Effects of Sodium Gluconate and Phytase on Phosphorus Utilization in Broilers during 1 to 3 Weeks

One experiment was conducted using a total of 350 one week old Arbor Acre male chicks to investigate the effects of sodium gluconate (NaGlu, 0 and 2%) and phytase (0, 500 and 1000 U/kg) on phosphorus utilization. A 2×3 plus 1 factorial arrangement of treatments was adopted. A positive control diet, adequate in nonphytate P and Ca without NaGlu and phytase, was used. Chickens were randomly assigned by body weight to seven treatments and each dietary treatment was replicated 5 times. The experiment lasted from week 1 to 3 posthatching. The results showed that adding 2% NaGlu improved apparent retention of Ca (P=0.003and P (P<0.001) and decreased serum alkaline phosphatase activity (P=0.038)of chicks. Supplementation of phytase increased tibia ash weight (P<0.001), apparent retention of Ca(P=0.005and P (P<0.001). There was a significant interaction between NaGlu and phytase for tibia ash weight (P=0.030) and P apparent retention (P=0.008).

In this study, local sheldrakes were randomly arranged two groups, one of which were stocked in selenium- rich land and fed with selenium- rich rice, and the other group were stocked in seleniumless land and fed with compound feed. Then determine the amount of selenium in pectoral muscle and leg muscle of all sheldrakes. The results indicated that the amount of selenium in muscle of sheldrakes, stocked in selenium- rich land and fed with selenium- rich rice, was extremely higher than that of the other group (P<0.01). 

The objective of this experiment was to research the effects of four different protein dietary A, B, C and D, which were soybean meal, cottonseed meal, rapeseed meal and DDGS respectively on rumen fermentation and the synthesis of microprotein. Treatments were arranged as a 4×4 Latin square design, and four goats (30±2.5) kg fitted with permanent ruminal cannulaes. The results indicated that there was not significant difference among four groups on rumen pH (P>0.05); the NH3-N concentration of group A was higher significantly than that of group D (P<0.05). There was significant difference among four groups on the yield of MCP, the yield of group A is highest, it is higher significantly than that of groups B and C (P<0.05), and higher significantly than that of group D (P<0.01). There was significant difference among four groups on acetic acid, propionate acid, butyric acid concentration, and total VFA concentration of groups A and B were higher than groups C and D (P<0.05).

20 castrated Tibetan sheep which were randomly divided into four treatments (group Ⅰ, group Ⅱ, group Ⅲ and groupⅣ)were adopted to investigate the effect of different concentrate levels(150,300,450 g/d) and silage on production performance. The results indicated that average daily gain(ADG)of four groups were (41.00±5.48),(63.00±8.37),(68.00±9.75),(69.00±9.2) g respectively.ADG of group Ⅳ was higher than that of group Ⅰ,significantly(P<0.01), ADG of group Ⅱ and group Ⅲ were higher than that of group Ⅰ,significantly(P<0.05), there was no significant difference among group Ⅱ, group Ⅲ and groupⅣ. The highest feed efficiency showed at 300 g/d concentrate leve1(group Ⅲ), and group Ⅱ (the silage) showed to be more economical than group Ⅲ.

A study was conducted to investigate effects of environmental enrichment and stocking density on performance,serum cortisol concentrations, meat quality and sarcous pH of sheep, twenty four small tail han sheep were assigned to three treatments: ①confinement in a pen (A); ②confinement in a pen with a yard (B); ③confinement in a pen with a yard and environmental enrichment as well (C).The results showed that environmental enrichment and stocking density had no significant effects on growth performances(P>0.05), although they tended to improve it. A highly significant difference in surface cleanness existed among groups (P<0.01) and at Day 37, environmental enrichment and stocking density significantly significantly decreased serum cortisol concentration (P<0.05). Environmental enrichment and high stocking density did not affect meat yield and pH 45 min, pHult of sarcous (P>0.05). The results indicated that environmental enrichment and stocking density can improve sheep welfare.

55 Kazak male lambs chosed, body weight and body size were carried out at the north bank of Tekas natural grassland, Zhaosu horse farm, Yili Kazak Autonomous Prefecture in Xinjiang from May 15 to September 28, 2008. 20 lambs were selected for digestion. The results showed that the entire research period, there was a fluctuation in the growth of lambs pastured as the seasonal variations in forage. The growth of lambs reached the highest level at 5 months and 7 months, respectively. The body weight gain overally increased. There was significant difference in the average body weight gain of the lamb aging 7 or 8 months (P<0.05). However, there was obvious significant difference in average body weight gain of the remaining lamb (P<0.01). The monthly average daily gain were (187.8±55.7) g,(100.1±44.7) g,(174.6±45.2) g and (82.1±36.0) g, respectively.Growth rate of pastured lamb body size of 5 months old lamb was bigger than that of 6 months old one, 7 months old lamb was bigger than that of 8 months old. In addition to thoracic deep, the order of the cumulative growth rate of the pastured lamb body size from big to small were chest, body length, body height, thoracic deep, wide chest, tube circuit, respectively. The DM, OM, CP digestion rate of 5 months old lamb were significantly higher than that of 4 months old one(P<0.01). There was decrease in DM digestion rate between 5 and 7 months, the difference between 5 and 7 months was significant (P<0.01). There was consistent in OM digestion rate between 6 and 7 months, there was biggest when 8 months. There was decrease in CP digestion rate between 5 and 8 months, there was no significantly difference between 7 and 8 months, However, there was significant difference in the CP digestion rate for the rest of the months (P<0.01).

To study histochemical and ultrastructural effects of Se deficiency on mast cells of jejunums in the chicken. 10
chicks were randomly divided into two groups (control group and Se deficiency group). The chicks were killed by decapitation at 75 d and then the jejunums were collected. Microstructure and ultrastructure of jejunums were observed using routine toluidine blue(RTB),long toluidine blue(LTB),alcian blue safranin (AB-S), aldehyde fuchsin alcian blue(AF-AB), alcian blue-periodic acid Schiff reaction(AB-PAS)and transmission electron microscope technology. Results showed that these mast cells had heterogeneity in quantity,shapes,sizes,colouration,mucopolysaccharides with the different staining methods, it may hint that these cells were divided into different subtypes. The amount of mast cells increased obviously in Se deficiency group, it shows that mast cells are one of the substaintial immunocyte and play an important part in immunological regulation. Under transmission electron microscope,abundant homogeneous cytoplasmic granules of different electron density were seen in cytolymph, nucleolus was spherical or oval, complexs of Golgi and major mitochondrias were observed in periplast. In Se difeciency group,it was seen that MC increased significantly and were close together with degenerating cells,heterochromatin increased, nucleus became unsymmetric, periplast was sparse and vacuoles were observed after cytoplasmic granules released the content.

Under transmission electron microscope, the prehypophyseal cells in F1 hybrid-marrah buffalo have five marked cells types in common, their ultrastructural features appeared characteristically. Gonadotrophs are round in shape, which about 7.6 to 9.0 μm in diameter; the size of secretory granules is either big or small clearly, which has high electron density. Thyrotroph is about 6.0 to 7.5 μm in diameter and contains irregular nuclei in the centre mostly; their secretory granules are generally minor and tiny,distributed in the vicinity of plasmalemma. Adrenocorticotrophic hormone(ACTH)cells are ovoid in shape and 7.1 to 8.5 μm in diameter; irregular nuclei always is side incline; their secretory granules occurred in cytoplasmic periphery or one side. Somatotrophs are round, containing rounded nuclei and numerous secretory granules. Lactotrophs are almost the biggest cells in pituitary, numerous cells of various size are full of periplast. The results aimed to provide the morphology basic data for controlling reproduction endocrine in F1 hybrid-marrah buffalo.

To discuss the effects of the recombinant interleukin-2 on the immune function of the Gushi chicken, the gross anatomy of technology and the immunity technology were applied to investigate the impacts of the recombinant interleukin-2 on immune function of Gushi chicken. The antibody levels in the every immunized group which was injected interleukin-2 were higher than the average Gushi chicken Shigella vaccine group, and they were notably higher than the control group. The antibody levels of all the groups in the second to third weeks achieved the highest level, and they remained a constant level in the fourth to seventh weeks. The spleen index, the thymus index and the IBD index in the every immunized group which was injected interleukin-2 were basically higher than the control group and chicken Shigella vaccine group. Gushi chicken recombinant interleukin-2 could promote the rapid growth and maturity of the chicken immune organ, and could enhance the immune function of chicken to resist all kinds of infection of pathogenic microbiology.

A cDNA library for Moniezia expansa was constructed by switching mechanism at 5′ end of RNA transcript (SMART) technique using SMARTTM cDNA Library Construction Kit (Clontech). The total RNA was extracted from M.expansa using Trizol Reagent and mRNA was purified using Oligotex mRNA Kits. Single-strand cDNA was synthesized using PowerScriptTM reverse transcriptase, and double-strand cDNA was synthesized and amplified by long-distance PCR. The PCR products were digested by proteinase K. After digestion with SfiⅠ and size fractionation using SPIN-400TM columns, SMART cDNA was ligated to pBluescript II SK* vector. The ligation mixture was transformed into E.coli DH5α. The recombination rate of the library was 94.7%, and the titre was 2.52×105 PFU/mL. The size of inserts was about 0.5 to 3 kb, and the average length was 1.2 kb. 2642 ESTs from 5′-ends of the cDNA clones representing 1081 unigenes were obtained. The result displays M.expansa cDNA library is successfully constructed,and could serve as a valuable resource for screening and isolating specific genes for M.expansa.

In the north bank of Turks river the YiLi natural grassland field select 20 4-month-old Kazakh lambs with the similar weight (24.71 kg±2.07 kg),from 2008 May 15 to September 20 to test grazing lamb growth, feed intake determination, serum biochemical and hormonal. The results showed that :4 to 8-month-old lambs with average daily weight gain: (175±34) g, (92±36) g, (158±46) g, (81±36) g, lamb daily weight gain has two peaks: 4 to 5 months old and 6 to 7 months old, which were significantly higher than both 5~6 month old lambs and 7to 8 month old lambs (P<0.05), the changes in serum biochemical markers: there is no significant difference about ALP, LDH, GOT content between months (P>0.05); TP content 5 to 7 months higher than 4 to 8 months, and significantly higher than 6 months old lamb (P<0.05); ALB levels of 6,7,8 months old twere significantly higher than 4,5 months old lamb (P<0.05); GLB content of 5,7 months old lamb are higher than 4,6,8 month old lamb, 7-months-old lamb was significantly higher than 6 months old lamb (P<0.05); A/G, 4,6 month old lamb was higher than 5,8 months old lamb, and which was also significantly higher than the 7-month-old lamb (P<0.05);BUN content, there was no significant difference among 4,5,7,8 months old lamb (P>0.05), but every of them was significantly higher than 6 months old lamb (P<0.05); GLU content, 5,7 months old lamb were significantly higher than 4,6,8 months old lamb (P<0.05); TC, GPT, LDL-C, HDL-C content,4 to 8-month-old lamb gradually increased (HDL-C 5-month-old lamb decreased slightly), TC, GPT content difference between adjacent months was not significant (P>0.05), LDL-C, HDL-C content of 7 to 8 months old lamb were significantly higher than 4,5,6 months old lamb (P<0.05); TG content, the whole period with downward trend, only the adjacent inter—month—old lamb 7 months old lamb were significantly higher than the 8-month-old lamb (P<0.05); FFA content, 8-month-old lamb was significantly higher than the remaining months old lamb (P<0.05), 6-month-old lamb was higher than the 4-month-old lamb, and which was significantly higher than 5, 7-month-old lamb (P<0.01).

In order to evaluate the effects of supplemental levels of dietary vitamin E and immune stress on serum concentrations of corticosterone (CORT) of chickens, 540 one-day-old chickens (White Leghorn, WL) were fed diets with different vitamin E levels (0, 40 and 100 IU/kg). A completely randomized experimental design in a 3×3 factorial arrangement with 5 replications of 12 chickens per treatment was applied. Three different vaccinating ways were those for the first time at 5, 8 and 11 d respectively. The kinds of vaccines and the subsequent vaccinating intervals were identical. There were 5 repetitions per treatment, each containing 12 chickens. At the metaphase of experiment, each chicken was injected celiacly with E. coli lipopolysaccharide (LPS, 0.05 mg/chicken). Results showed that the supplementation of dietary vitamin E had significant impact on the serum concentrations of CORT.

VP1 gene of foot-and-mouth virus (FMDV) AsiaⅠ was expressed in insect cell/Baculovirus expression system to lay a foundation for research of FMDV AsiaⅠ VP1 protein function and serology diagnostic method. The gene VP1 was amplified from the recombinant plasmid of FMDV AsiaⅠ by PCR and inserted into Baculovirus vector pFastBacHTA to get recombinant plasmid pFastBacHTA-VP1. The recombinant plasmid pFastBacHTA-VP1 was transformed into DH10Bac and the VP1 gene was integrated into Bacmid by site-specific transposition. Subsequently, the recombinant Bacmid-VP1 was transfected into Sf9 insect cells. The expressed product was identified by SDS-PAGE and Western blotting. The soluble fusion protein was purified by Ni-NTA affinity chromatography. ELISA analysis showed that the protein purified was reactive with the positive serum against FMDV AsiaⅠ. The successful expression of FMDV AsiaⅠ VP1 gene in insect cell/Baculovirus expression system provide material for research of antigenicity of AsiaⅠ VP1 protein as well as the surveillance of serology antibody.

96 matrix antibiotic susceptibility testing kit was designed based on agar dilution method. According to National Committee for Clinical Laboratory Standards and the kid, susceptibility of 98 E.coli strains isolated from raisers in the main large scale farms, students of kindergarten and middle school to Enrofloxacin, Ciprofloxacin, Norfloxacin and Sanhuangtang. The results showed that the MBC and MIC was high symmetry of two methods. The average of difference was in one dilution range respectively and the coincident ratio all reached 90% in 0.5 μg/mL difference range. The range of relativity was 0.94 to 0.99 of MBC and MIC. We found that the parallelism of MBC and MIC was high from the index. The results of the two methods were closely compared to the total frequency of susceptibility, the coincident ratio was 93.93%. 96 matrix antibiotic susceptibility testing kit can determine MIC of Sanhuangtang exactly ranged from 125 to 500 mg/mL. We can conclude that 96 matrix antibiotic susceptibility testing kit is a new kit which can adapt to large scale lab antibiotic susceptibility determination that simple, exact and fast, large flux, economize, which can test 96 bacteria for one antibiotic or Chinese traditional medicine combined with 96 holes incubated board one time.

A recombinant Doppel plasmid without glycosyl phosphatidyl inositol (pDoppel 1-155) was digested by Not Ⅰ,then the objective gene was recovered and purified. The gene was introduced into male pronucleus of BALB/c mouse fertilized ovum by microinjection, then we got 32 mice which have been injected the Dpl 1-155 gene, in which 6 was positive by PCR; one F0 generation of Tg mouse expressing Dpl without GPI was mated with a wild-type mouse, then 21 F1 generation mouse were got, in which 10 were positive by PCR identification. Brain tissue of three F1 generation mice which were positive by PCR identification was collected, and was identified by Western blotting, there was 17 ku Dpl 1-155 protein expressed in the brain. So we have generated Tg mice expressing Dpl without GPI which provides Tg animal models for investigating the bionomics of Dpl protein.

SPA labeling technique for the gold-labled pond, BP26 purified by affinity chromatography for the detection antigen(T line), the colloidal gold test strip was designed to detect Brucellosis antibody, to discern infected mouse by Brucella abortus S19 and vaccinated mouse by BP26 KO Brucella strain YZ-2. There was no cross-reaction between the Brucella and the bacteria which have highly homology to Brucella for the strip test. Compared to other general tests, the sensitivity of the strip test is more than RBPT, and similar to ELISA; the coincidence of test results with ELISA was 100%, and the strip test was obviously more convenient. The colloidal gold test strip have the potential application to discern infected and vaccinated animals by Brucella.

The objectives of the present study were firstly to detect polymorphisms in the follicle stimulating hormone receptor (FSHR) gene, which provide a scientific basis for marker assisted selection for high prolificacy in Wenchang chicken. In this study, the FSHR gene as the candidate gene of chicken’s reproductive trait was analysis on its regions which encode most of the extracellular domain by PCR-SSCP. The result showed that 4 SNPs sites were found at region 2, region 4, region 6, region 9, respectively. They are C-49→T-49 mutation before the CDS’ 5-flanking terminal of the exon2, T43→C43 mutation in the CDS of the exon4, A12→G12 mutation after the CDS’ 3-flanking terminal of the exon6, G38→T38 mutation after the CDS’ 3-flanking terminal of the exon8. The fitness test indicated the gene frequency were all in Hardy-Weinberg balanced state (P>0.05). In conclusions, the polymorphisms of theses sites lay a foundation for the further research on relationship between the polymorphisms of FSHR gene and the reproductive performance of Wenchang chicken.

A pair of primers was designed and synthesized according to the bovine Hepcidin gene sequence in GenBank. Coding sequence of Hepcidin gene was amplified by RT-PCR technology from total RNA of yak’s live, the sequence of Hepcidin was analysed and phylogenetic tree of Hepcidin protein was constructed. The results showed that a 322 bp cDNA sequence of Hepcidin gene was cloned, including 289 bp open reading frames, encoding 82 amino acids which contained 22 amino acids coded signal peptide. Forecast for protein molecular weight and isoelectric points, was 8.88 ku and 9.24 respectively. This gene region had high conservation in the bovine species, compared with the ordinary cow, the homology of Hepcidin sequence was up to 99 percent.The phylogenetic tree conformed to the laws of species evolution. The research laid an important foundation on the full-length cDNA gene cloning and study.

Five Newcastle disease virus strains were isolated from chickens and pigeon of poultry farm in Xinjiang. The results of the hemagglutination test (HA) and hemagglutination inhabition test (HI) indicated that the 5 NDV strains had HA. The HA of strains all can be inhibited by NDV positive serum and can’t be inhibited by AIV (H5 subtype and H9 subtypes) positive serum. In this study, the F gene of strain XJ/3/07, strain XJ/7/07, strain XJ/9/08, strain XJ/10/08 and strain XJ/11/08 were amplified by RT-PCR and cloned. The sequence analysis showed that the full-length gene of F were 1662 bp, 1589 bp, 1676 bp, 1589 bp and 1662 bp and encoding 553,528,553,520 and 553 amino acids respectively. According to the analysis 〖JP+3〗of amino acid sequence of the cleavage site of F protein, the NDV strain XJ/10/08 was virulent and the others NDV strains were avirulent. Comparison the five NDV strains with the vaccine strain La Sota and strain TaiWan95, the nucleotide homology were 83.7% to 99.8% and 84.7% to 93.3%. The phylogenetic analysis suggested that the strain XJ/3/07, strain XJ/7/07, strain XJ/9/08 and strain XJ/11/08 were gene type Ⅱ, and the strain XJ/10/08 strain was gene type Ⅶ.

To study and modify the optimal methods of silver staining of DNA polyacrylamide gels electrophoresis, polymerase chain reaction (PCR)-polyacrylamide gel electrophoresis silver nitrate (AgNO3) staining were used to detect DNA fragments and pBR322 MspⅠDNA molecular weight. 2 silver staining methods were analyzed and evaluated. The sensitivity of all the band of pBR322 MspⅠ Marker developed was coequally 25 ng. However, comparison of time and step,advanced technique was more convenient. The advanced staining cost only about 25 minutes, and was characterized as simple steps,high sensitivity,little pollution,which was recycled and valuable for practical apply.

In this study, the genetic diversity of 232 samples of superfine merino sheep were studied, by using eight microsatellite markers mapped to sheep chromosome 1 and chromosome 3. Allele frequencies(p), mean heterozygosity(H),polymorphisms information content(PIC), and average effective number of alleles(N)were calculated. And various phenoltype including wool fibre diameter(WFD), fibre diameter dispersion(FD-D), wool fibre absolute strength(WAS), wool fibre relative strength(WRS), wool strength disperse(WSD), wool natural length(WNL), wool extend length(WEL) and fibre waviness(FW) were measured in the flock. Correlation analysis of Microsatellite loci with wool phenotype showed: there were great correlation between microsatellite BL-4 and WEL,microsatellite KRT2-13 and WEL(P<0.01), microsatellite Oarcp43 and WSD(P<0.01),microsatellite Bms1248 and WFD(P<0.01),the correlation between microsatellite Fcb5 and FW was significant(P<0.05), microsatellite OarDB6 and WAS was significant(P<0.05).

In this experiment, morula and blastocyst were bisected in three different partition liquid with micromanipulator.Demi-embryos were cultured and then the development rate was observed. Bisected embryos were co-transferred with trophoblastic vesicles (TRV).The results showed that morula were bisected in PBS +0.2 mol/L sucrose and PBS+5% PVP,splitting success rate significantly higher than in PBS (89.13%,86.73% VS 69.67%) (P<0.05),and the blastocysts rate and pregnancy rate were no significant difference (P>0.05); in PBS +0.2 mol/L sucrose and PBS+5% PVP in the bisection of blastocysts,splitting success rate significantly higher than in PBS (94.52%,92.52% VS 70.52%) (P<0.05), the blastocysts rate and pregnancy rate were no significant difference (P>0.05); the PBS were added in 0.2 mol/L sucrose and 5% PVP will help to improve splitting success rate of the dairy cow morula and blastocyst.

Most of the genes encoding inflammatory proteins involved in neutrophil migration and activation have been shown to contain κB sites for the NF-κB within their promoter region and therefore partly depend on NF-κB for their expression,suggesting that this transcription factor might play a critical roles in the pathophysiology of mastitis. In the experiment 5 Theileria annulata infected dairy cows and 5 healthy dairy cows were selected and RBC-C3bRR rate,RBC-ICR,the percentage of T Ea rosette and the lymphocyte transformation rate were measured respectively in 0th,5th,10th,15th,and 20th days.The result showed that all these indexes of the infected diary cow were significantly lower than those of healthy ones. It indicated that Theileria annulata inhibited the immune level of diary cow obviously, and with the changes of detect time,the immune level decreased gradually. 

The virus isolated from duck was identified as Newcastle disease virus by the HA/HI. The intracerebral pathogenicity index in 1-day-old chickens, the intravenous pathogenicity 6-week-old chickens were 1.79 and 2.45 according to the standard and methods of Newcastle disease pathogenicity judgment refer to OIE. We amplified its fusion protein gene by RT-PCR using the primer which got from the reported sequence of NDV in GenBank, cloned into the carrier and sequenced. The results showed that the isolated virus was the strong virulence NDV.

368 swine serum samples collected from some districts in Liaoning province were examined for antibodies against swine hemagglutinating encephalomyelitis virus (HEV) by hemagglutination inhibition test. The positive rates of antibodies against HEV was 44.3%. There was on clinical sign of HEV infection in any districts. The results showed that pigs had been infected subclinically by HEV in those districts.

A multiplex PCR assay for detection Streptococcus suis (S. suis, SS) serotypes 1, 7, 9 (SS1, SS7, SS9) was developed using the primers designed basing on the gene coding for CPS1I, CPS7H and CPS9H coding for the capsule (CPS) of SS1, SS7 and SS9, respectively. The DNA of standard SS1, SS7 and SS9 strains were used as the positive control to establish the multiplex PCR assay. The sensitivity, specificity and repetition assay of multiplex PCR were tested, and 11 purified S. suis strains of culture samples taken from clinic suspicious S. suis infected pigs and having been testified by routine bacterial isolating and identification were detected by the established multiplex PCR assay. The results indicated that the multiplex PCR assay was successfully established. The specificity and the sensitivity of the multiplex PCR assay revealed that the multiplex PCR threshold was 100 CFU/mL of S. suis, and no products were amplified from the genomic DNA of the standard SS2 strain and the other 6 kinds of pathogenic bacterial microorganism acting as the negative control. The repetition test indicated that the duplex PCR was repeatable. Total 3, 2, 1 of 11 purified S. suis strains of culture samples displied the CPS9H, CPS7H, CPS1I positive, respectively. The results showed that the established multiplex PCR method was highly specific and sensitive,and was suited to clinic rapid diagnosing of S. suis serotypes1, 7 and 9.

Colloidal gold was prepared with the tri-sodium citrate reduction method. To develop gold immunochromatographic assay for detection of specific antibodies of porcine circovirus type 2, protein A labelled colloidal gold was sprayed onto a conjugate pad, meanwhile PCV-2 capid protein and anti-protein A antibodies were striped in the test line position and in the control line position respectively. The color was appeared both in the test line position and in the control line position when the sample was positive. The color was appeared only in the control line postion when the sample was negative. Compared with ELISA, the sensitivity and specificity of the assay were 89.19% and 100% respectively. No cross-reactivity with PCV-1,HCV,PRV,PRRSV,PPV positive serum samples was observed.

Specimens were collected from sick or dead ducks from 9 aquatic farms of 5 provinces in China. 67 suspected strains were purified and then identified by VITEK-32 as E.coli.The results of bacterial susceptibility experiments by the broth micro-dilution test indicated that the tested bacteria presented 56.7%, 71.6%, 31.3%, 49.3%, 50.7%, 71.6%, 62.7%,0% and 56.7% resistance to Ceftriaxone Sodium, Ceftiofur, Cefepime, Cefotaxime, Levofloxacin, Ciprofloxacin, Gatifloxacin, Imipenem, Aztreonam, respectively. The resistant condition of the duck E.coli to the above-mentioned drugs was severely and cross-resistance between analogues should not be neglected.

The experiment was conducted to use three dosage levels of ceftiofur suspension (10,5,2.5 mg/kg) with twice in 72 h and ceftiofur sodium injection (10 mg/kg) with twice in one day for 3 days to cure piglet pleuropneumonia by intramuscular injection.The results showed that both ceftiofur suspension of three inclusion levels and ceftiofur sodium injection as medicine control group have significantly therapeutic efficacy on piglet contagiows pleuropneumonia and the survival rate was up to 80% to 100%, which was distinguished higher level than the infection control group(P<0.01).Especially,the high and the medium level of ceftiofur suspension had higher survival rate than the low levels of ceftiofur suspension and ceftiofur sodium injection(P<0.01).Artificial infection experiment indicated that ceftiofur suspension could reduce clinical symptom and decrease mortality.The levels of ceftiofur suspension(5 to 10 mg/kg) had better effection as injection on curing Actinobacillus pleuropneumoniae.

The results were as follow: 30.11%(243/807) checked sera antibody titers were ≥1∶8 which was regarded as positive.The positive rates in sows,postweaning piglets(40-day-old),growing pigs(80 to 100-day-old),growing pigs(150 to 180-day-old) were 56.94%(123/216),9.04%(17/188),27.65%(60/217) and 23.12%(43/186), respectively. In order to analyze the differences between RT-PCR and ELISA antigen detection methods in classical swine fever virus detection,120 clinical samples were detected by the two methods. According to the study released by the NCBI swine fever virus strain Shi-men E2 gene sequence, design and synthesis of a pair of detection of classical swine fever virus E2 gene PCR primer. Submitted to the 120 suspected swine fever disease compound extracted total RNA, after one-step RT-PCR amplification of E2 gene, checked out 32 samples were expected to contain swine fever antigen, positive rate was 26.7%. ELISA antigen detection kit checked out 38 samples were expected to contain swine fever antigen, positive rate was 31.7%. One-step RT-PCR detected 32 positive samples were also positive in the detection of ELISA. The establishment of classical swine fever virus RT-PCR detection of nucleic acid and antigen detection ELISA. Pathogen detection of classical swine fever supplied technical support for the provision of Yunnan.

This paper had studied the performance that the native yellow cattle was improved by Gelbvich and Belgian Blue using frozen wheatstraw semen which was bought from the beef engineering center of Henan province in 2003 to 2006.And showed that the 6 months development performance of F1 crossbred of Gelbvich or Belgian Blue were higher than the native’s. The male body weight of F1 crossbred of Gelbvich was 177.45 kg,and the F1 crossbred of Belgian Blue’s was 118.26 kg,and the native’s was 113.50 kg.Simultaneously those females body weight were 156.81 kg and 108.86 kg and 97.57 kg resp.

Based on the electrical bioimpedance theory and the latest technology, principle, software and hardware circuit of the oestrus detector are designed. The experiment results showed that the detector can accurately measure the electrical bioimpedance of the vaginal secretes of the oestrus bitch. The detector is of high precision, small volume, and high display stability, and overcome the shortcomings of the same type of equipment at home and abroad.
