This experiment used four primiparous Holstein cows infused with free linolenic acid (0, 100, 200, 300, 400 g/d) into the duodenum of dairy cows in cross-over design, the author investigated the effect of increasing amounts of free linolenic acid on the content of fatty acids and oxidation stability in milk fat. The results observed that the content of polyunsaturated fatty acids, especially α-linolenic acid increased significantly in milk fat, but the total activity of superoxide dismutase and total antioxidant activity were all have the tendency of decreasing, however, the thiobarbituric acid reactive substances have the opposite tendency. The results showed that fatty acid profile and the oxidation stability in milk fat would be influenced by increasing amounts of free linolenic acid infused into the duodenum of dairy cows.

To study the relationship between fluorosis and hepatocyte apoptosis and morphological changes in meat ducks, 160 8-day-old Tian-fu meat ducks were chosen and randomly divided into four groups,and 0, 350, 450 and 550 mg of sodium fluoride were added to conventional dietary per kg respectively. After challenge five ducks were killed in each group every eight day and TdT-mediated dUTP nick end-labeling (TUNEL) method was used for experimental fluorosis liver tissue handling and labeling. Percentage of hepatocyte apoptosis and morphological changes were detected on the 24th,40th and 56th day with the light microscope. TUNEL results showed that DAB staining positive cells appeared typical morphological characteristics and the percentage of apoptosis cell increased obviously compared with the control group. Fluorosis in meat ducks mainly caused early and middle hepatocyte apoptosis.With time extension and dose increase,sodium fluoride accelerated apoptosis of hepatocyte and developed into necrosis, indicating that toxic effects of excessive fluoride in vivo is closely related to hepatocyte apoptosis.

Methanogen is a kind of strictly anaerobic archaea that can transform organic matter and inorganic compounds into methane and carbon dioxide in an anaerobic fermentation. Methanogenesis usually occurs in the last step of anaerobic fermentation process. Being the key part of the nature carbon cycle,end-product of methane has been ranked as a second key greenhouse gas besides carbon dioxide which exacerbate global warming trend. In order to obtain a further understanding to the biological characteristics of different kinds of methanogens,researches on laboratorial techniques of isolating and purifying methanogens are also increasingly focused in last decades. This paper thoroughly reviewed research advances on typical habitat and biological role of methanogens,species classification and their physiological characteristics with focuses on techniques of strains isolation and purification,and the selective action of various mediums for methanogens.

For developing the typical testing standard of transgenic mammals and its derived food and the technical support for entry﹠exit animal quarantine, detection of the transgenic component in transgenic cow by multiplex PCR was developed in this study.The endogenous mitochondrial DNA(mtDNA) gene specific for cow identification, exotic human lysozyme(hLY),marker NPTⅡ were amplified according to the differential primers. The process of the multiplex PCR reaction was optimized. The system described herein represent simple, accurate and sensitive detection methods in which only one reaction was necessary to detect multiple target sequences that could be reliably used for the identification of specific lines of transgenic cows.

In order to investigate the mRNA expression of compact-related genes in buffalo in vitro matured and fertilized embryos, the Taqman probe and SYBR-Green Ⅰ quantitative real-time PCR(Q-PCR) method was used to analyze mRNA expression of Cx43, Cx31 and E-cad genes in pooled baffalo in vitro cultured embryos. Cx43 gene showed significantly higher expression in matured oocytes than in other stage embryos(P<0.05), reverse to Cx43 gene, Cx31 gene had the lowest expression in matured oocytes compared with other three stages, and gradually increased its expression along with the development of embryos.The level of E-cad mRNA expression did not vary among the tested embryos(P>0.05). The present study demonstrated that the three compact-related genes had expression in all stages of baffalo IVF embryos, but showed definitely different mRNA expression style.

The purpose of the present study was to examine the effect of different sequential cultures on the porcine parthenogenetic embryos of development after activation. The ordinary culture and sequential culture were used in vitro culture of procine oocytes. The results showed that NCSU-23 supplemented 0.4% BSA was a better culture media for the oocytes development after activated in the early stage, but mTCM-199 supplemented 0.1% PVA facilitates parthenogenetic embryos breakthrough 4-cell block and reach morula;the sequential cultures system of NCSU-23 supplemented 0.4% BSA and mTCM-199 supplemented 0.1 % PVA could improve the development of porcine parthenogenetic embryos.

Taking Tan sheep ear margin as the study material, we established successfully a fibroblast cell line by the method of explant culture directly. Observations on morphology, dynamic growth, determination of viability, analysis of karyotype, test of microorganism, transfecting fibroblasts with fluorescent protein vectors mediated(PEGFP-N3) by lipfectamine and another characteristics were detected. The results showed that population doubling time (PDT) of cells was approximately 36 h, cell viability was 95.8% after thawing, the status of cell After passage was constant, analysis of chromosomal karyotype indicated that diploid (2n=54) account for 97.5% in the cell line. Every index in the cell line met all the standard quality controls of ATCC。

In this study, the OmpL1 was amplified by PCR from a serovar Leptospire canicola interrogans, and it was cloned into plasmid pVAX1,then sequence analysis was done. These all got an expectation about the 960 bp of special tape, it’s sequence analysis and gene comparision with Leptospire hebdomadis,Leptospire autumnalis,Leptospire pomona and Leptospire australis, the homology of nucleotide sequences was 99.38%,93.77%,99.58% and 99.38%, the homology of deduced amino acid was 100%,95%,100% and 100%. It indicated that OmpL1 was a conserved antigen among the pathogenic Leptospira species, fusion protein OmpL1 might play an important role in diagnosis and prevention of Loptosirosis.

FSHα-subunit gene segments were amplified by PCR from the original constructed vector. Then the PCR product was TA cloned, sequenced and inserted into the carrier pGEX-6P-2. The recombinant plasmids were identified by restricted enzymes digestion, and then the positive clones were transformed into E.coli BL21. GST-FSHα fusion proteins were expressed via the induction of IPTG, and detected by SDS-PAGE. The results showed that PCR product was about 380 bp,and sequencing result was identical with GenBank. The pGEX-6P-2-FSHα positive clone produced a 39.3 ku fusion protein on SDS-PAGE gel. Successfully cloning and expressing Cervus nippon FSHα-subunit gene.

To study genetic variation of mitochondrial cytochrome c oxidase subunit Ⅲ (cox3) gene of Eimeria acervulina, the cox3 was amplified by polymerase chain reaction (PCR) and 572 bp of partial cox3 (pcox3) was obtained from five strains of E. acervulina. By sequence comparison and comparative analysis with the cox3 gene of some protozoa of Phylum Apicomplexa avalidable in GenBank, the results showed that there was no difference in the nucleotides of the pcox3 sequences obtained from five strains of E. acervulina, and there were distinct differences in the nucleotides compared with other protozoa of Phylum Apicomplexa. The sequence diferences were 51% and 50% compared with Plasmodium and Leucocytozoon respectively, and was less than 50% compared with Theileria. It was the first time that cox3 of E. acervulina was reported. The results of the present study provided a foundation for further studies of population genetics of Eimeria.

Wenyonella philiplevinei is one of important pathogens of duck coccidiasis. To seek specific genetic marker of the protozoan, the fresh stool was obtained from a duck house in Guangdong, then coccidial oocysts were collected by Sheather’s sucrose floatation procedure, and these oocysts were identified as W. philiplevinei according to morphology. The partial 18S rDNA was amplified by PCR, cloned and sequenced after the genomic DNA was extracted. The genetic relationship between the isolate and other protozoan was compared. The results showed that the sequence was closer to other coccidians of Eimeriidae. Phylogenetic analysis indicated that duck-derived W. philiplevinei isolate belonged to another branch of Eimeriidae. It is concluded that the 18S rDNA sequence provided an useful genetic marker for the differentiation of W. philiplevinei from other related protozoa.

The reverse transcription loop-mediated isothermal amplification (RT-LAMP) had been built as a rapid and exact detection method which could play an important role in the prevention and control of Newcastle disease. Two pairs of specific primers were designed based on the sequences of SCFV E gene available in GenBank, targeting the 6 distinct sequences of E gene. A reverse transcriptase loop-mediated isothermal amplification(RT-LAMP) for the rapid detection of CSFV was developed by using the specific primers. The results showed that the detection limit of the CSFV-RT-LAMP method was 10-5. The amplification could be finished within 1 h with good specificity and high sensitivity. And the developed method can meet the demands of field detection of CSFV infection.

Tissue samples were taken from the dead geese of breeding goose farm suspected to be colibacillosis infection,8 pathogenic E.coli strains were isolated and identified,the results of identification using O and K serogroups showed that 8 isolates were confirmed to be O88K61(4 strains) and O44K74(3 strains) serotype respectively, except 1 unidentified strain,endotoxin was not detected in the identified E.coli strains above. The antibiotics sensitivity test with 14 kinds of antibacterial drugs indicated the 8 isolates were highly sensitive to Amikacin,moderately sensitive to Vancomycin,Neomycin and Furazolidone,and resistant to Amoxinllin,Streptomycin,Kanamycin,Gentamicin and Tetracycline,the 8 E.coli isolates were cross-resistant to Enrofloxacin,Pioneermycin,Sulfadiazine,Norfloxacin and Doxycycline. The results above also revealed that at least 3 pathogenic E.coli serotypes with multi-drug resistance existed among the sick geese flock in the same farm.

Three swine influenza virus strains were isolated from 9-11-day SPF chicken eggs in our study of these, GXHZ isolation was subtyped as H1N2 by HI assay and RT-PCR, and GXLZ,GXXY as H3N2. We detected the EID50 of these isolated strains and performed inoculation to pig with GXHZ isolation. NA genes amplificated from 3 isolated strains showed that their nucleotide and amino acid sequences had highest homology to A/Sw/Hainan/1/2005 (H1N2) strain. Two substitution at 6 (K6→R6) and 20 (V20→M20) separately located on the amino acid cytoplasmic tail and nonpolar transmembrane domain of the NA molecules of isolated strains. Phylogenical analysis of the NA genes indicated that the NA genes of isolated strains were most closely related to H1N2 which derived from swine origin influenza virus. Moreover, three isolated strains were clustered with H3N2 strains isolated after 1996.

The method for detecting Lanzhou fat-tailed sheep fatty acid-binding proteins gene mRNA expression with real time PCR was developed. By isolated total RNA form the fresh omentum, kidney weeks adipose tissue, muscle and fat tissue sample using improved the extraction Trizol one step method and reverse transcribed to cDNA using random primers as primer, the results show, the standard curve were CtH-FABP =-3.24375x+38.34230 and Ct18S=-3.33137x+33.4573, had good linear dependence, and it was sensitive and specific. The coefficient of variation values for both intra-experimental and inter-experimental reproducibility ranged less than 1.8% and 10% respectively. The method for detecting Lanzhou fat-tailed sheep H-FABP gene expression with fluorescence quantitative polymerase chain reaction was developed successfully.

The indices of bovine growth traits, meat quality traits and carcass traits were studied to analyze the correlation within/between the 3 groups and reveal the interaction between traits in each groups. The phenotypic values of 17 traits from 327 steers were used to analyze the correlation in-depth study by ordinary correlation analysis and canonical correlation analysis. A total of 74 pairs of traits were highly significant ordinary correlation between 17 traits (P<0.01), and most correlation coefficients were below 0.5.The canonical correlation analysis showed that highly significant correlation coefficients of the first、second and third canonical correlation were obtained between the 3 groups(P<0.001). The first and second canonical correlation coefficients between growth traits and meat quality traits, growth traits and carcass traits, meat quality traits and carcass traits were 0.525 and 0.350,0.993 and 0.623,0.698 and 0.394, respectively. The canonical correlation analysis can more accurately reflect the correlation between traits of 3 groups than ordinary correlation analysis, and benefit for breeding and selecting of beef cattle.

In order to understand shape of lactation curve and factors which affect on the lactation curve traits of Sanhe cattle.The Wood model was used for fitting the latation curve of 486 lactation records of Sanhe cattle.The results showed that the lactation curve was well fitted by the Wood model, and the R2 was 0.93±0.004.ANOVA method was used for analysing the effection on lactation curve traits by non-genetic fectors such as farm operation, calving year, calving season and parity.The results indicated that the effects of farm operation was highly significant for c, tmax, per and Y305; calving year was significant for b and highly significant for c,ymax,Y305; calving season was significant for per and highly significant for a,b,c,Y305; parity was highly significant for a,ymax,Y305;other factors had no significant for remain lactation curve traits.

The allele, polymorphism information content, heterozygosity and mean effective number of allele were studied in 3 Xinjiang game chickens(Tulufan, Kashi and Yili) through 8 microsatellite locis. The results showed that 77 alleles were found, the polymorphism information content of all loci(in addition to sites outside the Yili cock ADL0298) exceed 0.5, and the average genetic distance of all groups was 0.2866. So the 8 microsatellite locis could be used for analysis of genetic diversity among 3 Xinjiang game chickens.

In mammals,the melatonin secreted by pineal gland was modulated according to the photoperiod and season. The hypothalamic-pituitary-gonadal axis accepted the messages from melatonin and integrated both external and internal status and appropriately altered the activity of the GnRH. All these supported mammals to maximize reproductive success and avoid breeding during inappropriate conditions. Reproductive rhythm was formed and fixed in the process of long evolution in mammals and some genes were identified to be related with reproductive seasonality. This review briefly introduced the relationships between MTNR1A,MTNR1B and KiSS-1/GPR54 genes and reproductive seasonality in mammals.

Male mammalian reproduction and development were affected by plateau ecological factors,such as illumination,temperature,high-altitude hypoxia and other factors. Wenchang,Anka and Zang chickens were used for SNPs discovery in exon 9 of Nramp1 gene using PCR-SSCP and sequencing methods. Two SNPs at C315G and C357T,and four genotypes of GG,GT,TT and AA were detected in three chicken populations. Allele T was the dominant allele in Wenchang and Zang chicken populations,while allele G was the dominant allele in Anka chicken population. All the populations were in Hardy-Weinberg equilibrium(P>0.05).There were no significant differences in three chicken populations(P>0.05).

In order to provide scientific basis for studying the origin and the genetic differentiation of Luchuan pig, we surveyed the mtDNA D-loop hypervariable regions of 17 Luchuan pigs and five haplotypes were detected. The haplotype diversity and the nucleotide diversity were 0.640 and 0.0074 respectively that suggested the genetic diversity of Luchuan pig was abundant. Analyzed with the same segments from the indigenous chinese pigs retrieved from the GenBank, we concluded that Luchuan pig was close with Ningxiang pig and Dahuabai pig which traditionally belonged to central Chinese breeds, but distant with Xiang pig and Diannan Small Ear pig which all traditionally belonged to south China breeds. It was inconsistent with the traditional pig breed grouping system.

Inactivated vaccines were prepared with three H9 subtype AIV isolated from Hebei, Henan and Shandong between 1998-2009. SPF chickens were vaccinated and 21 days later blood samples were collected for HI test. Five field isolates from Hebei, Henan, Shandong and Beijing were used to infect immunized chickens to examine the protectivity of the candidate vaccines made with above isolates. The HI test results showed that HI antibody levels against the H9 antigens triggered by the three candidate vaccines were apparently increased. Although there was certain degree of differences at the antibody titers raised with different vaccines, using homologous virus as antigen resulted in higher HI titers. The challenge experiments showed that H9 subtype AIV strains isolated at different time and places produced sound cross-protection. The study also demonstrated that the inactivated vaccine prepared with 1998 field isolate WD98 offered fairly good protection against current circulating H9 subtype AIVs.

To observe Chawu Mei, aloe and so on, 12 Chinese herbal medicines in the water extract of chicken pathogenic Escherichia coli O78 inhibition effects. Ebony, aloe, et al, boiling water to get the water extract of traditional Chinese medicine obtained solution was determined on the chicken disease Escherichia coli O78 minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC), of 12 Chinese herbal medicines for drug susceptibility testing, and better in vitro antibacterial effect of a single taste of Chinese medicine in vivo tests. Results showed that Platycodon grandiflorum, Forsythia, rhubarb, peony, Atractylodes almost no activity against chicken pathogenic Escherichia coli, ebony, aloe vera, Pulsatilla good antibacterial activity, including ebony, Pulsatilla and Aloe vera extract on chicken pathogenic Escherichia coli O78 of the MIC were 2.57, 2.06 and 2.31 mg/mL. Ebony, aloe vera, and the bulbul has a certain resistance to the role of chicken pathogenic Escherichia coli, for the development of new anti-chicken pathogenic Escherichia coli lay the theoretical basis for the drug.

Bacteria DY9 strain was isolated in Sichuan province from a dead duck. DY9 strain was investigated by biochemical reaction test, nucleotide sequence of 16S rRNA and cam gene. The results showed that DY9 strain was Riemerella anatipestifer(RA) and the 50% lethal dose (LD50) was more than 2.3×1010 CFU/0.5 mL for 14-day-old ducks. In animal assay, the dead ducks did not characterize fibrinous inflammation and there did not have RA in the livers. The RA did not recover virulence after 10 passages in ducks and not loss virulence after 30 passages in vitro culture in chocolate agar. Morphology of duck embryo fibroblast treated with culture filtrate of DY9 strain was not changed. At vaccinated 14 days, the oil-adjuvant inactivated vaccine and the live bacteria had the relative percentage survival (RPS) with 100% and 79.52% respectively, and at 21 days, they were both 100%. The results indicated that DY9 strain RA was low-virulence field strain, and the virulence was stabile.

To study the influence and mechanism of norcantharidin(NCTD) on growth of lung cancer cell line A549 and Lewis, the inhibitory rate, cell cycle arrest, mitogen activated protein kinase(MAPK) were detected by wst-1, flow cytometry and immunoblotting technique,respectively. IC50 of NCTD to A549 and Lewis was 13.5 μmol/L and 9.5 μmol/L,respectively. This study showed that the inhibition of NCTD on A549 and Lewis. G2/M phase blocking rate was 26.30%,29.44%,49.83% and 81.35% at 24 h after NCTD treated cells, dose of NCTD was 0,6.25,12.50 and 25.00 μmol/L. The results indicated that one of the major antitumor mechanisms of NCTD may be through inhibiting proliferation.

To establish a SYBR-Green I fluorescent quantitative PCR method to detect the minicircles of the kinetoplast DNA of Leishmania,special primers were designed and synthesized to this conserved sequence. The special fragment was cloned into pMD18-T vector after PCR amplification,and then was transformed into E.coli DH5α. After the positive recombinant plasmid was identified,it was used as quantitative template to generate standard curve and melt curve. Results showed that the standard curve had a good linear relationship between cycle threshold(Ct) and template concentration,and the correlation coefficient was 0.996. We have successfully established a SYBR-Green I fluorescent quantitative PCR method to detect Leishmania,which can be used in the diagnosis and epidemiological surveillance of Leishmaniasis.

To express ORF3 gene of porcine circovirus 2(PCV-2) in E.coli, analyze the antigenicity of ORF3 protein, according to the amino acid sequence of PCV-2 strain in GenBank, a pair of specific primer was designed to amplify the fragment encoding ORF3 gene. This fragment was cloned into the multiple cloning sites of pET-32a (+) vector. The recombinant plasmid was designated pET32a-ORF3 and was transformed into E.coli Rosetta-blue (DE3) pLacⅠ competent cells. Expression of recombinant protein was induced by addition of final concentration of 1.0 mmol/L IPTG under 37 ℃ condition. The length of PCR product of interest was 315 base pairs. The size of recombinant amino acid was 29 ku. The results of western blotting showed that it could be recognized by anti-His tagged mouse monoclonal antibody and could not be recognized by swine serum infected with PCV-2. The results showed that ORF3 of PCV-2 was expressed successfully in E.coli and the ORF3 protein didn’t exhibit reaction genicity. These results were benefit to research for function and characteristics of ORF3 of PCV-2.

This experiment was based on investigating the reproductive function of male animals treated with Ivermectin, in order to provide the basis for clinical safe medication and animal breeding. We chose the adult male rats as the experimental model, all of them were divided into 0.17 mg/kg (low dose group), 3 mg/kg (middle dose group) and 5 mg/kg (high dose group) groups, which were fed with Ivermectin of the corresponding dose, and control group was fed with saline instead of ivermectin. After 24 hours of the last administration, all the rats were executed to take testis and epididymis for analyzing the toxicity of the reproductive system. The results showed that there was a reproductive toxicity on testis and epididymis when the dose of ivermectin was over 3 mg/kg; Testis coefficient and epididymis were affected, such as the density and alive rate of the sperm were decreased, but the sperm viability was not influenced, and no results showed correlation with Ivermectin dosage in all groups.

This article studied the pharmacokinetics and bioavailability of 10% Ampicillin suspension in vivo of target animal-fowl. The result showed that the absorption of 10% Ampicillin suspension was quick; the reaching peak time was short,about 0.89 h; the elimination half-life was about 2.5 h; and the absolute bioavailability was 35.27%,lightly higher than the absolute bioavailability of Ampicillin solution (34.30%); the bioequivalence of 10% Ampicillin suspension with oral administration to fowl was 109.8%. 

32 strains of Escherichia coli (E. coli) were isolated from 50 E. coli infected chickens in six farms in Shandong. As a result, all of them were resistant to 12 antibiotics to a greater or less degree. In the screening test of plasmid DNA of 14 strains E.coli isolates and plasmid DNA fragments generated by Hind Ⅲ, simple agarosegel electrophoresis was employed. The results showed that most of E.coli isolated from the same infected flocks on the same farm had similar plasmid profile, and its fragments sizes were similar too, and that these isolated had common plasmids. The results showed that those isolates from different chicken flocks and had different plasmids were in different serotypes, the plasmids could be transferred in different serotypes of E.coli.

The hyperimmunized yolk antibodies against porcine circovirus serotype 2 (PCV-2) was obtained,and its cure effect was tested. After the hens were immunized with the inactivated vaccine of PCV-2 and eggs of hyperimmunity were collected, the hyperimmunized yolk antibodies against PCV-2 were prepared; the proof-test of live bacteria,safe experiment,antibody titer by double diffusion test and artificially infected piglets with the antigen were conducted; and the antibodies were used on the farms to evaluate the clinical effect. The antibody titer reached to 1∶64, the cure rate of artificial infection was 100% and the cure rate of clinic treatment test was 96%. The antibody was worth to be used in practice on the farms.

From 2006 to 2008,116 hepatitis B positive serums were detected in 610 dairy cow serums in Guizhou province, the mean positive ratio of HBsAg was 19%(116/610). 29 infectious bovine rhinotracheitis positive serums were detected by neutralization test in 534 dairy cow serums, the positive ratio was 5.4%(29/534). The antibody of dairy cow contagious pleuropneumonia were negative in 534 dairy cow serums by CFT. HBV particles were observed by electron microscopic observation. The results indicated that dairy cow type B hepatitis and infectious rhinotracheitis existed probably in Guizhou province. The breeding cow industry should pay close attention to dairy cow type B hepatitis and infectious rhinotracheitis.

The results showed that significant correlation was obtained between a majority of body size parameters. Significant correlation was obtained between a majority of body carcass parameters. And there was significant correlation between some of body size parameters and body carcass parameters. The results of this study provided basis data to the farther research,and provided theoretical basis for the improvement of the Sansui duck.

An experiment was conducted to determine the effect of chromium picolinate and magnesium aspartate on pork quality. 48 Duroc×Landrace×Yorkshire finishing pigs with initial average body weight (86.0±5.1) kg were divided into four dietary treatments(Ⅰ,Ⅱ,Ⅲ and Ⅳ) with 4 of each treatment. Treatment Ⅰfed corn-soybean meal (C-SBM, control), treatment Ⅱ fed C-SBM+1.0% magenesium aspartate (MgAsp), treatment Ⅲ fed C-SBM+0.3 mg/kg chromium picolinate (Crpi), treatment Ⅳ fed C-SBM+1.0% MgAsp+0.3 mg/kg Crpi. 2.8 kg upper diet/day was fed to each pig for seven days before transport to slaughterhouse. Pork quality data were collected and measured. Serum cortisol level was significant decreased after supplement with MgAsp, Crpi and MgAsp+Crpi (P<0.05). The glycogenolysis was decreased after supplement with Crpi and MgAsp+Crpi(P<0.05). The content of lactic acid, pH and drip loss of the back longissimus and the biceps femoris was decreased in pigs supplement with MgAsp, Crpi and MgAsp+Crpi than the control pigs, and the occurrence of PSE (pale, soft and exudative) pork decreased 75.1%, 49.8% and 75.1%, respectively(P<0.05). The water holding capacity was not different between four treatments. The drop loss, color scores of back longissimus and biceps femoris were better in pigs supplement with both MgAsp and Crpi than pigs supplement individually. These data indicated that MgAsp and Crpi may favor to each other for anti-stress and improvement the meat quality,addtion of MgAsp and Crpi together gave the best effect.

Intramuscular fat is one of the key factors to impact the meat quality,the intramuscular fat content is high,the marbling is rich,the myesthesia is tender,and the mouthfeel is good. This paper is started with the main factors of affecting the content of the intramuscular fat,analysis the factors of the different hereditary basis,age,slaughtering site and ration to impact the intramuscular fat. It provides a basis for producing high quality beef.

In this study the culture conditions and culture medium components of the Lactobacillus caseis-03 were optimized by means of single factor and orthogonal experiments. The results showed that Lactobacillus caseis-03 was anaerobic bacteria;the optimum culture conditions were as follows: initial pH 6.5, 35 ℃, inoculation 1.0%, static cultured in improved MRS medium for 24 h; the optimum culture medium components were as follows: peptone 12 g, yeast extract 6 g, beef extract 6 g, glucose 18 g, sodium acetate 5 g, C6H17N3O7 2.15 g, Tween 80 1 mL, MgSO4·7H2O 0.58 g, MnSO4·4H2O 0.05 g, K2HPO4 2 g, H2O 1000 mL. D610 nm value could reach 1.832, viable bacteria counts could reach 3.22×1010 CFU/mL in the optimum culture medium and conditions.

To study the hemostasis effect of disposable packing hemostasis on bleeding due to trauma of medium-sized artery,and compare the hemostasis effect with that of styptic powder.9 miniature pigs were divided into packing hemostasis group,styptic powder group and blank group.Establishing bleeding of forelimb medium-sized artery models on miniature pigs,haemostasis treatments were performed on individual group and parameters were measured, such as hemostatic time,regional maximum temperature and blood pressure.In addition,the tissues surrounding the haemostatic spot were picked for observation. When comparing the haemostatic effect,we found that the disposable packing hemostasis and styptic could stop bleeding quickly,and the effect of these two methods on hemostatic time has no difference;the temperature raised by styptic powder were higher than that of packing hemostasis,in addition, they all cause light tissue damage for raw surface and surrounding tissues. Disposable packing hemostasis could effectively stop bleeding caused by forelimb medium-sized artery,it could act as a secure and effective dressing for treating acute hemorrhage.
