Abstract: Continuous improvement in both culture-dependent and-independent tools of microbial diversity analysis and extending of animal species into minor ruminant not only add tremendous information into the public database of rumen microbial diversity, but also reveal that the structure of rumen microflora is affected by host genotype, growth development and health conditions and demonstrate a dynamic character with ratio regime and season. In the functional point, fiber degradation, methanogenesis and biohydrogenation of unsaturated fatty acid and related microbial issues were highlighted from physiological, genomic or metagenomic point of view. Although much progress has been made in 2009, how certain keystone members of the microbiota operate to maintain the stability and functional adaptability of this microbial organ still need to be answered which necessitate the combination of high throughput methods and multivariable analysis.

Abstract: This review summarized the research progress of feed additives in rumen fermentation in 2009. Ionophores, such as monensin alters volatile fatty acid (VFA) production in the rumen towards more propionate (Pr) production at the expense of acetate (Ac), which results in increased glucose availability to the animal through gluconeogenesis, decreases the production of methane by inhibiting H2-producing bacteria but not directly inhibiting methanogens. Ionophores decrease milk fat content by an incomplete biohydrogenation process of long-chain fatty acids in the rumen that interferes with de novo fat synthesis in the mammary gland. Adding fibrolytic enzymes and amylase to ruminants can improve the nutrient digestibility. Saponin decreases the production of methane by inhibiting rumen ciliate protozoa. The appearance and proliferation of tannin-tolerant bacterial species to enhance their tolerance to tannins and only condensed tannins extract influence the ruminal biohydrogenation. Essential oils reduce protein and starch degradation and an inhibition of amino acid degradation in rumen due to the selective action on certain rumen micro-organisms. DFM include yeast, lactic acid bacteria, Bacillus can increase in nutrient digestibility, alteration of the proportion of volatile fatty acids produced in the rumen, reduction in ruminal ammonia, and increase of ruminal microorganism population. But the beneficial effects of yeast products depend on lactation stage, diet characteristic and environmental condition.

Abstract:In this experiment fifty-four growing pigs were conducted to study the effects of Escherichia coli phytase and Aspergillus niger phytase substitute Calcium phosphate in diet on the growth performance,nutrient utilization,serum biochemical parameters and excretion of feces P and N for growing-finishing pigs. There were 3 treatments with control group,experiment groupⅠ, experiment groupⅡ, and normal diet was fed to control group,low-P diet adding phytaseⅠwas fed to experiment groupⅠ, low-P with adding phytaseⅡwas fed to experiment groupⅡ. Each treatment had 3 replicates with 6 pigs per pen. The addition of phytase in low-P diet improved ADG,reduced F/G,what is more,supplement Escherichia coli phytase to diet had significantly improved the growth performance of pigs. Adding phytase in low-P diet improved the apparent digestibility of DM,CP,CF,EE,Ca,P,GE respectively (P<0.05). The excretion of feces P and feces N was decreased by fed low-P with adding Phytase diet. The concentration of total protein,globulin,P were improved by adding phytase(P<0.05),the concentration of BUN,serum Ca was decreased by fed diets with adding phytase (P<0.05). The enzymatic activity of alkaline phosphatase in serum was not affected and the concentration of serum sugar was decreased by adding Aspergillus niger phytase; the enzymatic activity of alkaline phosphatase in serum was decreased (P<0.05) by adding Escherichia coli phytase.

Abstract:Bacillus species are one of important sources of probiotics preparation. The species exert beneficial effect on the health of animals gastrointestinal tract and the microecology balance. Many research interests are focused on the Bacillus probiotics for their advantage of alternative to antibiotics in feed.Spores of Bacillus were found that they could germinate and proliferate in vivo, and that their physiological activities, such as immunological enhancement, modulation of intestinal microflora and enzyme activities, could be released through spore germination. According to the newest research, the progress of the application and mechanism on Bacillus probiotics is reviewed in the paper.

Abstract: The experiments were conducted to study the effects of dietary energy (DE) and crude protein(CP) and calcium (Ca) and phosphorus(P) on growth performance and serum biochemical indices of the four nutrients in 70 to 90 kg newly imported Duroc from USA. The orthogonal 4×3 factorial trial was used and 9 kind of diets were designed to determine the requirements of dietary metabolizable energy and crude protein and calcium and phosphorus of newly imported Duroc from USA. The results showed that, ①the energy, crude protein, calcium level cannot significantly(P>0.05) effect in the growth performance, the phosphorus level significant(P<0.05) effected in daily feed intake;②along the increase of the dietary energy and crude protein level, the metabolic of energy and crude protein increased, but has no significant effect(P>0.05), along the increase of the dietary calcium and phosphorus level, the blood calcium and phosphorus increased,the phosphorus level extremely significant(P<0.01) effected in serum phosphorus. The best nutritive level in Duroc diet can be: digestive energy 14.2 MJ/kg, crude protein 18.6%, calcium 1.25%, phosphorus 0.73%, respectively.

Abstract:18 Chinese Holstein cows of 2 to 4 parity at mid-lactation were selected,randomly divided into 3 groups of six cows, group 1 and group 2 were added to each of 5% and 15% proportion of glutamate thalli protein, the control group does not add glutamate thalli protein in order to assess the influence on production performance of dairy cows. The results showed that 5% or 15% glutamate thalli protein was used in place of partial protein feedstuffs of dairy cows in summere,roughage intake was promote increased, and milk production was decreased slowly, but milk quality was not influenced significantly. 

Abstract: Study on the effects of Cu(Ⅱ)-exchanged montmorillonite nanoparticles on growth performance and some blood biochemical indexes of pigs.A total of 72 weaning pigs (Duroc×Landrace×Yorkshire) were used to investigate the effects of Cu(Ⅱ)-exchanged montmorillonite nanoparticles(MMT-Cu) on growth performance and some blood biochemical indexes of these pigs and verify MMT-Cu can promote growth performance more than antibiotics.The weaning pigs were allocated to three dietary treatments in a randomized complete block design.The dietary treatments were basal diet,basal diet + 20 mg/kg tylosin+100 mg/kg SM2 and basal diet + 1500 mg/kg MMT-Cu.The results showed that the addition of MMT-Cu to the diet decreased extremely significantly(P<0.01)the F/G during the whole test; the concentration of ALB,SUN,Ch,Tp in surum decreased extremely significantly (P<0.01); the concentration of Ca,GPT in serum increased extremely significantly (P<0.01) and TG,GLU decreased(P<0.05). These pigs feed the basal diet + 1500 mg/kg MMT-Cu increased the ratio of F/G,and there were various effects on ALB,Ca,GPT,SUN,TG,Ch,TP,GLU.

Abstract:In this paper, the author analyzes the mode of action of feeding probiotics on the intestinal immune function in piglets and discusses the mechanism of probiotics that promote the intestinal immune. When the probiotics into the intestinal tract, in the first, adheres to intestinal mucosa, then, stimulates to the T-cell of intestinal mucosa differentiation, produces immune-related cytokines and active substances, and promotes intestinal immune and immune synergy, so improves immunity, reduces the rate of piglet diarrhea, and increases the level of health and the performance of growth.

Abstract: To ascertain the nature of gelatinase and its role in the pathogenesis of R. anatipestifer, the enzyme from R. anatipestifer strain AF, was purified and its characterizations were analyzed, including stability for heat, effects of proteases inhibitors and metal ions on its activities and pathogenicity for ducklings. The results revealed that gelatinase was purified from the culture supernatant of R. anatipestifer strain AF by ultrafiltration, 70% ammonium sulfate precipitation, anion exchange chromatography, hydrophobic interaction chromatography and gel permeation chromatography. The sample migrated as a single band on SDS- polyacrylamide gel electrophoresis with a molecular mass of 59000. Its activity was inhibited by EDTA, Cu2+, Cd2+, Mn2+, Hg2+, PMSF, NBS, AAD, and by heating at 65 ℃ for 6 min. Purified gelatinase was proved to be a direct virulent factor on the basis of eliciting lethal toxicity to duckling by intravenous inoculation. The gelatinase not only enhanced the invasion of R. anatipestifer, increased bacteremia, shortened stage of latency and course of disease, but also raised mortality obviously. The results indicated that the gelatinase is metalloprotease, and it is virulence important role in pathogenesis of R. anatipestifer.

Abstract:Bacteriocin has potential for use as antibiotic substitute and novel green veterinary drugs.However,there was little bacteriocin which has been obtained through growth of natural producer strain and it was also difficult to purify them.With the purpose of achieving increasing yield, heterogenous expression of bacteriocin has been widely studied in recent years.This review focuses on the the development of heterologous expression systems and stratages for bacteriocin production.

Abstract: Experiment was designed to culture pulmonary artery smooth muscle cells of broiler with the enzymaic digestion method.Pulmonary artery media layer from 1 to 7 day old broilers was cut into less than 1 mm×1 mm pieces, and was digested for pulmonary artery smooth muscle cells by 0.1% collagenase type Ⅱ. The obtained cells were then cultured and subcultured. Identifications of the cells were performed with the morphological feature and immunohistochemical stain using monoclonal antibody against smooth muscle αaction.The results showed that live cells and HE stained cells were typical spindleshaped and overlapping layer form of vascular smooth muscle cells under phase contract and light microscope. αactin positive stained cells were more than 97%.Therefore,the results showed that 0.1% collagenase type Ⅱcould digest and isolate broiler PASMC. This method for culture of PASMC has advantages of easy performance, more living cells and short period for passaging.This method can be used for study in cadiovascular disease such as pulmonary hypentension and pulmonary vascular structural remodelling.

Abstract: In order to screen indicators for earlier diagnosis of diary cow mastitis by investigating the activity dynamic changes of enzymes in serum of rabbits at before and after infusion Staphylococcus aureus through teat canal. Eighteen healthy pregnant rabbits at seven days after parturition were infused with Staphylococcus aureus via teat duct to the fourth pairs of the mammary gland to establish animal model of mastitis. At exactly 2 h before inoculation and 6, 12, 24, 48, 72 h after infusion, taking the blood to determine the activity of alkaline phosphatase (ALP), lactatedehydrogenase (LDH), lactoperoxidase (LP) and myeloperoxidase (MPO). At different time after infusion, one rabbit was euthanatized and the mammary glands were removed and fixed for histopathologic evaluations. The results showed that the activity of ALP, LDH and MPO significantly increased within 6-48 h (P<0.05), and activity of LP significantly increased (P<0.05) within 12-48 h after infusion Staphylococcus aureus via teat canal. These four enzymes activities reached peak value at 12 h after infusion. The fitting degree (R2=0.9074) between LDH activity with different time at before and after infusion Staphylococcus aureus higher than other three enzymes. Mammary tissue damage was closely related to the LDH activity. The results suggested that LDH could be as a candidate indicator for earlier mastitis diagnosis to further study.

Abstract: This experiment was carried out to study effects of follicle size on collecting effect and developmental competence of goats oocytes derived from abattoir ovaries following maturation, fertilization and culture in vitro. In this experiment, the aspiration method was applied to collect the follicular oocytes. The results showed that follicle size directly affected collecting effect of oocytes and development of oocytes after IVMFC. The rate of collecting oocytes, the avaible oocyte and maturation was interrelated with the diameter of follicular positively. Both fertilization and cleavage rates of oocytes from 2.1 to 5 mm follicles were significantly higher than from >5 mm follicles and ≤2 mm follicles, which were suitable for embryo in vitro production. The oocytes from ≤2 mm follicles had the lowest developmental competence in vitro. Both fertilization and cleavage rates of oocytes from >5 mm follicles decreased ascribed to oocytes degeneration. We could improve the developmental competence of oocytes from >5 mm follicles by decreasing the cultural time in vitro.

Abstract: Establishing a method of PMCA for 22L strain PrPSc based on the method called protein misfolding cyclic amplification (PMCA) established by Saborio and Soto to research the biochemical mechanism of PrPSc formation. Under the existence of the anion detergent lauryl sodium sulfate (SDS), the method can directly degrade the products that producted by repeated amplification, and can in the brain tissue homogenate of hamster effectively imitate the replication mode that PrPSc run in vivo in a accelerated way. Therefore, it can amplify PrPScto high concentrations in a few hours. We found that in vitro unbolted brain tissue homogenate is more efficiently than purified reconstituted about PrPSc conversion, this discovery implied that highly efficient PrPSc conversion may associated with other unknown host factor. Researching indicated that the improved PMCA technique that we established can be used to research the biochemical mechanism of PrPSc formation.

Abstract: To determine the biological characteristics and immunogenicity of the molecular and virulence deleted vaccine strain YZ-2 of Brucellosis. The biological characteristics of Brucella abortus YZ-2 was evaluated by dyeing, toxicity, generation stability; vaccination of mice with YZ-2, the humoral immunity and cellular immunity were detected, and the results were analyzed. The research found that the biochemistry characteristics of YZ-2 and S19 were nearly identical, and the colonies and the suspension have disparity. The variation experiment proved that YZ-2 had become rough type from smooth type. Virulent experiment suggested that strain YZ-2 had lost some residual virulence of the parental strain, also YZ-2 has good heredity stability. Research verified strain YZ-2 has the immunogenicity as well as the parent strain. The results suggested that the safety of YZ-2 is better than parent strain, and the stability is well, has the same immunogenicity as parent strain; YZ-2 has good prospects to be used.

Abstract:According to the gene sequences in GenBank of porcine circovirus type 2 (PCV2), porcine pseudorabies virus (PRV) and porcine parvovirus (PPV), three pairs of specific primers were designed for amplifying the four specific fragments of PCV2, PPV and PRV 350 bp 270 bp and 191 bp in length, respectively. The results indicated that multiplex PCR is both sensitive and specific, providing a new method for the clinical diagnosis and epidemiological research of the three diseases.

Abstract: The hydrophilicity, antigenic index and surface probability of glycoprotein B (gB) of infectious bovine rhinotracheitis virus (IBRV) were analyzed by biosoftware DNAStar-Protean. Amplified the sequence which codes the amino acid residues from 385th to 550th in major antigen determinant region of gB glycoprotein. Then the purified products were cloned into pMD18-T vector and sequenced. The truncated gB gene was inserted into pET-28b vector which was transformed to competent Escherichia coli DE3 for expressing. The SDS-PAGE showed that the fusion protein of 21.35 ku as expected. Western blotting and indirect ELISA analysis that the recombinant protein had a specific positive reaction with standard positive blood serum. The concentration of purified protein was 2.0 mg/mL and the purity was 95.27%. The preliminary indirect ELISA method was developed by using the purified recombinant protein of gB as antigen. The results above suggested that the assay was confirmed to be specific, sensitive and quickly, which can be used as antigen for diagnosis.

Abstract: According to the E2 genes of bovine viral diarrhea virus (BVDV) which were obtained from GenBank, the target fragment of about 640 bp of isolate Yak was amplified by reverse transcription polymerase chain reaction (RT- PCR), cloned into pMD18- T vector and sequenced. The positive recombinant clone was identified by restriction enzyme digestion. Sequence comparison and phylogenetic analyses showed that isolate Yak fall into the BVDV genotype 1 and were distant from other subgroups. There is no insertion or variation of E2gene. It had the highest sequence identity (68.8%) with the Australia Braidwood strain. The low sequence identity with other BVDV- 1 strains indicate that strain Yak had an independent evolutional pathway.

Abstract:Through RTPCR amplification of tissue or blood sample from some of pig form of Huanan area, 14 sequences of the ORF5 gene and 13 sequences of the partail Nsp2 gene were obtained. Sequencing and comparative analysis showed that two sequences exhibited deletion in the Nsp2 gene. There was 96.9% to 99.1% identity between those sequences of Nsp2 gene, sharing 85.1% to 86.4%, 67.2% to 68.9%, 54.1% to 55.9%, 67.6% to 68.9% identity with CH1a, VR2332 LV and MLV respectively. The identity among those sequences of ORF5 gene was 98.8% to 99.7%, sharing 94.4% to 95.2%, 86.7% to 87.9%, 54.1% to 55.7%, 86.4% to 87.6% identity with CH1a, VR2332, LV and MLV respectively. No deletion was exhibited in the 14 ORF5 gene sequences, and the sequences of ORF5 genes were analysed and compared with those of ATCC VR2332,CH1a, LV and MLV strains, there was 98.8% to 99.7% identity among those sequences of ORF5 gene, sharing 94.4% to 95.2%, 86.7% to 87.9%, 86.4% to 87.6% and 54.1% to 55.7% identity with CH1a, ATCC VR2332, MLV and LV respectively. And the homology comparision between the deduced amino acid sequence and other strains were 91.5% to 93.5%, 86.6% to 88.6%, 85.6% to 87.6%, 54.7% to 56.2% respectively. The results showed that the prevalent PRRSV strains in China belonged to North American in genotype, and the result showed that those virus probable origin of one strain, and prevail throught the transportation of the pigs.

Abstract:Melanocortin receptor-4(MC4R)gene in mink was cloned and sequenced,in order to provide theoretical foundation for further research of the corelevantion of MC4R gene in mink and fat traits,and location. MC4R gene of mink was amplified using particular primers,cloned by polymerase chain reaction (PCR) method and sequenced,and spliced by using DNAMAN. The length of MC4R gene in mink was 816 bp. The sequences had been sent to GenBank and registered. Comparison of the fragment sequence with that of other mammal animals MC4R gene was carried out through BLAST in GenBank,the results showed that the homologies of the nucleotide sequence with river otter,dog,fox,panda,river horse,boar,goat,human and rat were 89% to 95%,MC4R gene of mink was successfully cloned in this study,indicating it was quite conservative among species.

Abstract: Exon 2 of prolactin gene were detected by PCR-SSCP and the DNA fragments were sequenced.The correlation analysis between the polymorphism of the exon 2 and egg-laying and broodiness traits were carried out respectively.The results indicated that there were two polymorphic sites in exon 2.A C/T mutation at the position of the 3838 bp was found,which resulted in 3 genotypes(AA,AB and BB).Furthermore,an amino acid changed from Leucine to Phenylalanine. The frequencies of genotype AA and allele A were the highest.The least square analysis showed that the first broodiness day of BB type chickens was later than AA and AB type chickens (P<0.01), the times of broody of BB chickens were more than AA and AB type(P<0.05). But there was no significant difference in egg production among the 3 genotypes.

Abstract:In this study,the dog semen was frozen with different cryopreservation,including glycerol and glycol,for better cryopreservation result. It was conducted to compare different concentration of glycerol or ethylene glycol’s effects on frozen sperm’s recovery rates and compare different balance time. The glycerol as cryoprotectant is better than ethylene glycol and the concentration of glycerol is 4%,the average recovery rate of frozen semen is highly,60 min of the balance time was more helpful to the survival of frozen sperm. The study found that cryopreservation at 4% of glycerol,balance 60 min at 4 ℃ achieved better sperm recovery for canine semen cryopreservation,with an average of 39.21%±0.013%recovery rate of frozen semen,and the results were rather stable. 

Abstract:To research the cucumber and lettuce’s combination to promote the role of fracture healing and the initial mechanism, 60 white broiler, weighing 1.75- 2.25 kg,were randomly divided into an equal number of high- dose group, middle dose group, low- dose group, positive control group and negative control group, then, to fracture the broiler’s middle of right leg to cause transverse closed fracture model. Different proportions of cucumber and lettuce seed powder extract were gavage feeding to experimental group. Musk bone capsule was gavage feeding to positive control group. Negative control group were fed with saline. Subsequently each group were adopt dynamic observation, histological observation, serological and radiological observation. Experimental group that were fed middle dose of cucumber and lettuce seed powder extract could promote systemic metabolic response, increase serum alkaline phosphates' levels, speed up the absorption of hematoma, increased serum calcium and phosphorus concentration and improve the quantity and intensity of callus generation. The results showed that cucumber and lettuce can increase calcium and phosphorus concentration, promote calcium and phosphorus transfer to the fracture line to promote fracture healing.

Abstract:The residues depletion of Enrofloxacin(ENR) and Ciprofloxacin(CIP) in the pig tissues was studied by using HPLC method. Enrofloxacin microcapsules was mixed-fed on consecutive seven days. After the last fed,five pigs were slaughtered and collected tissue samples. Tissue samples were extracted by phosphate buffer solution, purified through C18SPE. Then the extract was dissolved with a mobile phase: 0.05 mol/L phosphoric acid/triethylamine-acetonitrile (82 +18).Finally, the samples were detected by HPLC after filtration. Results showed that, when the sample concentrations were 0.02 μg/g to 1 μg/g, the linear relationship was good. The detection limit was 20 μg/kg.The recovery was between 72% to 90%,and the coefficient of variation was under 10%.The depletion of enrofloxacin in muscle and fat was fast, and the sum of concentration of ENR and CIP was declined, lower than LOD on the 5th day. However, compared with fat and muscle, the depletion of drugs in liver and kidney were slower. The sum of concentration of these in kidney was 91.48 μg/kg at the 7th day. Integrated total residues in tissues, we can identify Enrofloxacin microcapsules withdrawal time is 7 days.

Abstract:Infection of chicken with protozoa Eimeria spp. is quite common with a global geographical distribution, which can cause serious clinical diseases (ie, Coccidiosis) in the host. Chicken infected with Eimeria spp. remains a severe problem for chicken health. As a major way in controlling the coccidiosis in chickens, the use of anticoccidal drugs has caused the development of drug resistance of coccidia and makes it hard to prevent coccidiosis in chickens. Meanwhile, drugs residues cause serious consequences to the environment and human health. Study on coccidiosis vaccines for chicken has become one highlight for many veterinarians. This paper summarized the immunologic mechanism and research progress on chicken embryo attenuated method, physical and chemical method and precocious strain selection method of coccidiosis attenuated vaccine for chicken. The application prospect of coccidiosis attenuated vaccine for coccidiosis control was also illustrated.

Abstract: To investigate the clinical veterinary effect of porcine recombinant interferon (IFN)-alpha in promoting growth and preventing viral disease, the test was processed with total 40 piglets in Guangdong province. As result, the feed to gain ratio of group 1 and 2 had decreased to 6.3% and 17.23% respectively, the average daily weight gain had improved-3.69% and 24.85%, the survival rate had increased to 6.6% and 14.3% separately, the rate of diarrhea had decreased to 11.5% and 26.9%, the total controllable ratio of the diseases had increased to 40.1% and 55.49% separately, comparing with group Ctr. During the observation period, the 4 piglets of group Ctr were infected by PRRSV and 1 of them was death, none was infected in group 1 and 2. In conclusion, the result has showed a good promoting growth′s performance and the notability to prevent viral disease of the piglets.

Abstract: The objective was to study effect of different antibiotic combinations on growth of rumen bacteria with a Hungate’s roll tube technique to isolate anaerobic fungi from rumen fluids of dairy cows. Penicillin, Streptomycin, Chloramphenicol and Metronidazole in different levels were added to 9.0 mL solid or liquid mediums prior to the anaerobic inoculation. Based on counting results of bacteria and fungi up to three day, the combination of 0.8 mg/mL Penicillin and 1.2 mg/mL Streptomycin did not inhibit the growth of rumen bacteria in mediums. However, the combination consisted of 0.8 to 1.2 mg/mL Penicillin, 1.2 to 1.8 mg/mL Streptomycin and 0.004 to 0.08 mg/mL Metronidazole significantly inhibited the growth of rumen bacteria without negative impact on that of anaerobic fungi (P<0.05). Similar inhibition also occurred for the combination consisted of 0.8 to 1.2 mg/mL Penicillin, 0.6 to 1.2 mg/mL Streptomycin, 0.1 to 0.3 mg/mL Chloramphenicol and 0.004 to 0.008 mg/mL Metronidazole. In general, antibiotic resistance of rumen bacteria should be paid more attention, and the combination of different antibiotics in this study might provide an alternative reference for the effective control of bacteria growth in the application of a roll tube technique to isolate anaerobic fungi from the rumen fluids. 

Abstract:In order to scientifically evaluate the drug sensitivity of pathogenic Escherichia coli strains isolated from piglit’s yellow dysentery and the cure effects of different drug administrating ways, the pathogenic Escherichia coli(E. coli) strains were first isolated from piglets sickened yellow dysentery in 25 large-scale pig farms from Henan province. The virulence tests were carried out by analyzing pathogenicity of the isolated E. coli strains to mouse. Drug sensitive tests of the isolated bacteria strains on 20 kinds of antibiotics were carried out to estimate the most sensitive antibiotics drugs. Three different ways of drug administrating which including oral administration, intramuscular injection and intraperitoneal injection were tested to evaluate the best drug administration way. The results demonstrated that total 25 strains of E. coli were isolated from 25 sickened pig farms. All the isolated E. coli strains displayed the pathogenicity to mouse with 100% morbidity rate. Drug resistance appeared to each antibacterial drug, but the drug resistance rate was different (from 4% to 100%). Among the 20 antibacterial drugs, Norfloxacin,Floxacin,Neomycin and Rifampicin showed higher drug sensitivity to 25 E. coli isolated strains than the others, which displayed the high sensitivity rates of 92%,88%,88% and 80%, respectively .Three different ways of drug administration exhibited the different therapeutic effects on clinic sickened piglets. The drug administration ways of oral administration and intraperitoneal injection demonstrated higher cure rates (92.3% and 95.0% respectively) than that by drug administrating of intramuscular injection (58.8%). The oral administration showed slightly higher cure rate than the intraperitoneal injection, which was no significantly statistical difference between them.

Abstract:The erythrocyte agglutination assay for detection of the antibody to gE protein of pseudorabies virus in porcine was established by the anti- human red blood cell single chain fragment variable- pseudorabies virus gE bifunctional fusion protein as antigen. The optimal work concentration of antigen was 55 μg/mL. The dilution of sera was 1∶20. The reaction time of sera was 15 min. It revealed a negation reaction with positive sera of classical swine fever virus (CSFV), porcine parvovirus virus (PPV), porcine reproduction and respiratory syndrome virus (PRRS), Japanese encephalitis virus (JEV), Brucella and serum of swine inoculated by PRV gE deleted vaccines. It revealed a positive reaction with PRV standard positive serum. The total coincidence rate of the negative and positive reaction reached 100%. The results showed that the established erythrocyte agglutination assay has the advantages such as high sensibility and strong specificity and can be used to detect the porcine serum antibodies to gE protein of PRV.

Abstract:The minimum inhibitory concentrations (MIC) of Herba Taraxaci, trimethoprim and Herba Taraxaci combined with Trimethoprim against Staphylococcus aureus, Escherichia coli and Salmonella were measured individually by the method of microdilution checkerboard techniques. Then the fractional inhibitory concentrations (FIC) indices of Herba Taraxaci combined with Trimethoprim were calculated. The results showed that the FIC values on Staphylococcus aureus, Escherichia coli and Salmonella were respectively 0.75, 0.75,1. The effect of their combination taken on additive action. The number of Staphylococcus aureus, Escherichia coli and Salmonella which had been inhabited by Herba Taraxaci combined with Trimethoprim for 1 h, 2 h, 4 h and 8 h, were enumerated by plate counts methods. Then the killing rate of Herba Taraxaci combined with Trimethoprim were calculated. The experimental data were fitted by least squares method. The optimal dose of Trimethoprim which combined with Herba Taraxaci against Staphylococcus aureus, Escherichia coli and Salmonella in vitro, were respectively 4.0 mg/g,1.8 mg/g and 2.0 mg/g. 

Abstract:Faecal samples of puppies after artificial trophozoite infection were examined for Giardia cysts by zinc sulphate floatation. The results indicated that trophozoite could infect puppies and the shedding of Giardia canis cysts in puppies was intermittent. The period of intermittence was 7 to 8 days. Giardia cysts were detected after 3 days,cysts shedding peaked in 13 days after inoculation ,and lasting for 3 to 4 days. In the intermittent period,we also detected a little amount Giardia cysts everyday. The infected puppies had the symptoms of diarrhea and depressed and weight loss.

Abstract:Diseased chicken collected in the Zhumadian district chicken farms were examined. The investigation showed that pathogenic E. coli was main reasons that caused chicken most of death. Drug- sensitivity test to 16 drugs was performed with 10 isolated pathogenic E. coli strains. The results suggested that most of E. coli strains were highly susceptible to Lincomycin- Spectinomycin, Ceftriaxone, Amikacin, Nemomycin, Ofloxacin. The frequency of drug- sensitivity was 80%, 60%, 60%, 60%, 50% respectively. Drug- sensitivity to Colistin and Florfenicol were 10% and 20%. All isolated pathogenic E. coli strains showed resistence to Thiamphenical, Doxycycline, Enrofloxacin, Norfloxacin, Ciprofloxacin, Streptomycin, Amoxicillin, Ampicillin and Oxytetracycline.

Abstract:Based on a series of 16S rRNA sequences of Haemophilus parasuis (Hps) published in GenBank, a conservative region was found. Four specific primers targeting the 6 distinct sequences of the conservative region were designed. A loop-mediated isothermal amplification (LAMP) for the rapid detection of Hps was established by using those specific primers. The results show that LAMP has a high specificity, and is more sensitive than PCR method under 63 ℃. Compared with the complex isolation and identification methods of Hps, LAMP is simple, low cost, and could be completed in 1 h. 

Abstract:A covey of swine whose clinical symptoms looked like Haemophilus parasuis disease were checked by lab technologies. Research as bacteria observation, cultural characteristics and biochemical properties of Haemophilus parasuis were done. According to the sequence of bacterium 16S rRNA, special primers were designed for PCR amplification. The results showed that the strain was tentatively identified as Haemophilus parasuis. The drug sensitive tests showed:the strain was sensitive to Ciprolfloxacin, Kanamycin, Doxycycline, Ampicillin, Amikacin, Gentamicin; but resistant to Tiamulin, Norfloxacin.
Preparation of Clinical Mastitis Induced by Staphylococcus aureus in Rabbits

Abstract:To establish rabbits mastitis model, 104 CFU/mL/3 to 4 kg (body weight) of Staphylococcus aureus that was isolated from the milk of dairy cattle with mastitis were infused into the third of mammary gland in 16 healthy rabbits. When the clinical mastitis model was challenged, observe its manifestations, pathological changes in breast tissue morphology, and regress the breast tissue fluid after established model 72 hours. The results showed that: ①Post- challenged treated group expressed significant clinical signs, manifested as reduced dietary wishes, and even waste must; body temperature, heart rate and respiratory rate significantly increased; model swollen breast areas.②Challenged district of breast tissue examination found inflammatory cell infiltration, destruction of the organizational structure and so on. ③Regression tests were positive by both morphological and biochemical identification was identified as Staphylococcus aureus. The results demonstrated that the rabbits experimental mastitis model infused with Staphylococcus aureus via teat duct was the same as immunopathology and inflammatory response, and can be used as a pathological mode1.
