Molecular Cloning and Characteristics of cDNA Encoding Porcine nectin-2 for PRV Receptor

Abstract: In order to understand the structure and function of porcine nectin-2 gene, we cloned and sequenced the cDNA encoding porcine nectin-2 from the brain of pig. The 1440 bp cDNA of porcine nectin-2 encoded a polypeptide of 479 amino acids consisting of a 32-residue putative signal peptide, a 330-residue ectodomain, a 23-residue transmembrane domain, and a 94-residue cytoplasmic domain. The ectodomain contained 2 potential N-linked glycosylation sites and 6 cysteine residues. The nucleotide sequence similarity of nectin-2 between pig and dog, horse, house mouse, human, rhesus monkey, cattle, Chimpanzee were 85.4%, 85.7%, 78.6%, 82.1%, 82.1%, 81.9%, 82.1%, and the amino acid sequence similarity were 84.5%, 83.0%, 74.7%, 75.7%, 76.4%, 78.4%, 75.5% respectively. This study will lay a foundation for understanding the interactions of PRV with host.
