The rumen was inhabited by numerous microbes,which form a complex microbial community that deeply affects ruminant physiology.This microbiota is dominated by relatively few divisions that are highly diverse at the strain/subspecies level,which together with its collective genomes (microbiome) provide us with genetic and metabolic attributes the ruminant have not been required.New studies taking advantage of molecular microecological and metagenomic are revealing how the rumen microbiota has coevolved with us and how it manipulates and complements our biology in ways that are mutually beneficial.However,how certain keystone members of the microbiota operate to maintain the stability and functional adaptability of this microbial organ still need to be answered,which necessitate the combination of high throughput methods and multivariable analysis.

Rumen regulation is one of the most important methods to improve feed efficiency and animal performance in ruminant industry. Changes of the high grain to forage rate in diet, protein composition in diet and physical state of feed effected ruminal fermentation in large extent. Supplemented with ionophores and organic acids could reduce methane production, and improve ruminal fermentation. Additionally, enzyme preparations, directfed microbials and extractions from plant also improved ruminal fermentation in different way. This paper summarized the last two years advance in rumen regulation, including diets control, supplemented with ionophores, enzyme preparations, organic acids, directfed microbials, extractions from plant, et al. Prospect and suggestion was referred in the end.

To study the effects of Robdosia serra (Maxim.) Hara on the growth performance and serum biochemical parameter of threeyellow chickens, 210 onedayold threeyellow chickens were divided randomly into 7 groups,and each group had 3 duplicate.GroupⅠwas given basal diet.The groupsⅡand Ⅲ were added 0.2% enrofloxacin and 1% ShuangHuangLian powder to the basal diet respectively. The groups Ⅳto Ⅶ were added 1%,2% and 4% Robdosia serra (Maxim.) Hara to basal diet respectively.After 49 days of feeding,the growth performance and serum biochemical parameter were determined.The results showed that,compared to the control group,added 2% Robdosia serra (Maxim.) Hara into the basal diet could remarkably increase the daily gain from 13.1% to 22.7%,and decrease the ratio of feed/gain from 8.1% to 14.2%, and the effect was better than ShuangHuangLian and enrofloxacin. 2% Robdosia serra (Maxim.) Hara could also increase the content of serum total protien, globulin and glucose,maintain the content of serum albumin at a stable level,and decrease the content of serum total cholesterol of threeyellow chickens which were 35 days old.

Substitute Ala for the sites of Ser 32 and Ser 36 in porcine IκBα gene,to establish the fundament for mechanism research of the mutant IκBα inhibition on delayed xenograft rejection.QuikChange SiteDirected Mutagenesis Kit was first uesd for the mutagenesis of Ser 32 site,then the kit was subsequently used for the mutagenesis of Ser 36 site,but failed. Finally OEPCR was used for the mutagenesis of Ser 36 site. These two mutagenesis methods were combined to achieve sitespecific mutagenesis at the sites of 〖JP2〗Ser 32 and Ser 36 in porcine IκBα gene.These two methods each has a unique,and each has their own shortcoming.So,in order to get better result by less work,mutation methods should be chosen flexibly according to the real need.

Prolactin receptor (PRLR) gene was studied as a candidate gene for the prolificacy in sheep. Two pairs of primers were designed to detect single nucleotide polymorphisms of exon 10 of PRLR gene in both nonseasonal estrous breed(Hu sheep) and seasonal estrous breed(China Merino, Romilly Hills and Romilly Hills×China Merino) by PCRSSCP.The result showed that the products amplified by primer P1, P2 displayed polymorphisms. For primer P1, sequencing revealed two mutations (53A→G and 81G→A) of PRLR gene in the genotype AB and only one mutation (53A→G) was found in the genotype BB in comparison with reported sequence.And these mutations did not cause any amino acid change.For primer P2, sequencing revealed one mutation(89T→C) in genotype CC,DD,CE,EF and this mutation resulted in an amino acid change of Pro→Leu.Another mutation (146C→G) of PRLR gene in the genotype DD and this mutation resulted in an amino acid change of Ala→Gly. One mutation (132G→A) was detected in the genotype EF and CE,this mutation resulted in no amino acid change. One mutation (167G→A) was also detected in the genotype EF and led to an amino acid change Pro→Leu. The result of χ2 independence test indicated that the distribution and constitution of genotypes at P1 and P2 of the exon 10 of PRLR gene showed significant differences among 4 sheep breeds(P<0.01).

Grampostive bacterium, identificated with Streptococcus suis serotype 2 by Rapid ID 32 Strep reagent of Streptococcus suis, was isolated in Guangxi. PCR assays were performed for detection of cps2J and three virulence factors, muramidaserelseased protein(MRP), extracelluar factor (EF) and suilysin gene(SLY) of Streptococcus suis. The sequence analysis revealed 98.8% homology to that of cps2J in GenBank. Virulence factors of Streptococcus suis serotype 2 were sly+mrp+ef+, which indicated that the cause of acute death of pigs was highvirulence Streptococcus suis serotype 2 in the pig farm. It was showed that determined bacteria caused infected rats death and leaded to infected rabbits showing temperature hoist to 40.3 ℃ and death due to blood poisoning. 

Karytype analyses were carried out in 309 pigs which were produced from three hybridized combinations of 13/17 Robertsonian translocation pigs by means of culturing whole peripheral blood.〖JP2〗 Three kinds of karyotypes appeared in their hybrids the normal karyotype pigs (2n=38), the heterozygous pigs (2n=37) and the homozygous pigs (2n=36). No mutation in exon 1 of porcine somatostatin gene was detected by means of PCRSSCP analysis in hybrids of three kinds of hybridized combinations while two single nucleotide polymorphisms (T520C,G521T) in 266~602 bp fragment of porcine somatostatin gene was detected by using the same method. But only two genotypes were detected: AA and AB. The result of genetic polymorphism analyses showed that the genotype frequency of AA(the limit of frequency was 0.58~0.77)and the allele frequency of A (the limit of frequency was 0.79~0.90)were both obviously higher than AB(0.23~0.42)and B(0.10~0.21) in hybrids of three kinds of hybridized combinations and pigs of different karyotypes. The genotype frequency of AB in the heterozygous pigs (0.42)were much higher than the genotype frequency of AB in the nomal pigs(0.25) and the homozygous pigs(0.40).The result of 2 test showed that genotype of all the populations reached HardyWeinberg equilibrium(P>0.05). 

To investigate the immuneregulatory effects of sargassum polysaccharide(SP) on mice. The immunosuppressed model in mice was induced by dexamethasone(Dex).The TI, SI, lysozyme concentration and ACP, SOD, XOD, CAT, NO, MDA in thymus and spleen were analyzed to investigate the immuneregulatory effects of SP. The TI, SI, lysozyme concentration, ACP, NO was increased in normal and immunosuppressed mice following treated with SP. The CAT activity in immunosuppressed mice and the SOD activity in normal mice was both enhanced by treatment of SP. Furthermore, the MDA concentration in spleen in normal mice and the XOD activity in thymus and spleen in immunosuppressed mice was also enhanced following treatment of SP. Therefor,the SP could improve the immune function of the mice.

The morphous and distribution rule of primoridial germ cells (PGCs)in chicken’s embryo was studied. The results showed that there were three aggregation peaks during the migration of the PGCs. When hatched 19~22 hours, most PGCs gathered at the germinal crescent. Then, when hatched 48~55 hours most PGCs migrated at the heart and dorsal aorta. At last, most PGCs ecesised at the ganadal ridge when hatched 4~5 days. The shape of PGCs in the germinal crescent liked round or orbicularovate, there were large glycogen granule in the cells. PGCs in the blood was round. And PGCs in the gonad were round or orbicularovate and had spurious. The nucleus was all round or orbicularovate at one side of the cell. The diameter of the cell was 10~22.5 μm and the nuclear was about 7.5 μm.

The superovulated donors of white cashmere goat were artificially inseminated (AI) with frozen sexed Xsemen to produce gender preselected embryos, then performed fresh embryo transfer. Results: ①The average numbers of ovum collection from the group of AI with frozen sorted semen and the group of fresh unsorted semen were 13.3 and 12.5 respectively. The rates of fertilization of these two groups were 29.6%(55/186) and 94.0% (281/299), the former was significantly lower than the latter(P<0.01). ②The lambing rates of gender preselected embryos transferring and untreated embryos transferring were 42.2% and 58.1% respectively. There was significant difference between the two groups (P<0.01). ③The rate of correct offspring in gender preselected embryos transfer group was 100%, which was significantly higher than untreated groups’57.4%(P<0.01). The results was proved the feasibility of gender preselection in offspring of white cashmere goat by sorted X and Ysperm, artificial insemination to produce embryos, and embryo transfer to obtain the kidlets.

The objective of this article was to investigate the effect of 4 different chemical activation reagents on the rate of development of bovine reconstructed embryos produced by intracytoplasmic direct injection. The reconstucted embryos were randomly divided into 4 groups to be activated:
Ionomycin+6DMAP; Ionomycin+Cycloheximide (CHX);7% ethanol+6DMAP;7% ethanol+CHX. The reconstructed embryos were activated for 5 min in Ionomycin and 7% ethanol. 6DMAP,4 h; CHX, 5 h. The reconstructed embryos were cocultured with cumulus cell monolayer. The rate of cleavage of the 4 groups were 52.6%,52.8%,53.8% and 54.2%, respectively. There was no significant difference among the 4 groups in terms of the rate of cleavage. The rate of blastocyst at day 8 were 20.0%,8.5%,10.2% and 6.1% for groupⅠ,Ⅱ,Ⅲ and Ⅳ, respectively. The rate of blastocyst of group Ⅰ was significantly higher than that of three other groups. The rate of hatching blastocyst of group Ⅰ showed very significant difference with that of group Ⅱ,Ⅲ and Ⅳ (10.7%,2.3%,3.0% and 1.8%, respectively, P<0.01). The results showed that all of the 4 kinds of activation reagents could successfully activated the reconstructed bovine embryos. The group Ⅰ gained the highest rate of blastocyst and hatching blastocyst, which demonstrated that Ionomycin+6DMAP benefited the development of bovine reconstructed embryos and could obtain an ideal rate of blastocyst. Ionomycin+6DMAP were ideal activation reagents for bovine reconstructed embryos produced by intracytoplasmic direct injection.

The effects of 17βestradiol (E2) on development of chicken primordial germ cells (PGCs) in vitro were investigated. Primordial germ cells, which were isolated from genital ridge of stage 28 chicken embryos with the method of EDTAtyrpsin collection, were purified by means of differential attachment technique and cultured in culture mediums of different 17βE2 concentrations (0 ng/ml, 5 ng/ml, 10 ng/ml and 20 ng/ml).The results indicated that there was no significant difference in development of PGCs between the group with 0 ng/ml and 5 ng/ml 17βE2. However, compared with the groups with 0 ng/ml and 5 ng/ml 17βE2, the groups with 10 ng/ml and 20 ng/ml 17βE2 had significant difference (P<0.05). But there was no significant difference in development of PGCs between the group with 10 ng/ml and 20 ng/ml 17βE2. The findings indicat that 17βestradiol of certain concentration (10 ng/ml, 20 ng/ml) promotes PGCs development in vitro.

Avian infectious bronchitis virus (M41,H120,HN99) were inoculated in chicken embryos.The eggs and the respective embryos were weighed and embryo:egg weight (EE) ratios were calculated within 6 days post inoculation.The EE ratios were compared with the uninoculated control embryos and EID50 could be calculated objectively. EE indices were also calculated by dividing the EE ratios of inoculated chicken embryos by the mean EE ratio of uninoculated controls. The results indicated that values of M41 and HN99 differ significantly from the respective fourth day EE ratio of uninoculated control embryos, and H120 differ significantly from the respective fifth day EE ratio of uninoculated control embryos, then the preliminary determination is to be infectioned by the infectious bronchitis virus. This method would be useful to observe embryo lesions especially in field strains of infectious bronchitis virus, which does not cause observable dwarfing until several embryo passages.

Relationship between amounts of antigens and effect of vaccine is important in the rseearch of new vaccine.The antigens(146S complete virion), inducing main neutralizing antibody of FMDV from vaccine,was abstracted to understand the relationship.Different contents of the antigen were inoculated into Guinea pig and the antibody response was studied by virus cell neutralization test. The results showed that antibody titer hadn’t increase when antigens over 1.5 μg, in opposite , the antibody titer decreased after booster immuniration when the contents of antigen over 15 μg.

On the basis of foot and mouth disease virus (FMDV) inactivated bivalen
t(O type, AsiaⅠ type) vaccines(called conventional FMDV vaccines blew ) using 206 adjuvant (France SEPPIC company), a double adjuvant inactivated bivalent FMDV vaccines was designed, it possessed stronger delayed release effect than conventional FMDV vaccines. According to the lymphocytes proliferation reaction (MTT), LPELISA and challenge protection experiments,the lymphocytes stimulation indices (SI=1.235±0.060) of double adjuvant vaccines group was higher than conventional FMDV vaccines group (SI=1.115±0.035) and control group (SI=1.010±0.045); compared with conventional FMDV vaccines group and control group, the celluar
celluar immunity level of double adjuvant vaccines group was significant than conventional one (P<0.05) and significant stronger than control one (P<0.01). However, as full dose immunity, antibody titer of conventional FMDV vaccines and double adjuvant vaccines group had little difference, 1/3 dose and 1/9 dose groups were especially lower than above two groups for less antigen and strong delayed release effect. The results of challenge protection efficiency showed that conventional adjuvant vaccines group was a little higher than the FMDV vaccines group, the former O strain was 11.84 PD50 and AsiaⅠ strain was 9.0PD50; the the latter O strain and AsiaⅠ strain all were 9.0 PD50. The results suggested that double adjuvant vaccines could induce better cellular immunity, possessed stronger delayed release effect and supported better challenge protection efficiency than conventional FMDV vaccines.

36 strains Escherichin coli were isolated and obtained from 7 pig farms in XiangFan. Through the bacterial colony and bacteria morphological observation, biochemistry reaction and animal experiment, the bacteria was identified as pathogenic E. coli.The drug sensitivity test was performed by disk diffusion method. The results showed susceptibility rate to Amikaain, Florfenicol, Gentamycin, Mequindox, Neomycin was 81%, 42%, 39%,33%,31%,repectively;resistance rate to Amoxicillin, Chloromycetin, Terramycin, Kanamycin, Gentamycin was 86%, 72%, 56%, 47%, 47%,respectively. According to above results, Amikacin was the most susceptible drug to against pathogenic E.coli, and Florfenicol was the secondary choice.

Nipah virus (Nipah virus) is a paramyxovius branch (Paramyxoviridae) nonsegmental singlestrand negative RNA virus, isolated and named for the first time in 1999 from the Malaysian town of Nipah encephalitis in the cerebrospinal fluid of patients. It is mainly against the central nervous system and respiratory system, causing acute fever, headache and different levels of consciousness. In this article,the diagnosis and treatment of canine spleen hematoma were analyzed and generalized, especially the operation was discussed for providing theories and directive meanings to diagnose and treat this disease.

The physical and chemical properties of wool were assayed, the correlations between these properties and the main economic traits were analyzed in super fine wool sheep and the wool qualities were evaluated by gray theory in different strains of Chinese Merino. The results show that: the average fiber diameter of super fine wool strain is significantly lower than the other strains(P<0.01), but the average wool curvature is significantly increased (P<0.05); there is no correlation between fiber diameter and the other traits (P>0.05); wool yield increase with the increase of wool cluster length (P<0.05); antistress elasticity is enhanced (P<0.01); there are significant positive correlations among clean wool’ glossiness, whiteness and yellowness (P<0.05). The comprehensive quality of wool of super fine wool strain is the best comparing to other strains.

Body composition traits of Chaohu ducks selected within the breed were tested and analyzed to breed the new meattype strains of Chaohu ducks. Sixty Chaohu ducks (thirty males and thirty females) with 10 weekages were slaughted and detected the carcass traits including carcass weight, eviscerated weight with giblets, eviscerated weight, breast muscle, leg muscle, abdominal fat, subcutaneous fat and so on. Statistics analysis was carried out for these traits. The results showed that there were no significant differences(P>0.05) between male and female ducks for the percentage of carcass weight, eviscerated weight, leg muscle weight, subcutaneous fat and the shearing forces of breast and leg muscle. The female ducks’ percentage of eviscerated weight with giblets, breast muscle and lean meat was significantly higher than that of the male ducks’ (P<0.05). The male ducks’ ratio of bones weight to meat weight was higher than the female ducks’ (P<0.05). Analysis of correlation revealed that there was a significantly positive correlation among the different section weight index (P<0.01). There was no sinificant correlation between the percentage of different section weight and the body weight(P>0.05). There was a significant correlation(P<0.05) between sex and the percentage of eviscerated weight with giblets, breast and leg muscle weight, and heart weight. The better carcass performance and qualities were found in the elementarily selected Chaohu duck population from the experiment. 

Taking Duroc×Landrace×Yorkshire hybrid pig as test material, this paper has studied the influence of adding biogas residue in diet on composition and content of intramuscular fatty acid. The results showed that besides the content of oleic acid in longissimus dorsi have significant differences, other fatty acids have no significant differences. Based on analysis adding biogas residue in diet will not bring adverse effect to pork flavor and other meat performance from the composition and content of intramuscular fatty acid, but should pay attention to the tendency of adding high ratio biogas residue in diet will lead to the increase of polyunsaturated fatty acids content, thus may cause meat quality becoming worse.

The left halfside longissmus doris, semitendinous muscle and middle gluteal muscle of 10 months bull calves of Xinjiang brown cow, Holsteins, Angus and Simmemtal male calf are separately chosen and analyzed about meat quality in the experiment. The result indicate the content of myoglobin is differente in variety breed calves’beef and the sequence is Holsteins>Xinjiang brown cow>Simmemtal>Angus that the difference is not remarkable (P>0.05). The Angus’ fat content is highest and collogen content is lowest in four breed that the difference is remarkable (P<0.05). Myoglobin, crude fat, crude protein content and dehydration rate are different in the same variety and different sites, the difference is unremarkable (P>0.05), but it has the certain rule, the content of myoglobin in semitendinous muscle and middle gluteal muscle content is higher than longissmus doris. Collagen protein content of longissmus doris is lower than in middle gluteal muscle and semitendinous muscle on same variety and different sites.

To evaluate emergency work of animal epidemic prevention after earthquake of Mianyang that belongs to earthquake stricken area symposium, looking up documents of animal disease prevention,checking the work of emergence response, survey and experimental methods were taken.The results showed that the leaders attached importance to emergency work of animal epidemic prevention after earthquake diseaster and cooperated with other departments timely to prevent major animal epidemics.The animal epidemic prevention and control have brought noticeable achievements by using scientific normative emergency treatment in time.There is no prevalention and potential risk of major animal epidemics and zoonoses. But there are few factors to cause animal disease in local places.
