Genetic diversity of exon 3 in insulin-like growth factor-Ⅰ(IGF-Ⅰ) gene was studied using PCR-SSCP technique in seven, eight and nine generations of Jinghai yellow chicken population in the present study, and calculated the genotype, gene frequency, chi-square value and some indicators of genetic polymorphism. The results showed that alleles increased with decreasing the number of generations; genotype distribution between the three generations achieved a significant(P<0.05) or very significant(P<0.01) difference; heterozygosis increased with the number of generations increased; the locus polymorphism of three generations were moderate. Indeed, the initial inference elected to stay on the IGF-Ⅰ gene had an impact on 31 points, the loci could be applied in breeding practice.
