine could degrade antioxdative activity, aggravate the level of lipid peroxidation, increase the content of free radicals, and induce the damage of lymphocyte.

PCR-RFLP and PCR methods were adopted to examine the polymorphisms of ESR gene and FSH-β gene in Meishan and Yorkshire Sows nucleus herds,The results showed that in Meishan nucleus herd the frequencies of A and B alleles of ESR gene were 0.353 and 0.647, the frequencies of A and B alleles of FSH-β gene were 0.836 and 0.164; In Yorkshire nucleus herd the frequencies of A and B alleles of ESR gene were 0.409 and 0.591, the frequencies of A and B alleles of FSH-β gene were 0.069 and 0.931.

In order to detect the distribution of myostatin(MSTN)in different tissues of Landaise goose, polymerase chain reaction(PCR) and sequencing was to be used. The results shown: the MSTN was detected in cerebrum, cerebellum, heart, liver, spleen, lung, kidney, pancreas, abdomen adipose, subcutaneous adipose,skeletal muscle.The wide distribution of MSTN gene in tissues reveals that it may have important physiological functions.

Melanin is distributed widely in many organisms including bacteria, fungi, plants and animals. Lighter variation soft pigmentation in humans is associated with diminished number, size, and density of melanosomes, the pigmented or ganelles of melanocytes. Here we reviewed that zebrafish golden mutants share these melanosomal changes and that golden encodes a putative cation exchanger SLC24A5 that localizes to an intracellular membrane, likely the melanosome or its precursor. The human ortholog is highly similar in sequence and functional in zebrafish. The evolutionarily conserved ancestral allele of a human coding polymorphism predominates in African and East Asian populations. In contrast, the variant allele is nearly fixed in European populations, is associated with a substantial reduction in regional heterozygosity, and correlates with lighter skin pigmentation in admixed populations, suggesting a key role for the SLC24A5 gene in human pigmentation. Finally, we reviewed research progress on melanin, SLC24A5 gene and Blackboned sheep. 

MicroRNAs (miRNA),one of the small moleculay RNA family, are single strand endogenous noncoding 21 to 23 nucleotide RNA molecules that keep high conservation in the process of evolution. Although they are tiny,In eukaryotes,microRNAs played important roles in the course of development and gene expression regulation, typically by forming perfect or imperfect duplexes with target messenger RNAs. In 2004, Epstein-Barr virus, was the first virus shown to encode and express miRNAs. Recently, miRNAs have also been cloned from Kaposi's sarcoma-associated herpesvirus, mouse γherpesvirus68 and human cytomegalovirus. In addition, the SV40 miRNA have been shown to negatively regulate large T-antigen (TAg) expression. This review will summarize recent findings on the prediction, expression and functions of miRNA and virally encoded miRNAs.

Embryonic stem cells (ES) have the unique capacity to proliferate extensively while maintaining pluripotency. ES cell lines can also be generated from human blastocyst embryos, and are considered promising donor sources for cell transplantation therapies for diseases such as juvenile diabetes, Parkinson’s disease, and heart failure. However, as for organ transplants, tissue rejection remains a significant concern for ES cell transplantation. Another concern is the use of human embryos. One possible means to avoid these issues is by reprogramming the nuclei of differentiated cells to ES celllike, pluripotent cells. Several extrinsic signals such as LIF, BMP and Wnt can support the selfrenewal and pluripotency of embryonic stem (ES) cells through regulating the “pluripotent genes”. A unique homeobox transcription factor, Nanog, is one of the key downstream effectors of these signals. Elevated level of Nanog can maintain the mouse ES cell self-renewal independent of LIF and enable human ES cell growth without feeder cells. In addition to the external signal pathways, intrinsic transcription factors such as FoxD3, P53 and Oct4 are also involved in regulating the expression of Nanog. Functionally, Nanog works together with other key pluripotent factors such as Oct4 and Sox2 to control a set of target genes that have important functions in ES cell pluripotency. These key factors form a regulatory network to support or limit each other’s expression level, which maintains the properties of ES cells.

Seven paternal half-sib families from Liangshan semi-fine wool sheep were used to detect quantitative trait loci (QTL) for birth weight. The population was genotyped for 11 microsatellite markers on ovine chromosome 1 by PCR. QTL analysis showed that quantitative trait loci (QTL) affecting birth weight was detected between ILSTS004 and CSSM004 in B3K5055 family. LOD was 3.338 and confidence level was 0.95~0.99.

The pharmacokinetics and residues of ciprofloxacin hydrochloride in the milk were investigated in 5 cows suffered from endometritis following singleintrauterine administration at the dosage of 2.5 per cow. Blood and milk were sampled at different intervals after administration and ciprofloxacin hydrochloride concentrations were determined by RP-HPLC method with a fluorescent detector. Difloxacin was used as intra-standard. The concentrationtime data were analyzed with MCPKP program. The ciprofloxacin hydrochloride concentrationtime data were fitted to a twocompartment open model with first order absorption in two cows suffered from endometritis after singleintrauterus. The main pharmacokinetics parameters were as follow: T 1/2α 1.26 h, T1/2β9.2 h, AUC 28.336 mg/L·h, β 0.3571 h-1. The ciprofloxacin hydrochloride concentration-time data were fitted to a three-compartment open model with nonabsorption in other three cows. The main pharmacokinetics parameters were as follow: T 1/2α0.916 h, T1/2β49.20 h, AUC 7.6296 mg/L·h,Clβ 1.582 L/kg·h, β 0.642 h-1. The results showed that ciprofloxaxin hydrochloride was absorbed fast and distributed extensively, but it was eliminated slowly in the blood after singleintrauterine administration.

In order to understanding the prevalence of intestinal parasites in sheep in Hennan province, 1265 sheep faecal samples from 5 farms were detected by the convention methods. The results showed that 5 groups parasites were detected in stool specimens, 2 groups were protozoon (coccidian, cryptosporidium), 2 groups were nematode (trichuris ovis,strongyloides stercoralis), 1 group was cestode . As a result,the average infection rate was 93.12%(1178/1265), thereinto, the infection rate of cryptosporidium sp was 3.10%, eimeria sp was 88.94%, trichuris sp was 10.83%, strongyloides sp was 59.11%, cestode was 43.96%,respectively. The results showed that sheep parasites mixed infections are more serious, in addition, the mixed infection of eimeria sp,tapeworm and strongyloides stercoralis took up 60.10% of the total infections rate. The results showed that sheep infection rate was reduction with the age increased.

The minimal inhibitory concentration (MICs)and the effects of reserpine on MICs of Ciprofloxacin against Staphylococcus aureus were determined using standard agar dilution method.Antibacterial activity of ciprofloxacin was ideal strengthened by reserpine.zone of inhibition of Staphylococcus aureus was enlarged by ciprofloxacin.Sensitivity of detect was advanced in organize.our investigation found that center concentration converted 0.5 μl/ml to 0.0625 μl/ml and limits of detection converted 0.125 μg/ml to 0.015625 μg/ml after reserpine. limits of detection was reduced 8 times.

In this article, 100 selected Yunling black goats were studied in 5 groups randomly and 20 goats in every group, using the method of single-factor experiment design in 5 grads according to the protein’s requirement of goats of NRC in the Lactation, with the aim of studying effect of diet pellets with different nutritional level on ewe of yunling black goat. The results could be concluded that as the whole ewe’s weight would decrease in lactation period, but with the increase of nutrition levels, the weight decrement would descent. At the same time, ewe lactation had a positive correlation with the level of nutrition.

Cornstalk silages of two varieties namely Jidan4011(JD4011) and Sidan29(SD29) were prepared with bankersilo. Digestibility and digestion rate (k) of organic cell wall (OCW) of cornstalk silage and cornstalk in the rumen were determined with in vitro method .The potential digestibility of DM and OCW of cornstalk silage was 57.4%, 48.9% for JD4011, and 58.8%,53.9% for SD29, respectively .The k value of OCW was markedly increased by ensiling and the respective value for the two types were 4.52% and 4.85% for the silage and cornstalk of JD4011, and 5.81% and 7.11% for the silage and cornstalk of SD29. The results confirmed that digestibility and digestion rate of OCW in cornstalk were improved by silage fermentation , the cornstalk with high OCW content was selected to make silage.
