Effect of β-mannanase on Performance and Apparent Nutrient Availability in Laying Hens

The objective of this study was to investigate the effect of β-mannanase added in SBM based diets on production performance and apparent nutrient availability of laying hens. Six hundred healthy Roman hens (laying hens of 33 weeks old) were selected and randomly divided into 3 groups with two hundred hens in each group. The control group and experiment group 2 and 3 were fed with a same diet added 0%, 0.03% and 0.05% β-mannanase respectively. The study lasted 35 days with the results showing that the egg weight (laying hens of 33 to 37 weeks old) of experiment group 2 and 3 increased 5.12% (P<0.05) and 7.08% (P<0.05) respectively than control group 1; the eggshell thick increased 11.60% (P<0.05) and 9.39% (P<0.05). The egg production rate of experiment group 2 and 3 (laying hens of 51 to 55 weeks old) increased 4.20% (P<0.05) and 5.55% (P<0.05) respectively more than control group 1. The nutrient availability of dry matter, organic matter, crude protein and calcium for experiment group 2 and 3 (laying hens of 36 weeks old) raised 6.15% (P<0.05) and 4.10% (P<0.05), 6.36% (P<0.05) and 6.60% (P<0.05), 2.04% (P<0.05) and 3.21% (P<0.05), 3.84% (P<0.05)and 7.08% (P<0.05) respectively more than control group 1; the egg weight raised 1.32% (P<0.05)and 2.46% (P<0.05). Figures above clearly demonstrate that β-mannanase helps to advance the production performance and apparent nutrient availability of laying hens.

The effects of formic acid on the fermentation quality, nutrient, biochemical oxygen demand of the watermelon skin silage were studied. The watermelon skin of Guilongxian and Guanlong with 0.3% and 0.6% of formic acid were bagged ensilaged. After sampled and analyzed, the results showed that in comparison with CK, formic acid significantly decreased the pH value, NDF and ADF content of watermelon skin silage(P<0.05), and there were no significant difference in the CP content, amount of the effluent , and BOD5 of effluent. The CP content of Guilongxian watermelon skin silage were significantly lower that of Guanlong(P<0.05), while the NDF and ADF content significant higher(P<0.05).

In order to study the effect of the symbiotic ferment culture of Lactobacillus rhamnosus and Saccharomyces cerevisiae on serum biochemical parameters and morphology of duodenum of broilers, 240 one-day-old broilers for a experimental period of 42 days,were randomly allotted into six groups, each with four replicates,and each replicate contain ten broilers.The control group was provided with basic diet,the antibiotic group was fed with basic diet supplemented with aureomycin at the level of 600 mg/kg, and the four test groups were fed with basic diet by providing water containing the complex probiotics,at the level of 0.1%,0.5%,1.0%,1.5%,respectively. The results showed that the concentration of Ca,P,total albumen,albumin and alobulin were significantly increased in serum of broilers by the complex probiotics (P<0.05),the concentration of total cholesterin and the proportion of albumin and globulin were decreased significantly (P<0.05),and the length of the villus in duodenum of broilers was increased significantly (P<0.05),the morphological characteristics of mucosal in duodenum was also improved. The results indicated that the complex probiotics had a certain impact on some biochemical in serum and duodenal morphology for broilers.

To study compound supplement of beneficial bacteria and traditional Chinese medicine on growth performance of rabbits,thirty 70-80-day old German white rabbits with same initial body weight were randomly divided into two groups,control group and experimental group.In experimental group, their basal diets supplemented with 3% of the beneficial bacteria and traditional Chinese medicine compound preparation,which the mixtures were feed for 42 days.The results showed that the feed conversion rate was increased 39.34%,the diarrhea rate was decreased 80%, and the mortality was decreased 75% in experimental groups.There were no significant difference on the slaughter indicators in two groups, and there was no poisoning symptom in experimental group;compared with the control group,the contents of arginine and ammonia of leg muscle were increased significantly in experimental group,but the levels of protein and amino acid were increased,but were not significantly.There was significant improvement in the total serum protein and serum glucose, and the total cholesterol and low density lipoprotein were decreased.Adding compound supplement of beneficial bacteria and traditional Chinese medicine in rabbit’s diets was helpful to improve the feed palatability,increase the feed conversion rate, regulate blood fat, improve quality and taste of rabbit meat.

To elucidate the genetic differences and genetic relationships among four chicken lines (Gushi Fast-feathering line,Gushi slow-feathering line, Short-green shell egg line and Yellow plumage-Silky black-bone chicken line) at genome DNA level and provide the basic data for utilization the heterosis between them, the RAPD assays were conducted in this study by using blood samples of the four chicken lines.40 primers which had produced good polymorphie RAPD markers were selected from 186 random primers.The genomic DNA pools from the total genome DNA of the four chicken lines were assayed by using the primers screened and 387 reproducible amplified fragments which were 100 to 2220 bp long were obtained.Average band sharing coeficient and genetic distance index among four chicken breeds were estimated according to RAPD polymorphie data.The results revealed that the genetic relationship between Gushi slow-feathering line and Short-green shell egg line was closer,which had a closer relationship with Gushi Fast-feathering line, respectively. whereas the genetic relationship between Yellow plumage-silky black-bone chicken line and other line was the most distant than any other pairs.

Preparative method of colloidal gold labeled monoclonal antibody against penicillic acid (PA) from Penicillium cyclopium was established and its reaction condition was optimized. The activity of the gold-labeled probe was determinated. Results showed that the optimization pH
was 8.2, the minimum proteins labeled 1 mL colloidal gold was 25 μg. It showed that the probe and anti-mouse anti-goat immnoglobulins were compaginated. The study provided foundation for colloidal gold immunochromotography assay against PA.

S1 gene of two strains of infectious bronchitis virus (IBV) isolated in Guangxi and Sichuan were amplified by RT-PCR technique and the cDNA sequences of those strains were determined and analyzed. Phylogenetic relations based on the nucleotide and amino acid sequences were analyzed among isolates and the reference strains in GenBank by DNAStar software. The results demonstrated that isolates had close relationship with J2, Q1 and T3, and less relationship with traditional IBV vaccine strains H120 and 4/91.

The DNA internal transcribed spacer (ITS) and 5.8S rDNA of ascaris from the Puma concolor and the Ursus thibetanus in Guangzhou zoo were amplified by PCR respectively using a pair of conserved primers and the amplicons were cloned into pGEM-T Easy vector. The positive clones identified by PCR amplification were sequenced and analyzed, and compared with relevant sequence in GenBankTM. The results revealed that the ITS and 5.8S sequence of the ascaris from Puma concolor and the ITS-2 sequence from Ursus thibetanus were significantly similar with that of Toxascaris leonina and Baylisascaris transfuga available in GenBankTM respectively. We concluded that the ascaris from Puma concolor represent the T. leonina, and that from Ursus thibetanus represent the B. Transfuga.

With the development of transgenic technology, it has become one of promising biotechnology to produce valuable pharmaceutical proteins using mammalian mammary gland as a bioreactor. Mammary gland bioreactors offer a high-yield,low-cost way to produce bioactive commercial pharmaceutical proteins that can not be produced efficiently by other methods and initiate a new approach for genetic engineering pharmaceutics. Recombinant human proteins produced by the mammary glands of genetically modified transgenic mammals have shown great commercialization prospects. So far, more than one hundred recombinant proteins have been successfully expressed in mammalian gland bioreactors, about thirty of which are in different phases of clinical trials. Antithrombin Ⅲ (ATrynR), the first protein for human of antithrombin deficiency. Here, the principles, advantages, research advances and industrialization process of mammary gland bioreactors were reviewed and problems and strategies for protein production were also discussed.

The ORF5 cDNA encoding GP5 protein of porcine reproductive and respiratory syndrome virus (PRRSV) stain Liaoning was amplified by RT-PCR. The cDNA fragment was further cloned and the complete sequences were determined. The results showed the cDNA of ORF5 was 603 bp, encoding 200 amino acids. Compared with the ORF5 gene of typical North American stain (VR2332 strain) and European strain (LV strain), it was found that there was 89.5% and 55.7% homology at amino acids level respectively. It indicated that the PRRSV strain Liaoning belongs to North American type.

In order to get high quality gold-monoclonal antibody of progesterone, the colloidal gold particles of 30 nm was prepared by the tri-sodium citrate reduction method, and then identified by two different ways. The key experiment conditions to develop gold-monoclonal antibody of progesterone were optimized. The spheroidal colloid gold particles of 30 nm was shown in color of bright red, and the equal particle ultrastructure was observed through transmission electron microscope(TEM); the minimal stable concentration (MSC) of gold-monoclonal antibody was 16 μg/mL, and the suitable stable concentration (SSC) was 19.2 μg/mL. The pH 8.5 was appropriate. Obvious low electron density halo was obversed around the particle of gold-monoclonal antibody complex, and the D520 nm was in the range of 0.2 to 0.4, thereby preparing for further research on the gold immunochromatography test strip of rapid diagnosis of pregnancy.

According to the conserved sequence of PCV2 published in GenBank, a pair of primers for amplification of PCV2 specified fragment was designed and a PCR assay for PCV2 detection was established. The results showed that the PCR was sensitive and specific, and useful to detect the clinic samples. It was helpful for clinical diagnosis and epidemiologic survey of PCV2.

Pairs of specific primer were designed according to the conserved coding region of sheep,Bos taurus Hairless and β-actin genome from GenBank.We synthesized the first strand cDNA by reverse transcription PCR after isolated the RNA from the skin tissues.The amplification products were ligated into the pGEM-T vector after recycling and purification,and trans formed into competent Top10 cells eventually.Plasmid DNA was purified and identified by restricted enzyme digestion,PCR amplification and sequencing.The positive plasmids enter into the real-time RT-PCR steps after gradient dilution from 103 to 107.The standard curve and regressive curve were generated automatically by the DNA continuous fluorescence detection system.The results showed that no specific amplicons was found according to the derived melting curve of the RT-PCR products, indicating the specification of the present primers.The correlation coefficients of the standard for the Hairlessand β-actin genes were 0.998 and 0.999, respectively, suggesting the strong linear relationship between the two groups.The results of repeatability experiments showed that the Ct values hold well for the recombinant plasmids with nine times PCR amplification under the same condition,and the coefficient of variation(CV)was less than 6% among groups(except of 107 copies/reaction),suggesting the great repeatability and stability of the standard curve.Therefore, the construction of the standard plasmid and standard curve established the foundation of the procedures focusing on the quantitation of Cashmere goats Hairless gene mRNA expression in skin tissues.

Uncoupling proteins (UCPs) belong to the family of mitochondrial transporter proteins that may uncouple the transport of protons across the inner mitochondrial membrane from electron transport, hence generating heat rather than energy. The study was conducted to determine the effect of diffent heat serum, diffent sheep individuals,sperm density,cumulus’effect and different culture media on the development of embryos from in vitro fertilization.The results showed that heat three days serum affect greatly the cleavage rate (CR) and the blastocyst rate (BR)(P<0.01);and the different sperm how ever they are freezed or fresh have significant affect on CR and BR. No significant differences in CR and BR were found both sperm densities of 2×106 to 6×106 sperms/mL.The cumulus of oocyte have no effect on IVF;there were no different effect on BR and CR of the culture media of SOF and CRLaa.

In order to improve pig parthenogenetic blastocyst adhesive rate, the experiment researchs both the culture medium and the feeder layer which can affect porcine embryo adhesive ability. Mouse, porcine and bovine fetal fibroblasts were adopted as feeder, to explore the development of the porcine parthenogenetic blastocysts cultured in DMEM or NCSU-23 or DMEM/ NCSU-23 medium. Bovine fetal fibroblasts feeder could betterly promote the porcine parthenogenetic blastocysts adherent growth, there were significant differences between the adhesion rate of bovine fetal fibroblasts feeder (33.67%) and that of mouse fetal fibroblasts feeder (19.08%)(P<0.05) , while was no significant difference between the adhesion rate of bovine fetal fibroblasts feeder and that of porcine fetal fibroblasts feeder (23.47%), the adhesion rate of mouse embryonic fibroblasts feeder group and that of the porcine fetal fibroblasts feeder groups (P>0.05). In bovine fetal fibroblasts feeder group, the blastocyst adhesion rate (22.53%) cultured in the porcine embryo culture medium NCSU-23 was significantly higher than in the DMEM culture medium (10.41%) and DMEM/NCSU-23 (12.05%) half mixed culture medium (P<0.05), and there was no significant difference between cultured in DMEM culture medium group and DMEM/NCSU-23 half mixed culture medium group (P>0.05). Bovine fetal fibroblasts feeder and porcine embryo culture medium NCSU-23 could better promote development of porcine parthenogenetic blastocyst.

Guizhou white pig is a special Guizhou pig breed native. In the present study, the polymorphism of exon 15 to 17 of BF gene about 733 bp was measured with the method of direct sequenceing in Guizhou white pig. The results showed that there was a transversion at nucleotide position 41 of BF gene in the 16th exon led to a conservative Glutamic-to-Aspartic substitution at amino acid position 14. AA, AC and CC e genotypes were detected in Guizhou white pig population, the frequencies of allele A and C were 0.88 and 0.12 respectively.

This study was designed to explore the effects of Letrozole which is an P450 aromatase inhibitor on ovarian development in mice. SPF degree of Kunming female mice were randomly divided into control and 2,4,6,8,10,30,50 g/kg of Letrozole dose groups, consecutive administration for 3 d. At the 9, 15 and 30 d after stopping administration, the ovarian development was detected. The results showed that: ①The ovary index was significantly (P<0.05) increased in 6 and 8 g/kg groups at the 9 and 30 d,and 4 to 10 g/kg groups at the 15 d compared with control group, however, it was significantly (P<0.05)decreased in 50 g/kg groups at the 9 and 30 d compared with control group and 2,4,6,8,10 g/kg groups; ②The number of primary follicle was significantly(P<0.05)increased in 4,6,8 g/kg group at the 9 and 15 d and 6 g/kg group at the 30 d compared with control group and other dose groups, however, it was significantly(P<0.05)decreased in 30 and 50 g/kg groups at the 9 d and 50 g/kg group at the 15 d and 10,30,50 g/kg group at the 30 d compared with control group and other dose groups; ③The number of secondary follicle was significantly (P<0.05) increased in 4,6,8 g/kg groups at the 9,
15 and 30 d, however, it was significantly (P<0.05) decreased in 30 and 50 g/kg groups at the 9 d compared with control group; ④The number of mature follicle was significantly(P<0.05)increased in 4,6,8 g/kg groups at the 9 d and 6 and 8 g/kg groups at the 15 d and 2,4,6,8 g/kg groups at the 30 d, however, it was significantly (P<0.05) decreased in 10 to 50 g/kg groups at the 9 d and 30 and 50 g/kg groups at the 15 and 30 d compared with control group. These results indicated that the concentration of Letrozole with 4,6,8 g/kg can inhibited the P450 aromatase activity to some extent, reducing endogenous estrogen synthesis and promote the development of ovaries and follicles in mice.

Satellite cell is an accessible adult stem-cell,which occupying a specialized niche beneath the basal laminae of myofiber.Although we have much more understanding about satellite cell according to the years’ researches, there is a lot of questions which are unknown. Recently, people focus on one question:how is satellite cell self-renewal regulated. CAPN1 gene as the major gene which controls beef tenderization was concerned by researchers. In this study,we reported the 4685 bp loci genotype frequency and allele frequency in Chinese Red Steppe,Yanbian Yellow cattle, Red Angus×Simmental, Limousin×Simmental, Germany Yellow cattle×Simmental and Charolais×Simmental cross. The results showed that genotype frequency and allele frequency were significant different in these population. As the predominant genotype in Yanbian Yellow cattle,the frequency of CT genotype was 0.53,the frequency of CC and TT genotype just were 0.27 and 0.20 respectively. The TT and CT genotype had very high frequency in Germany Yellow cattle×Simmental cross, reached 0.50 and 0.40 respectively, the CC genotype frequency only was 0.10, the T allele frequency reached 0.70, and the C allele frequency only was 0.30. The others cross cattle had similar genotype and allele frequency to Yanbian Yellow cattle. The most interesting result in this study was the genotype distribution in Chinese Red cattle,we only found one genotype TT which suggested C allele was not exist in Chinese Red cattle. The complement of genotype frequency test of 4685 bp loci in CAPN1 gene in triple cattle laid a foundation to research the relationship of 4685 bp loci to meat tenderness.

The genomes of six production groups chickens were screened using microsatellites to get the genetic distance of every combination, and analysed the relation among the heterosis of the major economic traits. The results showed that the absolute value of relation coefficient were range between 0.0651 and 0.9344. Brest width,eviscerated yield with giblet, leg muscle yield, etc, were main relation with genetic distance. Born weight and shank length were minus relation with genetic distance, and also obtained that three targets could be used to predict the hybrids offspring’s production level, so the microsatellite markers genetic distance was important for the predict of heterosis.

In order to increase the quality of transgene vectors and the transgenic efficiency, the construction and the development course of the lentiviral vector were reviewed. In order to increase the biosafety, lengthen the loaded fragment of target DNA, and extend the host cell of the lentiviral vector, all accessory genes of lentivirus were deleted, certain exogenous regulatory sequences were introduced, and the envelope glycoprotein was replaced also, which gives the lentiviral vector a broad range of target cells and a higher biosafety, transfer efficiency and expression efficiency. What’s more, the developed transfection method laid the foundation for integration and optimization of transgenic technology. The integration and optimize between lentiviral vector and other transgene technologies will help to create a simple, efficient and economical transgene technology.

Genetic variation of Luxi,BLACK,Bohai cattle in Shandong and one foreign breed,Japanese black breed,was assessed by analyzing a random sample of 185 animals using 10 microsatellite loci. A total of 58 alleles were identified, averaged about 5.8 ranging from 4 (BM1818) to 8 (ETH225 and TGLA53). All the microsatellites were highly polymorphic,and various genetic variability measures,including observed heterozygosity (0.2270 to 0.5459), expected heterozygosity (0.6082 to 0.7230) and polymorphism information content (0.5482 to 0.6791), and effective numbers of alleles was 2.5417 to 3.5849. Considerable genetic differentiation was observed and 34.49% of the total genetic variation originated from the differences among breeds. Then we constructed UPGMA tree by genetic distances (DA). Luxi cattle clustered first with Bohai breed, then BLACK cattle and Japanese black clustered.The sequences of PCR amplification fragment of 10 clones of 3 microsatellite loci in BLACK cattle were accepted by GenBank, the GenBank accession numbers were GQ368896,GQ368897,GQ368898,GQ368899,GQ368900,GQ368901,GQ368902,GQ368903,GQ368904,GQ368905,respectively. All of 10 microsatellite loci were effective markers for analysis of genetic relationship among 4 beef cattle breeds.

Multivariate factor analysis and principal component analysis were used to decompose the correlation matrix of body measurement trait of 30 Xinjiang Brown cattle of age about 18 months, then indexes of Xinjiang Brown cattle body measurement trait were studied by the principal components analysis, the 7 principal components which contributing rate accumulated up to 89.505% were chose. The results showed that there was big potential of mass selection for the rump length, hip width, thurl width, pin bone width and body weight, there were high significant correlation among body weight,body length, heart girth, hip width(P<0.01), and there was significant correlation with thurl width(P<0.05). The first principal component reflected the entire growth and development condition of Xinjiang Brown cattle, the second principal component reflected rump characteristics, the third principal component reflected back body characteristics.

In this study, PCR-RFLP method was established according to a novel SNP site (C218T) within the intron 4 of sheep insulin-like growth factor binding proteins-3 (IGFBP-3) gene. Using this method, 353 Chinese Merino sheeps (Xinjiang Junken type) were tested and illustrated three unique binding genotypes of CC (249 bp), CT (249/216/33 bp) and TT (216/33bp). Frequencies of the genotype CC, CT, TT were 0.52, 0.42, 0.07, respectively, and frequencies of the allele C, T were 0.72, 0.28, respectively. Different genotypes slightly affected several wool traits of Chinese Merino sheeps. Staple length (SL) in individuals of genotype CC was significantly shorter than that in individuals of genotype TT (P<0.05), follicle density in individuals of genotype CC was significantly higher than that in individuals of genotype CT and TT (P<0.05).

The polymorphism of growth hormone (GH) gene was detected by PCR-RFLP method, and association analysis with performance trait of Jining goat were performed. The results showed that the endonuclease Hae Ⅲ polymorphism was found in GH gene in Jining goat population, and it was in non-Hardy-weinberg equilibrium using the χ2 test. AA genotype had a significant effect on body heigh at 3 and 12 months old, birth weigh, weaning weight, carcass weight, meat hindquarters weight (P<0.05), and AA>AB>BB. In conclusion, the allele A was a benefical gene on the growth and debelopment in Jining goat population.

To study the genetic effect of forllicular-stimulating hormone beta subumit (FSHβ) on the number born alive (NBA) trait and its application in pig reproduction trait selection, the polymorphism of FSH β was detected by the method of PCR and association analysis between different genotypes and NBA trait were carried out in 802 Large white sows with 3418 parities from two commercial breeding pig companies which were Hubei Tianzhong and Yichang Zhengda. The frequency of A allele was 0.447 in the Hubei Tianzhong sows population and 0.312 in the Yichang Zhengda sows population. It was demonstrated that the effect of different genotypes of FSH β loci on the NBA was AB>BB>AA at the first parity, AA>AB>BB at the second parity and AB>AA>BB at the later parities and all parities, respectively, but no significance was found between different genotypes. The results showed that the effect of A allele was better than B allele on the NBA in large white. The sows of AA genotype have more NBA than the sows of other genotypes. Thus, increasing the number of sows with AB genotype on the FSH β loci might bring us more litter pigs in commercial pig production.

To study the traditional Chinese medicine effects on the biochemical parameters of serum and immunologic organ of broilers and research its influence on the some intermediary metabolism and immunologic function, 120 AA broilers(1-day-old) were randomly divided into 3 groups, groupⅠ(control), groupⅡ(each was fed clinic preventive dose i.e. 0.1 g/d of complex of traditional Chinese medicine)and group Ⅲ(each was fed 0.3 g/d).After 30th days, the immunologic organal index and biochemical parameters of serum(TP,GLB and ALB) were determined. As the result, TP of group Ⅲ were markedly more than group ⅠandⅡ, but among the groups, the GLB and ALB had no differences. During the experiment, the absolute weigh and index of groupⅡ and Ⅲ organs had some improvments. The results showed that the traditional Chinese medicine played a significant role in the immunologic function of broilers. 

To assess the pharmacological effects of compound of Fructus mume and Semen ziziphi spinosae extracts against infectious bursal disease (IBD) in the laboratory. Firstly, the chickens without the attenuated IBD vaccine were inoculated with the IBDV LX strain in order to establish the virus disease model. Secondly,Fructus mume and Semen ziziphi spinosae extracts were formulated according to the previous study. Chickens were administered orally with 400,200 and 100 mg/kg herb extracts.The chickens were administered with astragalus polysaccharide as the control group.All birds were monitored daily and evaluated the efficacy.The results showed that: ①After experimental infection with IBD,average weight gain of the infected chickens was 81.85% in 200 mg/kg group and 48.24% in astragalus polysaccharide group. As for survival rate, the infected chickens were 87.5% and 100% in above group,respectively.Average weight gain was significant difference in 200 mg/kg group as compared to that of astragalus polysaccharide group. No significance was found between two groups. The chicken bursal index was 0.187 and 0.123, respectively in 200 mg/kg group and in astragalus polysaccharide group while the spleen index were 0.361 and 0.286, respectively in above two groups. There was significant difference (P<0.05) between two groups.②In second experiment, the relative weight gain in above mentioned groups were 29.55 and 46.1 while survival rate was 100% and 80%, respectively, which was no significant difference between 200 mg/kg group and astragalus polysaccharide group. The chicken bursal index were 0.22 and 0.25 while spleen index were 0.34 and 0.36, respectively. Statistical analysis showed that no significant difference was found in immune organ index between two mentioned groups (P>0.05). Chickens were orally administered with 200 mg/kg Fructus mume and Semen ziziphi spinosae extract for 5 days, resulting in maximum 80% survival rate against the artificially challenge with IBD. The improvement of immune organ index contributed to the promising pharmacological effects.

To determine the contents of Aspirin eugenol ester microemulsion(AEE-ME) and the related substances in AEE-ME, RP-HPLC method was used. The high performance liquid chromatographic conditions were as followed: ODS column (4.6 mm×100 mm,5 μm) was used as stationary phase and methanol-water (70∶30) was used as mobile phase; the detection wave length was 279 nm; lowing rate was 1.0 mL/min; temperature was 30 ℃. The calibration curves was a good linearity in the range of 1.25 to 40 μg/mL for AEE at 279 nm(r=0.9923),the average recovery rate was 99.93%, and RSD was0.84%. The results showed that the method was suitable for detection of the content and the related substances of Aspirin eugenol ester and could be used in quality control.

An experiment was carried out to evaluate the effect of intramuscular injection of different concentrations of sulfated glucomannan on protection and immune function of broilers artificially infected with infectious bursal disease virus. 270 1-day-old AA broilers were randomly divided into five treatments ,Ⅰ,Ⅱ,Ⅲ,Ⅳ group with 55 birds,Ⅴ group with 50 birds. At 18-day-old three groups were injected with 1, 2 and 4 mg/mL concentrations of sulfated glucomannan and each 1 mL for 3 days, the other two groups as a virus control and conventional control. At 21-day-old except normal control the other four groups were infected with the same dose of bursal disease virus, a trial period for 3 weeks. The results showed that injection of high concentrations of sulfated glucomannan can reduce morbidity and mortality of chickens significantly; in 1 week post-infection high concentrations group compared with the control group was able to significantly promote lymphocyte proliferation (P<0.05); in the two weeks post-infection, antibodies of high concentrations group was significantly higher than the virus control group (P<0.05). The rest period of each group compared with the viral group was able to raise antibody levels but the difference was not significant (P>0.05).

The distribution and depletion of Clenbuterol in swine urine and blood after treatment with dietary Clenbuterol were studied. Ten pigs(60 kg±5.7 kg) were used to determine the residues and depletion of Clenbuterol in swine. Pigs were fed 4 mg/kg Clenbuterol HCl for fifteen consecutive days. Samples of urine and blood were collected to determine Clenbuterol concentrations by GC/MS. The results showed that the mean concentrations of Clenbuterol were 393 to 1131 μg/L in urine, and 6.8 to 22.1 μg/L in blood during feeding period. The concentrations of Clenbuterol were rapidly decreased. With a 19-day withdrawal period, the mean concentrations of Clenbuterol were 2.19 and 0.49 μg/L in urine and blood, respectively. Moreover, there was a significant positive correlation between the concentration of Clenbuterol in urine and the concentration of Clenbuterol in blood. The correlation coefficient was 0.9385 and 0.9402 during feeding period and withdrawal period, respectively.

In order to investigate the crucial reason which led to acute death of newborn hamsters on a hamster farm in Changchun and response paroxysmal disease, the death hamsters were treated by laboratory examination and one bacterial strain was isolated from hamsters which died acutely in this study. The isolated bacteria was identified as one Escherichia coli tract inflated extremely. Drug sensitivity test showed that isolated bacteria was sensitive to many kinds of antibiotics,but had strong drug resistance to Tetracycline antibiotics, Sulfame-thoxazole and Trimethoprium (SXT), and Gentamycin could be used to treat infection caused by this E.coli strain. All results obtained from this study provided well foundation for the further research on the pathogenesis,prevention and control of acute colibacillosis’ producted by bacteria.

To investigate the pharmacokinetics of Ivermectin emulsion compound in vivo, and to provide the medical information for the veterinary clinical diagnosis, 7 goats as experimental models were selected, and each goat was given orally Ivermectin emulsion compound by dose of 0.1 mL/kg (Ivermectin 0.2 mg/kg, Albendazole 10 mg/kg). Blood samples were respectively taken 5 mL from Jugular vein at 0.5, 1, 2, 3, 4, 6, 8, 12, 16, 24, 36, 48 and 60 h after administration, then the serum were separated and stored at -20 ℃. High performance liquid chromatography (HPLC) method was used to detect serum drug level. The results showed that: ①The metabolism of Ivermectin in goat: the drug concentration was 0.112151 μg/mL after 0.5 h; the first peak time was 0.302702 μg/mL after 4 h, and the second peak time was both 0.258284 μg/mL after 16 h and 0.011118 μg/mL after 60 h. ②Albendazole metabolism in goats:the drug concentration was 0.049285 μg/mL after 0.5 h; the first peak time was 4.95283 μg/mL after 8 h, and the second peak time were both of 5.694551 μg/mL after 16 h and 0.06434 μg/mL after 60 h.The metabolized time of Ivermectin and Albendazole of Ivermectin emulsion compound in goat was very short, and the drug concentration in serum was not too low until 60 h.

To explore the safety of Potassium dichromate used as the solution for coccidiosis vaccine preservation, the acute toxicity test and toxicity test with low dosage in chicken were conducted by oral administration. The clinical symptoms, gross lesion and histopathologic changes were observed. The results showed that LD50 of potassium dichromate was 0.501 g/kg body weight, which was more than 160 times that the recommended amounts for clinical use (about 3.125 mg/kg body weight). Heart, liver, lung, proventriculus and kidney of chicken were damaged in varying degree with the dosage of more than 0.464 g/kg body weight. 5 or 20 times of the dosage in routine immunization had no harm to chicken.

A total of 12 nasal swabs and 4 lung specimens were collected from a goat flock with infectious pleuropneumonic disease in Lezhi, Sichuan province. Pathogen isolation and PCR detection were performed for all samples. The results showed that Mycoplasma ovipneumoniae was isolated and identified from 9 nasal swabs and all 4 lung specimens, while any of Mycoplasma mycoides cluster members, Pasteurella multocida or Mannheimia haemolytica was not found. This indicated that Mycoplasma ovipneumoniae is responsible for the pleuropneumonic disease in this flock. It is also suggested that Mycoplasma ovipneumoniae is one of the important pathogens causing Mycoplasma pneumoniae in goats.

he purpose of this paper was to study biochemical characteristics, drug sensitivity, serotype and pathogenic infection of Riemerella anatipestifer(RA) from geese. One goose isolate from an infected farm of the suburb of Yangzhou were isolated, cultured, stained, tested by microscopy, biochemical identified, and slide agglutination test. The results showed that the isolate was Ⅱ serotype RA. While no Newcastle disease virus, avian influenza virus, and gosling plague virus were isolated by hemagglutination test (HA) and agar-gel precipitin test (AGP).At the same time, the isolate was used to infect 15-days-old geese,the isolate attacked the geese through intravenous injection, subcutaneous injection and intranasal three infected ways. The results showed that RA had strong pathogenicity to Yangzhou goose, which prompted us, the Rimer ella anatipestifer pathogenicity and dissemination should also be taken fully into account in the immunization programs for goose.

The positive rate was 7.65%. The result showed that the infection rate of toxoplasmosis on yak was relatively high in Qinghai province.

To grasp the number and species of bacteria in the surrounding environment of the scaled pig farm and provide the basis for prevention and control of the bacterial disease of pig, the air, drinking and sewage water, faecal and soil samples were collected for bacteriological isolation and identification which from two-scale pig farms in Guizhou province. The results showed that the numbers of bacteria were respectively 1.17×104 to 94.40×104, 0.15×102 to 1.83×102, 5.00×105 to 32.00×105, 0.46 ×106 to 26.40×106 and 0.30×105 to 52.30×105 CFU/g in the above-mentioned samples, and of these bacteria, there were mainly Staphylococcus, Streptococcus, Escherichia, Salmonella, Yersinia, Proteus, Pasteurella, Listeria, Bacillus, Clostridium and so on. The results suggested that the numbers of bacteria in the other samples except drinking water and soil, were in line with requirement of national regulation for livestock and pourty farms, while there were some conditioned pathogen or pathogen,and these investigations provided an important reference data for prevention and control of pig bacterial diseases.

Epidemiological investigations were carried out in several layer-densely-cultured areas and the result showed that the avian leukosis epidemic situation was caused by subgroup J avian leukosis virus infection and mostly occurred in commercial layers with seriously economic loses. Furthermore, 665 serum samples, 688 cloacal swabs and 180 hatching eggs were detected by using ELISA method,and the results demonstrated that the layers especially the commercial layers were seriously contaminated by subgroup J avian leukosis virus in these layer-densely-cultured areas in Hubei province.

Canine hip dysplasia is a complex progressive disease which is caused by genetic and other factors, and it usually occurs in puppies of large-breed dogs whose age is between 4 and 10 months. The typical clinical features are pain and lameness, and the main pathological changes are flattening of the acetabulum, deformation of the femoral head, different degree laxity of the hip joint, incomplete dislocation or total dislocation, and even developed into a serious degenerative arthritis. There are many studies which concern the genetic characteristics, method and standards of diagnosis, various treatments since the disease was first discovered. This paper reviews pathogenic factors, control measures, method commonly used in diagnosis and treatment of canine hip dysplasia, in order to provide references for whole understanding of the disease and further deep research.

The seroprevalence of toxoplasmosis in pigs was investigated of Xishuangbanna state in Yunnan province.711 serum samples were collected from Jinghong, Menghai and Mengla, which included 680 rural “free-range” pigs samples and 31 slaughterhouses samples, and the Toxoplasma gondii antibodies were detected by indirect haemagglutination (IHA), the test was considered positive when a layer of agglutinated erythrocytes were formed in wells at dilutions of 1∶64 or higher. The results showed that 174 serums were positive, and the seroprecalence was 24.47%. The data of the present survey indicated that pig infection with T. gondii was serious in Xishuangbanna state, it was necessary to enhance integrated strategies and measures to prevent and control T. gondii infection in pigs.

This study compared effects of tow group sizes(20 pigs per peng and 10 pigs per peng, body weight from 17 to 40 kg) and tow feeding methods(free feeding vs rationed feeding, both 10 pigs peng, body weight from 34 to 60 kg) in a 40 days period, each involving 60 Seghers hybrid pigs. For the pigs raised in large group size(20 pigs per peng) and that raised in small group size(10 pigs per peng), the formers’ growth rate declined 4.95% (P>0.05), their daily feed intake decreases 9.72% (P>0.05), their feed conversion efficiency increases 5.45% (P> 0.05), and they were given gross profit reduction of 0.14 yuan per pig. For the pigs raised by free feeding and that raised by rationed feeding, the formers’growth rate rised 13.3% (P<0.05), their daily feed intake increased 16.87% (P<0.05), their feed conversion efficiency declined 3.14% (P> 0.05), they were given gross profit increase of 11.31 yuan per pig.

The objective of this research is to find the influence on specific microRNAs in bovine mammary gland epithelial cells by Semen vaccariae active isomer and prolactin. Quantitative real-time PCR was used to detect the expression changes of microRNAs (21, 125b, 143 and 195) in the mammary gland epithelial cells with dibutyl phthalate (DBP) and prolactin (PRL) at different time. The results showed that both DBP and PRL could repress the expression of miRNA-125b, miRNA-143 and miRNA-195 in mammary gland epithelial cell; DBP could repress the expression of miRNA-21,while the influence of PRL on miRNA-21 was not certain. It is the first time to demonstrate that the expression of the microRNAs can be changed by Semen vaccariae active isomer (DBP)and prolactin.

This experiment is to establish methods of detecting pefloxacin in HBS-EP buffer by SPR biosensor, which can provide a basis for sample detection. The pefloxacin-ovalbumin conjugate was covalently immobilized on CM5 chip and the antibody was mixed with pefloxacin of different concentrations to construct a standard curve. To obtain lower limit detection, the diluted antibody of 1000 times was injected at a flow rate of 7 μL/min for 4 min. It was shown from the experimental results that a rapid detection of pefloxacin in buffer using SPR biosensor had been established and the limit of detection was 0.8 ng/mL, the detection time was 4 min.
