The purpose of this experiment was probing the effect of multienzyme in the miscellaneous meal diet and prepara-〖WT〗〖HT〗〖HJ〗〖LM〗

tions on growth, metabolism and the blood indices in weanling piglets.48 weanling piglets was used and divided into 2 groups. 1% multienzyme was added in the experimental group on the basic of control group. The experiment was divided into two stages, earlier stage (1 to 21 d) and later stage (22 to 50 d). The results showed that experimental group was better than control group in the daily gain, the feed to gain ratio(F/G) and the incidence of diarrhea in 1 to 21 d (P<0.05),and in full distance test (1 to 50 d) experimental group was better than control group in the daily gain, F/G and the incidence of diarrhea, but only F/G was different markedly (P<0.05).In 21 d the digestibility of all the nutrients of experimental group were better than those of control group (P<0.05) except crude ash. In 50 d the digestibility of crude fat and crude fibre of experimental group were better than those of control group (P<0.05), and the others had no difference. The results of blood biochemical indexes showed that in 21 d and 50 d the experimental group were all better than those of the control group in TP and ALB, but had no obvious difference; the experimental group was lower than that of control group in BUN and only had obvious difference in 21 d (P<0.05); the experimental group was higher than that of control group in GLU and only had obvious difference in 21 d (P<0.05).

The aim of this experiment was to study the effects of 4 kinds of compound additives on performance and serum biochemical index in Black-bone silky fowl. One hundred and eighty chickens at the age of three hundred days were selected and randomly allocated to five treatment groups with three replicates, four diets containings four different compound additives (0.5%, 0.2%, 0.2% and 0.2%) and basic diet were given to feed these chickens for 78 days in order to. The results showed that laying rates were increased with diets of compound additives. When compared to control group, the laying rates of group 1, 2 and 4 were significantly increased by 6.09%, 13.82% and 6.57% (P<0.05), respectively. The values of average egg weight and the feed conversation rate of group 2 were the least in these treatment groups and were decreased by 2.05% and 12.5% compared to control group (P<0.05). The content of total cholesterol (TC) and glucose (GLU) of group 1 were decreased by 21.24% and 8.93% (P<0.05), and the content of creatine kinase (CK) was increased by 32.73% (P<0.05) compared to control group. There were no difference between group 2 and control group at all the serum biochemical index. The concentration of malonaldehyde (MDA) and CK of group 3 were lower than that of control group (P<0.05). The content of GLU of group 4 was increased by 13.97% (P<0.05) and the content of TC was decreased by 13.72% (P<0.05) compared to control group.

An experiment was conducted using three Chinese Holstein cows in early lactation under hot environment,in a 3×3 Latin square design,to determine the effects of the basal diets supplemented with 0,8,15 g/d niac in each cow as control group A,group B,group C on performance,maintenance behavior and ion chroma of serum.The feeding experiment had been carried out for 63 days.The results showed :① compared with control group A,milk yield of group B and C was increased by 29.9% (P<0.05),22.0% (P<0.05),respectively,and 4% fat corrected milk yield was increased by 28.5% (P<0.05),21.5%(P<0.05),respectively,there was no significant difference between group B and C (P>0.05);dietary niacin had no effect on the content of milk fat, milk protein and milk sugar; ②compared with control group, there was no effect on the hours of standing and lying, the number of micturition and defecation, the number of drinking-water,but rumination time was significantly increased (P<0.05). Also, there was no significant difference between group B and group C (P>0.05); ③compared with control group A, the Ca2+,K+,Na+ chroma of serum of group B and group C was increased in diffferent extent, but there was no significant difference between group B and group C (P>0.05 ).The Mg2+ chroma of serum was increased,and the Cl- chroma of serum was decreased, but not reached significant levels (P>0.05). As performance,maintenance behavior and ion chroma of serum were concerned,niacin supplementation with 8 g/d in the ration of lactating cows had the best effect and released the effect of heat stress in hot environment.

To observe the influence of Chinese medicine Qingongsan on the T,B lymphocyte of spleen and peripheral blood in mice, the flow cytometry method were adopted to determine the percentage of CD3+〖KG-*2〗, CD8+,CD4+ and CD19+ lymphocytes and CD4 +/CD8 + value. Chinese medicine Qingongsan signifcantly enhanced the percentage of CD3+, CD4+ and CD19+ lymphocytes and CD4 +/CD8 + value in peripheral blood and recovered the percentage of CD3+,CD3+CD8+and CD19+ lymphocytes in mice inhibited by cyclosporin. The results showed that Chinese medicine Qingongsan can significantly enhance the immune function and recover the immune function in mice inhibited by cyclosinfluence.

In the cytochrome P450(CYP) superfamily, cytochrome P4502A5(CYP2A5) is induced by a number of structural diverse of hepatotoxic compounds in the murine liver, plays a unique up-regulation in the process of hepatitis and hepatotoxicity. Researching the mechanism of expression and regulation, catalysis, induction or inhibition the metabolism of CYP2A5 gene, and chemical xenobiotics metabolism, play a key role in guiding the rational drug use of human clinical, preventing from cancer diseases, improving drug efficacy and reducing the side effects of drugs. This paper gives an overview of the characteristics of CYP2A5, the affecting factors and their relationship with the disease.

To assess brain cell apoptosis induced by fluoride in their offspring, 16 one-month old Wistar albino rats were divided randomly into two equal groups, each with six females and two males. The first group of rats served as the untreated controls; the second group received high fluoride(HiF) in their drinking water(100 mg/L NaF); After the animal model was established, the rats were allowed to breed, and 16 offspring rats in each group were randomly selected for the experiments. The treatment for these second and third generation rats was the same as for their parents. At 0, 10, 30, 60 and 90 days after birth, these offspring rats were anesthetized and their brain cells prepared for flow cytometry. In comparison with the controls, the percent of brain cell apoptosis in the offspring rats in fluoride groups was obviously higher, especially in third generation rats. In comparison with the second generation, brain cells apoptosis in different aged third generation rat was increased, especially at 10, 20, 60 and 90 days(P<0.05). These results indicate that fluoride can accumulate in different generation, and the longer the period sustained, the severer damage of the brain in offspring animal occurred. 

To investigate the effects of Saruma henryi olive in analgesia and peristalsis, the pharmacological experiments were carried out by some classical methods, such as mice writhing test,mice models of hot-plate and toner test. The purpose of this study was to compare the levels of antibody titer to PRRSV and screen the best immunization method for PRRSV. The nucleocapsid protein of PRRSV was used as immunogen to immunize BABL/c mice by traditional immunization, NC filter immunization, intrasplenic immunization and gel particle containing special bands. Then the titer of positive sera was detected by indirect ELISA, and the specificity was analyzed by Western blotting. After the third immunization, the titers of serum in different mice groups immunized by the four groups were 1.28×105, 1.28×105, 3.2×104, and 6.4×104, respectively. The results showed that the four immunity methods had obtained high titers polyclonal antibody, comparing with traditional immunization, the other three methods had more advantage in conserving antigen. Intrasplenic immunization was the best immunization method, which had the best immunity effect,using the least antigen and the shortest time.

The methods of extracting trehalose from S.boulardii was preliminarily studied in this experiment. Through the contrast between three kinds of extractant and using single and complex treatment,grope the best way to extract trehalose in S.boulardii: extractant is 5% trichloroacetic acid, water bath at 100 ℃ and 30 min after 10 min ultrasonication. Extracting supernatant was hydrolytic and determined glucose with the biosensor and calculate the content of trehalose.

According to the sequence of chicken insulin-like growth factor (IGF-1) gene which submitted to GenBank, designing a pair of primers. The partial fragment of IGF-1 gene was cloned and sequenced on layer and meat quails. The length of the fragments were 806 bp and 804 bp. Now the two sequences had been sent to GenBank and registered comparison of the fragment sequence with that Gallus gallus IGF-1 gene was carried out through BLAST in GenBank, the result showed that the homologies of the nucleotide sequence was 93% with Gallus gallus. And two strains of quail had the homology of 99.38% in the nucleotide level.

As porcine circovirus type 2 (PCV-2) is an important pathogen in swine, a PCR method was established by using a set of primers derived from the published nucleotide sequence of the PCV-2 genes deposited in GenBank. In order to confirm its specificity, the PCR was performed by using the template DNA from the reference virus, and the expected products of 536 bp were only detected in PCV-2, whereas no specific band was detected in other virus such as PRV, CSFV, PPV and PK-15 cells. The lowest DNA consistence which could be detected by the PCR method was 29 ng/L, and the sequence of aim band was belong to PCV-2. To further investigate the prevalence of PCV-2, 91 clinic samples were obtained from Shanghai and circumjacent regions in 2008, and submitted to PCR detection. The data indicated that PCV-2 in 52 samples were positive, while positive proportion was 57.14%. The clinical experiments indicate the PCR method could be used in diagnosis and epidemiology investigating.

To study the expression of the duck hepatitis virus 1(DHV1) VP1 gene in E.coli ,one outer primer pair and one inter primer pair containing EcoR Ⅰ and Sal Ⅰ according to complete genome of DHV1 were designed. PCR products were re cycled by outer and inter primer pair .The PCR products and pET-32a vector were digested by EcoR Ⅰ and Sal Ⅰ to construct the recombinant plasmids.Then the recombinant plasmids were transfected into E.coli BL21 and Rosseta. After digestion, PCR and sequencing the target gene were insert expression vector correctly. With different concentrations of IPTG induced bacteria express VP1 gene, the bacterias were collected and examined by SDS-PAGE.The results showed that the VP1 gene fragments hard to express in E.coli Rosseta and BL21.

In order to develop novel piglet’s Paratyphoid-ED bivalent vaccine, SLT-ⅡvB gene without signal peptide was amplified from pMDSLT-Ⅱ and subcloned into pYA3493. pYA2B was electroporated into C500crp-/asd- mutant. SDS-PAGE and Western blotting were done. Mice were i.p. immunized with C500crp-/asd-(pYA2B) and C500crp-/asd-(pYA3493) and i.p. challenged with 2 LD50 ED3 and 10 LD50 C78-1. 2 SLT-ⅡvB was expressed in C500crp-/asd- mutant. The C500crp-/asd-(pYA2B) immunized group was 60% protected mice against ED3 challenging and control groups couldn’t protect mice. All immunized groups induced 100% protected mice against C78-1 challenging. These results provided base for developing piglet’s Paratyphoid-ED bivalent vaccine and further improving of immunity effect.

A pair of primers was designed according to the sequence of porcine Th1-type cytokines,IL-2,IL-12p40 and IFN-γ gene as well as housekeeping gene β-actin,respectively,and a recombinant plasmid containing the target gene was constructed as a standard control for each cytokine.Then, a real-time PCR assay based on SYBR Green I for detection of IL-2,IL-12p40,IFN-γ and β-actin was established,respectively.Every assay had a good linear relationship between initial templates and Ct values,and the correlation coefficient of the standard curve was over 0.997.It was highly sensitive and had a detection limit of 100 copies/μL.It was highly specific and there was single specific melting peak of every cytokine.It was highly reproducible and had a coefficient of variation less than 3.5 percent for both intra- and inter-assay.The established assays were successfully used to detect IL-2,IL-12p40 and IFN-γ mRNA expression levels in peripheral blood mononuclear cells from piglets experimentally infected with porcine reproductive and respiratory syndrome virus (PRRSV).The high sensitivity,specificity and reproducibility of the assays indicated that the SYBR Green I real-time PCR could be used as an effective tool for detection and quantification of these Th1-type cytokines.

TGF-βRⅡ plays a key role in TGF-β signal transduction, TGF-βRⅡ was selected to study the effects of TGF-β signal pathway in the cycling growth of cashmere. Assessed TGF-βRⅡ expression by RT-PCR in the skin of Inner Mongolia Cashmere goats at different development stages. Results of this study showed that the expression of TGF-βRⅡ in January, March, May and September. TGF-βRⅡ might regulate the cycling growth of cashmere with binding ligand TGF-β.

The single nucleotide polymorphisms (SNPs) within the chemokine receptor CXCR1 gene in Holstein cattle,Luxi Yellow cattle and Bohai Black cattle were investigated by DNA sequencing technology,nested-PCR and CRS-PCR methods.Sequencing results showed that four SNPs in exon 2 were identified,which they were 291 bp (C/T),333 bp (C/T),337 bp (A/G) and 365 bp (C/T).The value of polymorphism information content indicated(0.25<PIC<0.5) that all the four SNPs were moderate polymorphism in Holstein cattle,Luxi Yellow cattle and Bohai Black cattle.Fur
thermore,C, C, A and C predominan allele were identical at 291 bp (C/T),333 bp (C/T),337 bp (A/G) and 365 bp (C/T),with the allelic frequencies 0.82/0.67/0.65,0.80/0.74/0.71,0.70/0.65/0.82,0.56/0.58/0.56 respectivly. Chi-square test indicated all the sites of Bohai Black cattle were in accordance with the Hardy-Weinberg equilibrium,while Holstein cattle meet Hardy-Weinberg equilibrium (P>0.05) at 291 bp (C/T),333 bp (C/T) and 337 bp (A/G),and the Luxi Yellow cattle meet Hardy-Weinberg equilibrium only at 365 bp (C/T).

The results showed that there was one single nucleotide mutation (A562→C562) at exon 2 of GDF9 gene in Qianbei-pockmarked goats,and this mutation resulted in an amino acid change: glutamine→proline.The results indicated that there were two genotypes (AA and Aa ) detected in Qianbei-pockmarked goats,genotypic frequency was respectively 0.7273 and 0.2727; frequency of A and a allele was 0.8637 and 0.1363. The results showed that there was one single nucleotide mutation (C480→G480)at exon 2 of BMP15 gene in Qianbei-pockmarked goats,and this mutation resulted in an amino acid change: glutamine→glutamate. The results indicated that there were two genotypes (BB and Bb) detected in Qianbei-pockmarked goats,genotypic frequency was respectively 0.7000 and 0.3000; frequency of B and b allele were 0.8500 and 0.1500.

The 3′UTR polymorphism of IGF-ⅠR gene was detected by PCR-SSCP in 58 red steppes. The results showed that two allelomorphic gene, three genotype of IGF-ⅠR gene were detected. The associativity was analyzed between the polymorphism of slaughter and carcass traits. The results indicated that procured meat percentage and the ratio of meat in bone for AB genotype was higher than AA and BB genotypes(P<0.05), the rate of fat coverage for AA genotype was higher than BB genotype(P<0.05),and was no significant difference with AB genotype(P>0.05).

Breeding data of 1835 individual Sanhe cattles recorded from 1977 to 2005 was collected at Breeding Centre of Inner Mongolia and used for genetic statistic analyses of milk yield (305 days), stature, body length, chest girth, shank circum and body weight. By using animal model, REML and DMU were utilized to estimate the genetic parameter. The results showed that the heritability of seven traits above was 0.26,0.18,0.38,0.21,0.28,0.20 and 0.39 respectively. The genetic correlation between body weight and body size was among 0.51 to 0.72.

The 7 microsatellite markers on chromosomes X were used to construct a linkage map of Cashmere goats with a 794 members half-sib pedigrees after paternity testing. The results showed that the number of alleles for 7 markers was 9 to 14, the heterozygosity was 0.585 to 0.918, the average heterozygosity was 0.726, and polymorphic information content (PIC) was 0.759 to 0.897, the average polymorphic information content was 0.834. Length of the linkage map of chromosome X we built was 139.4 cM, and the sequence of loci was coincident with the SM4.7, and could be used in quantitative trait loci (QTL) searching in Cashmere goats.

In the present study,the superovulation records of 218 2 to 5 year-old multiparous boer goats were analyzed,and the repeated superovulation schemes were designed according to superovulatory response and the number of recovered embryos. All of donor does were divided into five groups,according to records of first superovulation response,for repeated superovulation in decreasing FSH dosage at 60 days intervals. Group 1 only had follicle but no ovulation; group 2 was not in estrus after superovulatory treatment; group 3 had the number of corpus luteum≤4; group 4 had the number of corpus luteum 5 to 14,and group 5 had the number of luteal number≥15. The results showed that the effective utilization rate of donor of five groups were 100%,75.00%,71.88%,93.10% and 93.65% respectively,there was no significant difference(P>0.05); the number of all embryos of five groups were 14.00±5.73,12.10±6.78,11.77±7.26,13.84±7.04 and 18.03±9.06 respectively, group 5 was very significant higher than other groups(P<0.01); the number of recovered embryos of five groups were 12.81±6.04,10.33±8.12,10.08±7.14,11.34±6.58 and 15.37±7.64 respectively, group 5 was significant higher than 2,3 and 4 groups(P<0.05), but there was no significant difference with group 1(P>0.05) . In conclusion, it was feasible to adjust the does of FSH for repeated superovulation according to the effect of first superovulation. Repeated superovulation treatment by increasing FSH dosage could make good response for the poor or no response of the first superovulatory treatment and there were more than 70% of the effective donor to be used.

In order to study the genetic effect of the PRLR,RBP4 gene and the association between different genotypes and litter size in large white, 278 sows with 1418 litters record on litter size were detected for the polymorphism and association analysis. The results showed that the B allele of PRLR gene with the frequency 0.538 was higher than A allele. The effect of different genotypes on total number of born was present as BB>AB>AA at the first parity,second parity,later parities and all of the patities,but with no significancy. Assocoation analysis between different genotypes and the number of born alive showed that there were no significant in all 1418 parities. For the RBP4 gene, only AA genotype homozygous was detected in this population.

To help find the physiological functions of the chicken lysozyme gene,the current study was designed to investigate the association of chicken lysozyme gene polymorphism with chicken body weight. 200 female Jinghai Yellow chickens were involved in this experiment. To determine gene mutations,DNA analysis of the lysozyme gene,including single strand confirmation polymorphism (SSCP),and direct DNA sequencing was performed. Three single nucleotide polymorphisms(SNPs) were indentified,all of them were silence mutations that caused no alteration in amino acid sequence. Chi-square test showed that only genotypes of the 1411 locus were at Hardy-Weinberg disequilibrium. For the 111 locus,body weight of the genotype AA chicken at twelve and sixteen-week of age was significantly higher than that of genotype GG chicken (P<0.05). For 1411 locus,body weight of genotype GG chicken at four-week of age was significantly lower than that of genotype AA chicken (P<0.05). In the meanwhile,for 1426 locus,associations between genotypes with body weight were not significant (P>0.05). Statistical analysis of haplotype of the chicken LYZ gene and body weight approved the genotypes. Chicken of haplotype ANC were heavier than other chicken except four-week of age, and chicken of haplotype GAC were haeavier at four-week of age than other chicken. Therefore, the lysozyme gene might be -a potential marker for use in marker-assisted selection programs in Jinghai yellow chicken breeding.

The polymorphisms of the 2,4-dienoyl CoA redutase 1 (DECR1) gene in exon 2 site was detected using PCR-SSCP in 490 pigs of 5 pig breeds including Shanxi Mashen, Shanxi White, Landrace, Yorkshire and Duroc. The results showed that there was a polymorphism in the exon 2 site and three genotypes (AA, BB and AB) were found. Analysis of population genetics suggested that the frequency of B allele was dominant among the studied breeds. The results of Hardy-Weinberg equilibrium test indicated that Shanxi White and Duroc reached the equilibrium. All of the breeds had intermediate polymorphism information contents.

Application of immunoproteomic methods in research on Brucella abortus.Two-dimension electrophoresis was used to separate the membrane protein of Brucella abortus strain 544A grown under laboratory condition,and Western blotting was used to screen immunogenic proteins by serum of guinea and bov anti-Brucella abortus IgG.After ingel digestion,12 immunoreactive spots were identified by LC MS/MS.All the proteins were searched by the bioimformation tools,there were not only membrane proteins,but also cytoplasmic proteins.Most of them were involved in protein synthesis and metabolism.The immunoproteomic methods can be successfully applied in identifying immunogenic proteins of Brucella,which helps to search for other immunogenic proteins and lay a foundation for searching vaccine candidate antigens in future.

The liver and spleen samples of 600 unhealthy chickens were collected randomly to investigate infection of MDV,CAV,REV and REOV in Shandong province using Dot blotting. The clinic tissues came from six different districts including yantai, binzhou, linyi, tai′an, liaocheng and qingdao of Shandong. The results suggested that the checkout ratios of the four virus are 5.17% (MDV), 8.17% (CAV), 1.17% (REV) and 2.5% (REOV) respectively which were lower than the former research; certain degree duale infection and triple infection were found out. The result indicated that the immunodepressive virus are well controlled except individual flock in which the rate of infection was higher than the sum of other flocks; there was a significant discrepancy of the immunodepressive illness in different flocks where the management level were uneven.

In this study, 4 mL canine distemper virus being 104.5 TCID50/0.1 mL, was used to immunize wusuli-raccoon dog which was strain economic animal. After 10 days, animal blood was collected, while blood serum was disassociated. Neutralization test was made to evaluate the potency of antibody, during this test, virus was 200TCID50 , blood serum whose volume was equal with virus was diluted by times after times, while immunoglobulin was extracted crudely by saturated ammonium sulfate. The results clearly suggested that under the project that the blood serum was diluted for 2917 times, the immunoglobulin which was extracted crudely by saturated (NH3)2SO4 was diluted for 2344 times, there was 50% Vero cytoprotection without CPE. 24 white raccoon dogs with this disease were therapied by the whole serum above, and 22 ones was survival, therefore,we demonstrated that antiserum, made from strain animal such as fox, could be used tocure canine distemper virus.

126 portions of the lung tissue samples or nasal swabs with respiratory symptoms, collected from number of pig farms in Henan province, were used to isolate and identify Bordetella bronchiseptica(Bb).According to bacterial culture characteristics, Gram staining, biochemical tests and PCR, 11 strains Bb have been successfully isolated. Three important virulent factors(fha,prn,dnt) for Bb were tested by PCR, most of the PCR results obtained corresponding DNA target bands except that two strains Bb didn’t get prn gene target bands. Erythrocyte agglutination assay showed all kinds of strains were able to agglutinate pig and sheep’s erythrocyte at different degree. Skin necrosis in suckling mice showed all kinds of strains were able to produce skin necrosis to different extent. Virulent strains(HN0610, HN0806, HN0922, HN0827) were confirmed by mouse lethal.

According to the sequence of duck circovirus (DuCV) in GenBank (AY228555),two pairs of primers were designed by Primer Premier 5.0.A nested-PCR assay for rapid detection of DuCV was established.The results showed that a specific 340 bp fragment was amplified from liver,thymus gland,bursa of Fabricius,kidney,spleen collected from Wangjiang county of Anhui province,but no bands were amplified with templates extracted respectively from normal duck embryo,healthy duck liver tissue,duck plague virus (DPV),avian influenza virus (AIV subtype H9),Newcastle disease virus (NDV),infectious bursal disease virus (IBDV),reticuloendotheliosis virus (REV),chicken infectious anemia virus (CAV),Escherichia coli (E.coli) of duck origin and Riemerella anatipestifer(RA).Sensitivity of the first and second amplifications by the nested-PCR assay was 1 ng and 1 fg,respectively.The sensitivity of the second amplifications was increased by 106 times.Results showed that the nested-PCR assay could be used as a method for the diagnosis and detection of clinical cases of DuCV and epidemiological investigation.

Compound traditional chinese medicine (CTCM) had been selected successfully in the test,which was identified as the best anti-bacterial effect against colibacillosis in chickens. CTCM was conducted to the development of ultrafine powderthe common fine powder,the particle size of ultra-fine powder was smaller and distributed more regularly.Only fragment of tissue was observed as tissue cell was broken in ultra-fine powder while the tissue cell of common fine powder was still intact and the primary profile could be observed (e.g. stone cell and catheter) in common fine powder.The dissolution test showed that the dissolution of citric acid,main active component of Ebony,was increased by 78.6 percent under the same condition. Comparing the inhibition effect against avian E.coli,the ultra-fine powder through simple extracted showed a better antimicrobial behavior than that extracted from common fine powder of CTCM. ②The experiment in which the diseased chicken were treated by feeding with CTCM,indicated that the mortality of the high and middle dose groups of ultra-fine powder was significantly lower than the infected control group (P<0.01),however,the groups of the low ultra-fine powder dose and common fine powder had no significant difference compared to the infected control group (P>0.05); in the high dose group of ultra-fine powder,the mortality rate was significantly lower than the common fine powder group (P<0.05),and the cure and effective rate also showed significantly difference between the two groups (P<0.05).

In order to test the inactivated emulsion vaccine against Newcastle disease, infectious bronchitis and avian influenza (H9N2 subtype) safety, 14-day-old leghorn chickens were vaccinated through hypodermic injection in neck. The weight of each chicken was measured every day until 28-day-old. The statistical significance between the vaccinated group and the control was compared by t test. The results showed that there was no significant difference between the two groups weight. An indirect ELISA was established for the rapid detection of specific Japanese encephalitis virus (JEV) antibodies in swine using a recombinant E protein as an antigen. The recombinant E protein was expressed from recombinant plasmid pET28a-E in Escherichia coli. The various factors affected the experiments were optimized. This assay was optimized using 2 μg/mL E protein as coat antigen, the optimal coating condition of JEV for ELISA was incubated at 37 ℃ for 2 hours, 1∶160 dilution of testing serum and 1∶5000 dilution of HRP-labeled goat anti-swine IgG, the best blocking buffer was BAS with 1% consent ration. The critical value of positive and negative of ELISA was D492 nm=0.254, this method is not with the serums of CSFV, PRRSV, PCV2, PRV reaction, D492 nm<0.254, explain its specificity. Test against 150 serum samples from Jilin province by this assay detected 43.33% positive, which showed 90.77% agreement with ELISA kit test. Therefore, this study indicated that an indirect ELISA method for specifically detection JEV in serum was successfully established.

Iron dextran treated calf anemia were studied in this article,the author detected concentration of hemoglobin, cholesterol, malonaldehyde(MDA), catalane(CAT), in order to validate therapeutic efficacy of iron dextran.The results showed that hemoglobin and ferrous ion concentration were significantly higher in experimental group compare to control group (P<0.05); cholesterol and MDA were significantly lower in experimental group compare to control group (P<0.01); but the concentration of CAT were significantly higher in experimental group compare to control group(P <0.05).Therefore, iron dextran significantly increased ferrous ion concentration and hemoglobin level,meanwhile corrected oxidative stress because of iron deficiency. There might be good effect for calf anemia with iron dextran.

In order to provide a theoretical basis for rational use drugs in clinic, the regular pattern of different doses of Tilmicosin on blood pressure and heart beat in Saibei rabbits were studied. 24 healthy Saibei rabbits were arranged into 6 groups randomly, the doses of Tilmicosin of the groups were control group(0 mg/kg),Ⅰgroup(20 mg/kg),Ⅱgroup(40 mg/kg),Ⅲ group (60 mg/kg),Ⅳ group (80 mg/kg),Ⅴgroup (100 mg/kg) respectively. The blood pressure and heart beat were measured after 7 d feeding with different doses of Tilmicosin. The systolic pressure was decreased by the increased of Tilmicosin,the diastolic pressure was increased gradually as well as the heart rate.The change of clinical symptoms of V gronp was the most significant. The results showed that the suitable dose of Tilmicosin was 20 mg/kg and the safety dose of Tilmicosin for Saibei Rabbit was 100 mg/kg.

The post antibiotic effect (PAE) of apramycin and combined with amoxicillin or ampicillin against Staphylococcus aureus, Salmonella, Escherichia coli in vivo, were studied by the method of colony counting and neutropenia associated with mouse thigh with bacterial infection model. The experimental results showed that, the PAE of apramycin were conspicuous against Staphylococcus aureus, Salmonella, Escherichia coli in vivo, and were enhanced with the increasing concentration, showing strongly a concentration-dependent. Apramycin combined with amoxicillin or ampicillin had synergetic effect on the PAE of Staphylococcus aureus, Salmonella, Escherichia coli in vivo.

BTB (Bovine tuberculosis) is series of chronic disease caused by Mycobacterium bovis, Mycobacterium tuberculosis, Mycobacterium avian, et al. The lethality rate of BTB is one of the highest disease in all bovine transmissible diseases. It is very urgent to prevent and control the transimission and pervasion of this disease, since it can be transmited between domestic cattle and human and without effective treatment, which seriously affect the cattle industry and human health. The main measures of controlling BTB including in quanrantine cows on time and remove the infected cows. In recent years, with the development of life science, the traditional bacterial detection method, immunology detection and molecular biology methods are combined well together, particularly the development on molecular levels gradually improved the bovine tuberculosis of diagnostic method, meanwhile, many of specific Mycobacterium tuberculosis antigens and genes were found, which palyed a signifcant role in elucidating the molecular evolution and genetic variation of bovine tuberculosis Mycobacterium.

yometra is a common disease of canine. The majority of canine pyometra incidence was observed in metoestrus.It can be classified as open-cervix and closed-cervix. Pyometra is a uterine inflammatory disorder characterized by cystic endometrial hyperplasia, the increasing of progesterone and the release of estrogen, and by ascending uterine infection with vaginal bacteria. In this article, the etiology, diagnosis and treatment of canine pyometra were reviewed.

Using the natural infection disease clinical coccidiosis only 150 chicken for supplying DN-DC, Chinese betel nut, conventional drugs, such as, antifebrile dichroa, betelnut, artemisia apiacea, radix bupleuri, was compared with western medicine ball net.The results of experiments: herbal treatments effective cure broiler coccidiosis was 94%, than higher western medicine ball net (88%) 6 percent, compared with controls (62%) 32 percent. Chinese DN-DC anti-coccidia index (ACI) was 214.3, above western anti-coccidia index (ACI)for 183.2. Chinese herbal medicine (DN-DC) is efficient QiuChongYao resistance.

Experiments were conducted with twenty six-month old Liaoning cashmere goats with average live weight of 16 kg.The experiment was composed of two groups,there were cysteamine group and control group repectively.Each experimental group was feed by the same basic diet.The cashmere goats in the control group was only feed with basic diet,while the CS group feed with the basic diet added with cysteamine 100 mg/kg W.B. one time every week.Effects of cysteamine on the physical quality of down and hair in cashmere goats was studied in the experiment.The periods of experiment had sixty days.The results showed that CS group of down lengh and hair lengh,down stretched length,hair stretched length were significantly higher than those of control group (P<0.05).Compared with control group,down strength and hair strength,down elongation and hair elongation of CS group there were no significantly different than those of control group (P<0.05).The results indicated that the cysteamine made better the cashmere quality of cashmere goats.

Test selection 16 wo duroc×landrace×big white 8-day-old grew up in three-way cross suckling piglets, according to birth date, litter size were randomly divided into four similar groups of four nests. The control group (Ⅰ) basic diet supple mented with 0.01% fed aureomycin, test group (Ⅱ,Ⅲ, Ⅳ) feeding basic diet supplemented with 0.01%, 0.05% and 0.10% of Bacillus subtilis, Bacillus subtilis on the study production performance of piglets. The results showed that Bacillus subtilis test feeding pigs with chlortetracycline compared with the control group, piglet average daily gain significantly increased the incidence of diarrhea have been reduced, Bacillus subtilis aureomycin in each group were higher than the control group; Bacillus subtilis with 0.10% added as the best, but all groups were no significantly different (P> 0.05). The conclusion of Bacillus subtilis on improving the production performance of piglets with good results, Bacillus subtilis was fully capable of an alternative feed chlortetracycline.

This study was carried out to evaluate the effect of compound Leonurus Heterophyllus decoction on reproduction of sows. The diet of treatment group were supplemented with compound Leonurus Heterophyllus decoction one day, two days and three day postpartum respectively. The results showed that diets supplemented with compound Leonurus Heterophyllus decoction after delivery could shorten the time of postpartum lochiorrhea of sows and decreased endometritis significantly. Compound Leonurus Heterophyllus decoction groups had higher estrus rate than the control group. Moreover, the time of estrus of weaning sows in the three groups complied with compound Leonurus Hetrophyllus decoction was shorten significantly. The next litter size of treatment group was higher than the control group and the first group. From the results above, the author can conclude that compound Leonurus Heterophyllus decoction could enhance the reproduction performance of sows, decrease postpartum diseases and enhance the reproduction performance of sows.

To take home at the farmers formed the core group, the basis of the open cluster selection methods, the development of basic elimination criteria, a comprehensive selection index to guide finishing goats and eliminated, selection option.The results showed that the core group of age body weight of male and female goats were 29.77 kg±5.51 kg, 25.18 kg±5.67 kg, weight of adult male and female goats were 40.08 kg±7.73 kg, 34.90 kg±6.56 kg; based group of age body weight of male and female goats were 26.59 kg±3.68 kg, 24.09 kg±4.60 kg, weight of adult male and female goats were 35.33 kg±4.09 kg, 32.16 kg±6.83 kg; lambing rate was 152.33%. The significance test, breeding, after the core group, the basis of age group, the average weight of adult ewes was significantly higher than the average weight of pre-selection(P<0.01). No angle on a corner was the dominant, goats angle between the sexual and the non-existence of linkage.

A standard curve was constructed for detecting Pefloxacin by surface plasmon resonance(SPR) biosensor to provide a basis for the sample detection.Using amino coupling method to active the CM5 chip and fix FLE-OVA conjugate. The optimum immobilizing conditions of antigen and the concentration of antibody were confirmed by orthogonal experiment.The results showed that it responded desirable when the FLE-Ovalbumin was diluted to 50 mmol/L by Acetate buffer(pH 4.5). The limit detection and the range of the standard curve was respectively 1 ng/mL and 0 to 20 ng/mL. It obtained a lower limit detection when the antibody was diluted by 400 times.

In order to reducing badly atmosphere efficiency in henhouse, 5 henhouses were randomly allocated in Taoyuan foreign trade in Zhucheng city, each have 2000 10-day-age AA healthy henhouse. Enzyme was supplied with 0, 0.5, 1, 3, 5 g/kg respectively in broilers diet, which contains lichen sporangium, enterococcus faecalis, acid milk bacillus an so on. We collected the atmosphere in henhouse using Andersen-6 impacting airborne (FA-1,6-stage) fungal samples and CD-1 airborne samples. The result showed that, compared with the control group, the addition of 3 g/kg probiotics could remarkably decrease the air microbe quantity (P< 0.01), and it could dramatically decreased the fecal emission of ammonia ( P< 0.01) .
